Project description:DHR96 plays a role in regulating xenobiotic responses in Drosophila. Using a gain-of-function approach we test whether DHR96 is sufficient to affect detoxification genes in the absence of a xenobiotic insult. Experiment Overall Design: A transgenic line that expresses a full-length DHR96 cDNA under heat-shock control was generated by standard P-element-mediated transformation, using w1118 as a host strain. Control flies are therefore w1118 without any transgene. Larvae were staged at the molt from L2 to L3, and heat-treated after 3 hours for one hour at 37.5 degrees (in vials). Finally, RNA was extracted 4 hours later.

Effects of caffeine ingestion in Drosophila melanogaster male thorax and abdomen.

Project description:To screen for xenobiotic-metabolizing-enzyme genes (XME) induced by caffeine exposure, a microarray experiment (performed with a Drosophila pangenomic array (Agilent 4 X 44K) was carried out between control and individuals exposed to caffeine (18mM ). Among 23401 genes consistantly found in the three biological replicates, 13108 showed a significant variation. 749 genes were over-expressed whereas 1518 were under-expressed. Interestingly, several genes belonging to all major classes of detoxification enzymes including XME were among the most highly induced such as CYP12d1, CYP6a8 and CYP6d5. Caffeine ingestion induced gene expression in Drosophila melanogaster male thorax and abdomen was mesured at 12h after exposure to doses of 0, 18mM of caffeine. Three independant experiments were performed at each dose using different drosophila for each experiment.

Ectopic assembly of heterochromatin in Drosophila melanogaster triggered by transposable elements.

Project description:A persistent question in biology is how cis-acting sequence elements influence trans-acting factors and the local chromatin environment to modulate gene expression. We reported previously that the DNA transposon 1360 can enhance silencing of a reporter in a heterochromatic domain of Drosophila melanogaster. We have now generated a collection of variegating phiC31 landing-pad insertion lines containing 1360 and a heat-shock protein 70 (hsp70)-driven white reporter to explore the mechanism of 1360-sensitive silencing. Many 1360-sensitive sites were identified, some in apparently euchromatic domains, although all are close to heterochromatic masses. One such site (line 1198; insertion near the base of chromosome arm 2L) has been investigated in detail. ChIP analysis shows 1360-dependent Heterochromatin Protein 1a (HP1a) accumulation at this otherwise euchromatic site. The phiC31 landing pad system allows different 1360 constructs to be swapped with the full-length element at the same genomic site to identify the sequences that mediate 1360-sensitive silencing. Short deletions over sites with homology to PIWI-interacting RNAs (piRNAs) are sufficient to compromise 1360-sensitive silencing. Similar results were obtained on replacing 1360 with Invader4 (a retrotransposon), suggesting that this phenomenon likely applies to a broader set of transposable elements. Our results suggest a model in which piRNA sequence elements behave as cis-acting targets for heterochromatin assembly, likely in the early embryo, where piRNA pathway components are abundant, with the heterochromatic state subsequently propagated by chromatin modifiers present in somatic tissue.

The Mechanisms Underlying α-Amanitin Resistance in Drosophila melanogaster: A Microarray Analysis

Project description:The rapid evolution of toxin resistance in animals has important consequences for the ecology of species and our economy. Pesticide resistance in insects has been a subject of intensive study, however, very little is known about how Drosophila species became resistant to natural toxins with ecological relevance, such as α-amanitin that is produced in deadly poisonous mushrooms. Here we performed a microarray study to elucidate the genes, chromosomal loci, molecular functions, biological processes, and cellular components that contribute to the α-amanitin resistance phenotype in Drosophila melanogaster. We suggest that toxin entry blockage through the cuticle, phase I and II detoxification, sequestration in lipid particles, and proteolytic cleavage of α-amanitin contribute in concert to this quantitative trait. We speculate that the resistance to mushroom toxins in Drosophila melanogaster and perhaps in mycophagous Drosophila species has evolved as a cross-resistance to pesticides or other xenobiotic substances.

Understanding oxidative stress responses in pancreatic islet cells by single cell RNA-sequencing analysis

Project description:We found these ROS generation is regulated by lncRNA MALAT1 and genetic ablation of MALAT1 drastically reduced ROS level and oxidative stress in mouse islet cells with the benefits of improved insulin responses in MALAT1-/- mouse. The pancreatic islet consists of five cell types (α, β and γ/PP, δ and ε cells) and very little is known about the xenobiotic detoxification pathways in these cells and their sensitivity to toxicants. We utilized single-cell RNA sequencing to analyze the role of MALAT1 in regulating oxidative stress response and insulin secretion function in distinct pancreatic cell population. We also treat the isolated pancreatic islets with 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) (10 nM, 12h) to investigate the xenobiotic detoxification pathways regulation in both MALAT1 KO and WT pancreatic islets. Our result showed that a subset of genes in T2DM related pathways were significantly regulated in MALAT1 -/- β cells, with significantly unregulated INS1, INS2, and PDX1. Nrf2/detoxification pathway was also significantly activated in MALAT1 -/- β cells. In addition, MALAT1 expression level was elevated in the T2DM patients pancreatic islets cells. This study provides insights for mechanisms of regulation of oxidative stress by MALAT1-Nrf2 interaction which has the potential as a therapeutic target for the treatment of T2DM.
