Transcriptomics to discover splice variants involved in the browning of adipose tissue

Project description:Adipose tissue is the major depot for energy storage. Recent studies have shown that at least three types of adipocytes can be distinguished depending on their anatomical locations : 1) The classic brown adipocytes, i.e., brown adipose tissue (BAT); 2) The 'brite' (brown-in-white) adipocytes, i.e. inguinal white adipose tissue (iWAT); 3) The 'true' white adipocytes, i.e., epididymal white adipose tissue (eWAT). Two strains of mice (SV129 and C57BL/6J) were used in this study. SV strain is resistant to obesity and latter is prone to obesity. Pre-adipocyte cells were isolated from subcutaneous tissue (iWAT) to create four groups of cell cultures per strain of mouse.

Gene Expression in the BAT and WAT of ob/ob and ob/ob/Cidea-/-/Cidec-/- mice

Project description:ob/ob mice is an obese mice. CIDE family proteins including Cidea, Cideb and Cidec play important role in lipid metabolism. Cidea is mainly expressed in the brown adipose tissue (BAT). Cidec is mainly expressed in the BAT and white adipose tissue (WAT). We generated ob/ob/Cidea-/-/Cidec-/- mice to investigate the phenotype of fat tissue. ob/ob/Cidea-/-/Cidec-/- mice are lean when compared with ob/ob mice. The tissue weight and TAG content of BAT and WAT was extreamly decreased in ob/ob/Cidea-/-/Cidec-/- mice compared with that in ob/ob mice. We next extract the total RNA from the BAT and WAT of ob/ob and ob/ob/Cidea-/-/Cidec-/- mice, to perform microarray analysis using Mouse Gene 1.0 ST array system, Affymetrix. We then analysised the up-regulated and down regulated pathways.

Hypoxia-induced metabolic dysfunction in WAT

Project description:Background: Excessive white adipose tissue (WAT) expansion as in obesity is generally associated with chronic inflammation of WAT, which contributes to obesity associated complications. Low oxygen availability in WAT is hypothesized to be the initiator of this inflammatory response. Hypothesis: We examined the hypothesis that local tissue hypoxia is responsible for the initiation of inflammation in WAT. Research design and methods: Diet-induced obese male C57BL/6JOlaHsd mice, housed at thermoneutrality, were exposed to mild environmental oxygen restriction (OxR, to 13% oxygen) for five days and compared with mice kept at normoxia, after which WAT and serum were collected. Body composition, systemic metabolic parameters, WAT macrophage infiltration (as marker for tissue inflammation) and whole genome microarray analysis, and circulating adipokines were measured. Results: Five days OxR decreased body weight and fat mass, and increased blood levels of haemoglobin and haematocrit, as well as lactate to glucose ratio, which indicated systemic hypoxia. No difference in adipose tissue inflammation was found, which was supported by down regulation of inflammation-associated transcript levels of S100a8, Saa1, and Saa3. Serum metabolomics revealed an increase of branched chain amino acid Valine and propionylcarnitine. Adipokines CCDC3, CCK, and Adiponectin are reduced by OxR on transcript (Cck) or serum protein level (Adiponectin), or both (CCDC3). Conclusions: Mild oxygen restriction does not increase white adipose tissue inflammation in obese mice. However, a systemic adaptation together with a metabolic response in WAT was observed.

Transcription profiling of mouse white adipose tissue from PPARb/d knockout

Project description:Analysis of white adipose tissue of PPARb/d knockout mice. Data may point towards putative target genes of PPARb/d and thus the function of PPARb/d in white adipose tissue. Datasets were used to identify glycogen synthase 2 as novel PPAR target. Experiment Overall Design: Three month old male PPARb/d knock-out mice were on a mixed background (Sv129/C57BL/6). Wild-type littermates served as control animals. Animals received standard chow. Epididymal white adipose tissue (WAT) was removed and total RNA was isolated and pooled afterwards (five animals per group). Pooled RNA was hybridized to Affymetrix mouse genome 430 2.0 arrays arrays. Five microgram total RNA was labeled according to the ENZO-protocol, fragmented and hybridized according to Affymetrix protocols.

Project description:We found that the circadian protein PER2 interacts with the nuclear receptor PPARgamma to repress its activity. PPARgamma is a master regulator of adipogenesis and lipid metabolism and is very abundant in adipose tissue. We used microarrays to detail the global program of gene expression in adipose tissue lacking the per2 gene. This analysis identified several PPARgamma target genes up-regulated in adipose tissue from per2-/- mice. Per2-/- and per2+/+ male mice (Bae et al., 2001) were housed under 12 hr light/12 hr dark (LD) cycles. Mice 20 weeks old were sacrificed at the same time and adipose tissue (WAT and BAT) was collected for RNA extraction. 3 biological replicates per mouse/tissue.
