The four RNA sources, WT scWAT, Lr11 -/- scWAT, WT BAT and Lr11 -/- BAT, were prepared from subcutaneous WAT and BAT from wild-type mice and Lr11 -/- mice, respectively (n=3 each).

Project description:We run microarrays from three per group Sv129 female mice, ten weeks old, which were maintained at 28M-BM-0C (warm conditions) or 6M-BM-0 C (cold stimulated) for ten days, while standard animal house temperature is 22 M-BM-0C. After ten days, three types of tissue were collected: Brown Adipose Tissue (BAT), Mesenteric (visceral) White Adipose Tissue (MES) and Posterior Subcutaneous White Adipose Tissue (WAT) Different adipose tissue depots were taken for RNA extraction and hybridization on Affymetrix microarrays. Gene Expression in the BAT and WAT of ob/ob and ob/ob/Cidea-/-/Cidec-/- mice

Project description:ob/ob mice is an obese mice. CIDE family proteins including Cidea, Cideb and Cidec play important role in lipid metabolism. Cidea is mainly expressed in the brown adipose tissue (BAT). Cidec is mainly expressed in the BAT and white adipose tissue (WAT). We generated ob/ob/Cidea-/-/Cidec-/- mice to investigate the phenotype of fat tissue. ob/ob/Cidea-/-/Cidec-/- mice are lean when compared with ob/ob mice. The tissue weight and TAG content of BAT and WAT was extreamly decreased in ob/ob/Cidea-/-/Cidec-/- mice compared with that in ob/ob mice. We next extract the total RNA from the BAT and WAT of ob/ob and ob/ob/Cidea-/-/Cidec-/- mice, to perform microarray analysis using Mouse Gene 1.0 ST array system, Affymetrix. We then analysised the up-regulated and down regulated pathways.

Proteome characterization of mitochondria, ER, and mitochondrial-associated membrane from BAT and liver

Project description:Mitochondria are the provider of cellular energy from aerobic respiration in all tissues. However, only brown adipose tissue (BAT) mitochondria are able to convert chemical energy in the form triglycerides into heat (thermogenesis). To ask if cellular respiration or thermogenic function require a specific lipidomic environment, we investigated the lipid species composition of brown adipose tissue mitochondria, and compared it those purified from liver, skeletal muscle and white adipose tissue. We emplyed mass spectrometry-based proteomics to evalaute the purity of the mitochondia fractions isolated from murine BAT and liver tissue by differential centrifugation, by comparison of its full proteome with endoplasmatic reticulum, mitochondrial-associated membrane, and mitochondria fractions isolated by density gradient centrifugation.
