Project description:These gene expression profiles were measured to create a broad and balanced control for any project that examines gene expression changes in men exposed to a defined stimulus (see series 5105). Experiment Overall Design: 9 healthy men (aged 21 to 44) gave written agreement to participate to this study. At the time of blood withdrawal all participants were non-smokers, with normal BMI and did not obtain any medicamentation. We collected 12ml of whole blood and isolated T-lymphocytes out of 4ml and monocytes out of 8ml whole blood.

Dosage compensation mass spectrometry in platypus, chicken and mouse

Project description:Gene dosage imbalance of heteromorphic sex chromosomes (XY or ZW) exists between the sexes, and with the autosomes. Mammalian X chromosome inactivation was long thought to imply a critical need for dosage compensation in vertebrates. However, mRNA abundance measurements that demonstrated sex chromosome transcripts are neither balanced between the sexes or with autosomes in monotreme mammals or birds brought sex chromosome dosage compensation into question. This study examines transcriptomic and proteomic levels of dosage compensation in platypus and chicken compared to mouse, a model eutherian species. We analyzed mRNA and protein levels in heart and liver tissues of chicken, mouse and platypus.

Transcription profiling of blood from participants with subjective memory decline

Project description:We have tested effect of bilberry/grape-juice on whole blood cell gene-expression in a nine-week double blind, placebo-controlled, dietary intervention study of aged men (n=62 - 67y) with subjective memory decline randomized to bilberry/grape- or control group (placebo). Blood-samples were collected at pre- and post intervention. Ten individuals of each group, with high baseline plasma-isoprostanes (> 86 pg/ml) were selected for blood cell gene expression profiling (Affymetrix Human-Genome U133 Plus 2.0).

Profiling Rest Intervals between Sets and Associated Factors in Resistance Training Participants.

Project description:The aims of this study were: (1) to report on and analyse rest interval (RI) control between sets adopted by resistance training (RT) participants; (2) to evaluate how RT participants control RIs; and (3) to describe the factors associated with controlling RIs, such as, gender, RT experience, and professional guidance. Four hundred and fifteen volunteers (198 women and 217 men) answered a questionnaire about RI control. Among the participants, 89.9% (n = 373) reported receiving guidance during physical training, of which 74.5% (n = 278) received instruction from a sports and exercise professional. The proportion of subjects that reported controlling RIs was 71.6% (n = 297). Most subjects that reported controlling RIs (95.0%, n = 282) reported adopting an RI of 60 s or less. There is no association of RI control with gender and professional guidance. The RI adopted by most of the participants might be considered short (<60 s), which is not in line with most RT guidelines. The results of the current study could be used to improve attitudes toward RT.

Peripheral blood gene-expression in heavy vs. moderate drinkers exposed to three types of personalized imagery

Project description:Alterations in stress-related gene-expression may play a role in stress-related drinking and the risk for alcohol dependence. Microarrays were used to measure changes in gene-expression in peripheral blood in non-smoking, social drinking subjects exposed to three types of personalized imagery: neutral, stressful (but not alcohol- related), and alcohol-related cues. Gene-expression was measured at baseline, immediately after, and 1 hour after stimulus presentation. Subjects were allowed to drink up to 750cc of beer in a “taste-test” following stimulus presentation in each imagery condition, and the amount of beer consumed was recorded. Gene-expression levels were compared in 2 groups of non-smoking subjects (n=11/group): heavy drinkers (HD, defined as regular alcohol use over the past year of at least 8 standard drinks/week for women and at least 15 standard drinks/week for men), and moderate drinkers (MD, defined as up to 7 standard drinks/week for women and 14 standard drinks/week for men).
