To determine to effects of DNA methylation on gene expression in M. tuberculosis, we genetically deleted a predicted DNA methyltransferase encoded by Rv3263 and subjected wildtype, mutant, and complemented strains to global expression analysis. RNA was extracted from triplicate cultures of wildtype, methyltransferase deletion, and complemented strains.

Mycobacterium tuberculosis m4C DNA methyltransferase Rv3204 promotes mycobacteria survival under oxidative stress

Project description:ROS can damage cellular components and is a key player in many cellular signaling. Tuberculosis remains a major global public health concern largely due to the ability of its causative agent-Mycobacterium tuberculosis (Mtb) to subvert ROS toxicity. DNA methylation deficiency was previously shown to attenuate the Mtb virulence. But the role of DNA methyltransferase in Mycobacteria survival and the link with ROS are unknown. Mycobacterial protein Rv3204 is a DNA methyltransferase specific for N4-methylcytosine instead of N6-methyladenine (m6A) or 5-methylocytosine (m5C). The survival rate of m4C methyltransferase deletion mutants was lowered, accompanied by higher accumulation of ROS and failure of upregulating the transcription of gene engaged in ROS clearance upon rifampin(Rif) exposure.

Project description:ING1b and GADD45a are nuclear proteins involved in the regulation of cell growth, apoptosis and DNA repair. We previously found that ING1b is required to target GADD45a-mediated active DNA-demethylation via TET1 to specific loci. In order to study the impact of ING1-GADD45a on gene expression, we compared the expression profile of wildtype mouse embryonic fibroblasts (MEFs) with Ing1- and Gadd45a- single- or double-knockout (DKO) MEFs.

Project description:We used DNA microarrays to define the M. tuberculosis FurB regulon. For this porpouse we constructed a M. tuberculosis strain where furB was disrupted and its transcriptional profile was compared on DNA microarray with the transcriptional profile of its parental strain (H37Rv). A total of 32 genes included in 16 putative transcription units were shown to be up-regulated in the mutant relative to H37Rv. No repressed genes were identified. Seven representative genes were chosen from the microarray experiments and their expression was measured by quantitative RT-PCR using sigA as invariant internal control. In support to the gene expression profiling data, the mRNA levels of all selected genes was clearly higher in the furB mutant strain relative to H37Rv Keywords: comparative genomic hybridization
