Expression profiling of A.thaliana with the posttranscriptionally silenced Myosin XI-K

ABSTRACT: This study was aimed to determine roles of individual myosins in organelle trafficking in plant cell. Expression of Myosin XI-K was suppressed in transgenic A.thaliana by using RNAi approach. Project description:This study was aimed to determine roles of individual myosins in organelle trafficking in plant cell. Expression of Myosin XI-K was suppressed in transgenic A.thaliana by using RNAi approach. To ensure that the RNAi-induced gene silencing was specific and did not affect expression of other myosins, a microarray analysis was performed. Experiment Overall Design: Total RNA was purified from A.thaliana plants transformed with an inverted repeat construct harbouring a sequence derived from Myosin XI-K ORF. 3 independent groups of plants were used for comparison of the expression profiles of 17 myosin RNAs to a control group comprised of plants constitutively expressing GUS ORF as a transgene.

The Class VIII myosin ATM1 is integral for root apical meristem function

Project description:Myosins are evolutionarily conserved motor proteins that interact with actin filaments to regulate organelle transport, cytoplasmic streaming and cell growth. Plant specific Class XI myosin proteins direct cell division and root organogenesis. However, the roles of Class VIII myosin proteins in plant growth and development are less understood. Here, we investigated the function of an auxin-regulated Class VIII myosin, Arabidopsis thaliana Myosin 1 (ATM1), using genetics and live cell microscopy. ATM1 is a plasmodesmata-localized protein that is enriched in actively dividing cells in the root apical meristem (RAM). Loss of ATM1 function results in impaired primary root growth due to decreased RAM size and reduced cell proliferation in a sugar dependent manner. Examination of stem cell marker line expression in atm1-1 indicated impaired division of the lateral root cap and columella cells and a diminished auxin response. Transcriptome analysis of atm1-1 linked these growth defects to dysregulation of cell cycle and auxin pathway genes. Complementation of ATM1 loss-of- function mutant restored root growth and cell cycle progression in the root meristem. Collectively, these results provide novel evidence that ATM1 functions to influence cell proliferation and differentiation in primary roots in response to auxin and sugar cues.

Xi-reactivation in female human induced pluripotent stem cells (hiPSCs)

Project description:Female human induced pluripotent stem cell (hiPSC) lines exhibit considerable variability in X-inactivation status. Some lines maintain one transcriptionally active X chromosome (Xa) and one inactive X (Xi) from donor cells. However, hiPSC lines that have two Xas are infrequently produced. We show here Xinactivation status in female hiPSC lines depends on derivation conditions. hiPSC lines generated using the Kyoto method, which employs leukemia inhibitory factor (LIF)-expressing SNL feeders, frequently had two Xas. Lines derived on other feeders maintained an Xi. In addition, there appears to be a window in which SNL feeders promote Xi-reactivation. Upon differentiation, Xa/Xa hiPSCs silenced one X. The efficient production of Xa/Xa hiPSC lines provides unprecedented opportunities to understand human X-reactivation and inactivation.
