Project description:The temporal program of gene expression during a model physiological response of human cells, the response of fibroblasts to serum, was explored with a complementary DNA microarray representing about 8600 different human genes. Genes could be clustered in groups on the basis of their temporal patterns of expression in this program. Many features of the transcriptional program appeared to be related to the physiology of wound repair suggesting that fibroblasts play a larger and richer role in this complex multicellular response than had previously been appreciated. Project description:Foreskin fibroblasts CRL 2091 (ATCC) were serum starved for 48 hours, and harvested at the indicated time points after switching to media with 10% FBS essentially as described (Iyer et al., 1999). RNA from all of the sampled time points were pooled as reference RNA to compare with RNA from individual time points as described (Iyer et al., 1999). Expression data for serum stimulation of human primary fibroblasts

Project description:We used microarrays to profile gene expression changes following growth factor stimulation of primary human fibroblasts. We serum starved (0.1% serum) fibroblasts for 48 hrs and restimulated with 10% serum for 0, 2, 4, 6 and 8 hrs. Total RNA was extracted from 2 independent biological replicas for each time point and hybridized to expression arrays.
