Project description:Infection-related development of phytopathogenic fungi is initiated by sensing and responding to plant surface cues. This response results in the formation of specialized infection structures, so-called appressoria. To unravel the program inducing appressoria in the biotrophic smut fungus Ustilago maydis, we exposed cells to a hydrophobic surface and the cutin monomer 16-hydroxy hexadecanoic acid. Genome-wide transcriptional profiling under these appressorium-inducing in vitro conditions revealed dramatic transcriptional changes in almost 20% of the genes. Comparisons with the U. maydis sho1 msb2 double mutant, lacking two putative sensors for plant surface cues, revealed that these plasma membrane receptors regulate a small subset of the surface cue-induced genes. These genes comprised mainly secreted proteins including plant cell wall degrading enzymes that facilitate plant penetration and secreted effectors that are essential virulence factors, with functions after penetration. Our data also demonstrate specific effects on two transcription factors that redirect the transcriptional regulatory network towards appressorium formation and plant penetration. Thus, plant surface cues prime U. maydis for biotrophic development. Solopathogenic Ustilago maydis strain AM1 and its derivate AM1Δsho1Δmsb2 were grown to mid-log phase in YEPSL medium and resuspended in 2% YEPSL plus/minus 16-hydroxy hexadecanoic acid (HDA). The cell suspensions were sprayed on hydrophobic surface (Parafilm) and incubated for 12 h. As control, cells were sprayed on hydrophilic glass surface and incubated for 2 h. After RNA extraction Affymetrix microarrays were performed.

Project description:The success of plant pathogenic fungi mostly relies on their arsenal of virulence factors that are expressed and delivered into the host tissue during colonization. The biotrophic fungal pathogen Ustilago hordei causes covered smut disease on both barley and oat. The biotrophic interaction of the fungus with its host plant starts with formation of appressorium and subsequent penetration of invasive hyphae into the plant cell, in which the plasma membrane of the infected cell gets invaginated and encases the penetrating hypha. After penetration, the fungus grows towards and in later stages along the vascular bundles, which contain assimilates and metabolites, representing a source of nutrients for the fungus. In this study, we combined cytological, genomics and molecular biological methods to achieve a better understanding of the molecular interactions in the U. hordei-barley pathosystem. This study focuses on the genome-wide Agilent Microarray-based transcriptome analysis of U. hordei during early stages (20 hpi, 40 hpi, 3 dpi, 6 dpi) of the biotrophic interaction in comparison to axenic culture growth. This should give insights into the molecular processes during these stages by the identification of regulated genes. Furthermore, the regulation of putative secreted enzymes and effector proteins can be studied. Transcriptome analysis of U. hordei at different stages of host infection revealed 273 effector gene candidates with differentially expressed transcript levels. Furthermore, U. hordei transcriptionally activates core-effector genes that may suppress even non-host early defense responses. It could be shown that about one half of the U. hordei open reading frames are differentially regulated during at least one of the analyzed stages. Based on the transcriptome data, some of them were identified to be involved in stage-specific processes, e.g. in the nutrient acquisition by the induction of carbohydrate transporters during the later stages of infection. Furthermore, a set of strongly induced genes was identified, which encode for secreted proteins. The acquired datasets show a strong induction of stage-specific secreted effector proteins, whose functions remain unclear. Based on the expression values, a group of 16 promising candidates was chosen for functional characterization. Project description:In fungi, sexual compatibility is controlled by mating type loci that prevent self-fertilization. In the plant pathogenic fungus Ustilago maydis, the b mating type locus encodes a pair of unrelated homeodomain proteins, termed bE and bW. After fusion of two compatible, haploid cells, the bE and bW proteins form a heterodimeric complex, but only if they are derived from different, compatible alleles. The active bE/bW complex is required and sufficient to initiate pathogenic development of U. maydis, which is prerequisite for sexual reproduction of the fungus. However, the role of the b heterodimer during later stages of pathogenic development was unclear. To analyze b function during in planta development, we generated a temperature-sensitive bE allele (bEts) encoding a protein with a single amino acid alteration at the border of the homeodomain. This mutation leads to a stop in pathogenic development at the restrictive temperature in planta, while bEts strains show normal development at permissive temperature. At restrictive temperature, hyphae develop enlarged, bulbous cells at their tips that contain multiple nuclei, indicating a severe defect in cell division. DNA array analysis of bEts mutant strains in planta revealed a b-dependent regulation of genes coding for secreted proteins that were shown to influence fungal virulence. Our data demonstrate that in U. maydis the b heterodimer is not only essential to establish the heterodikaryon after mating of two compatible sporidia and to initiate fungal pathogenicity, but also to sustain in planta proliferation and ensure sexual reproduction.
