Project description:miRNA analysis of breast epithelial cell line with stem cell properties before and after undergoing endothelial induced epithelial to mesenchymal transition (EMT). miRNA expression anlysis was done on breast epithelial cells before and after endothelial induced EMT. D492 is a breast epithelial cell line with stem cell properties that undergoes EMT in 3D rBM coculture with endothelial cells. Total RNA was isolated from D492 and D492M (a mesenchymal derivative) at 50% and 90% confluency in monolayer.

Project description:The epithelial-mesenchymal transition (EMT) is an embryonic transdiffrentiation program which consists of the conversion of polarized epithelial cells into a motile mesenchymal phenotype. EMT is aberrantly reactivated during tumor progression, promoting metastatic dissemination. Herein, we demonstrate that EMT permissive conditions also favor tumor initiation by minimizing the number of events required for neoplastic transformation. We further demonstrated that even partial commitment of human mammary epithelial cells into an EMT program is sufficient to confer malignant properties, suggesting that the reactivation of embryonic EMT inducers participate to the primary tumor growth long before the initiation of the invasion-metastasis cascade. Human mammary epithelial cells (HMEC) were sequentially depleted in p53 through RNA interference (shp53), transduced with H-RasG12V and immortalized by hTert. Two different Tert/shp53/Ras cell population emerge that display either an epithelial (Epi) or a mesenchymal (Mes) phenotype. Gene expression profiles of the Tert/shp53 control cells and of tert/shp53/Ras/Epi and Tert/shp53/Ras/Mes were analyzed.

Project description:Epithelial stem cells self-renew while maintaining multipotency, but the dependence of stem cell properties on maintenance of the epithelial phenotype is unclear. We previously showed that trophoblast stem (TS) cells lacking the protein kinase MAP3K4 maintain properties of both stemness and epithelial-mesenchymal transition (EMT). Here, we show that MAP3K4 controls the activity of the histone acetyltransferase CBP, and that acetylation of histone H2B by CBP is specifically required to maintain the epithelial phenotype. Combined loss of MAP3K4/CBP activity represses expression of epithelial genes and causes TS cells to undergo EMT while maintaining their self-renewal and multipotency properties. The expression profile of MAP3K4 deficient TS cells defines an H2B acetylation regulated gene signature that closely overlaps with that of human breast cancer cells. Taken together, our data define an epigenetic switch that maintains the epithelial phenotype in TS cells and reveal previously unrecognized genes potentially contributing to breast cancer. Three separate trophoblast stem (TS) cell conditions were compared to define the gene expression changes that occur with the induction of epithelial-mesenchymal transition (EMT) in TS cells. These conditions were TS cells differentiated for 4 days (T^Diff), TS cells differentiated for 4-days and isolated following invasion through Matrigel (T^Inv), and TS cells with an inactive MAP3K4 (TS^KI4). All conditions were normalized to wild-type control TS cells (TS^WT). T^Diff and T^Inv were analyzed in triplicate. TS^KI4 was analyzed in duplicate in two independent biological replicates.

Project description:Epithelial-to-mesenchymal transition (EMT) allows progressive loss of epithelial traits and acquisition of mesenchymal identity promoting cell migratory and invasive properties. It is expressed by changes in gene expression program induced by changes of chromatin state and organization. To identify epigenetic landscape associated with EMT we performed chromatin immunoprecipitation (ChIP) experiments allowing nucleosome profiling (H3), distribution of histone modifications linked to actively transcribed (H3K4me3, H3K36me3 and H3K27ac) and repressed (H3K27me3) regions in epithelial (Epi) and mesenchymal (Mes) cells. These cell lines were generated by immortalization of primary HEK cells before and after naturally occured EMT.

Project description:Epithelial to Mesenchymal Transition (EMT) has been associated with cancer cell heterogeneity, plasticity and metastasis. It has been the subject of several modeling effort. This logical model of the EMT cellular network aims to assess microenvironmental signals controlling cancer-associated phenotypes amid the EMT continuum. Its outcomes relate to the qualitative degrees of cell adhesions by adherent junctions and focal adhesions, two features affected during EMT. Model attractors recover epithelial, mesenchymal and hybrid phenotypes, and simulations show that hybrid phenotypes may arise through independent molecular paths, involving stringent extrinsic signals.
Of particular interest, model predictions and their experimental validations indicated that: 1) ECM stiffening is a prerequisite for cells overactivating FAK-SRC to upregulate SNAIL1 and acquire a mesenchymal phenotype, and 2) FAK-SRC inhibition of cell-cell contacts through the Receptor Protein Tyrosine Phosphates kappa leads to the acquisition of a full mesenchymal rather than a hybrid phenotype.

Project description:The series of events that allows for the conversion of adherent epithelial cells into migratory cells is collectively known as epithelial-mesenchymal transition (EMT). EMT is involved in triggering neural crest migration during development and in the pathogenesis of diseases, such as cancer metastasis. Whereas Erk signalling is known to be essential for EMT, its influence on the epigenetic and transcriptional programme underlying EMT is poorly understood. Here, using a comprehensive genome-wide analysis of H3K27ac mark and gene expression in primary mammary epithelial cells undergoing EMT, we found that Erk signalling is essential for the epigenetic reprogramming underlying hallmark gene expression and phenotypic changes of EMT. We found that the chemical inhibition of Erk signalling during EMT prevents loss and gain of H3K27ac mark at regulatory regions of epithelial and mesenchymal genes, respectively, and results in a transcriptome and epigenome closer to those of epithelial cells. Further computational analyses identified a distinct set of transcription factor motifs enriched at distal regulatory regions that are epigenetically remodelled by Erk signalling. Altogether, our study reveal an Erk-dependent epigenetic remodelling of distal regulatory elements that results in a gene expression programme that is essential for driving EMT.

Project description:Cardiac fibrosis is a detrimental pathophysiological state involved in a number of cardiovascular diseases. Myofibroblasts mediate fibrosis by excessive remodeling of the extracellular matrix, which ultimately leads to tissue stiffness and impaired heart performance. Recently, it was shown that a substantial fraction of cardiac myofibroblasts may originate from the epicardium through Epithelial-to-Mesenchymal Transition (EMT). We have developed a cellular model of EMT in which adult murine epicardium-derived cells are differentiated into myofibroblast-like cells in the presence of Interleukin-1beta, Tumor Necrosis Factor-alpha, or Transforming Growth Factor-beta. Using this model of EMT, the microRNAome was assessed by microRNA (miRNA) arrays. Subsequently, expression levels of differentially expressed miRNAs were validated by qPCR. These miRNAs were targeted by transfecting epicardium-derived cells with anti- or pre-miRs prior to EMT initiation. The ability of the anti- or pre-miRs to inhibit EMT was assessed on a number of phenotypic markers. In this study we have identified a number of miRNAs that potentially play an intrinsic role in cardiac EMT. We speculate that by targeting those miRNA, the onset and long-term progression of cardiac fibrosis can be substantially reduced. Epicardial mesothelial cells were isolated and expanded from the epicardium of adult rats (8-10 weeks). Epithelial-to-mesenchymal Transition was induced by 10 ng/mL Interleukin-1beta, Tumor Necrosis Factor-alpha, or Transforming Growth Factor-beta1 for 48h. The assocciated differential microRNA expressions relative to a control treatment was computed by microRNA arrays. The experiment was conducted on biological quadruplicates for the control treatment and biological triplicates for cytokine treatments.
