Real-time quantitative PCR analysis of mouse lungs

ABSTRACT: 2 IL-15 KO AND 2 C57BL/6 mice were injected with 5X10^5 cells of the polyoma Middle T (pMT) primary breast tumor cell line intraveneously(IV). 2 days post injection, the lungs were harvested and one lobe of the lung was used to extract RNA from. We then utilized the SABBiosciences RT^2 Profiler PCR Array for Mouse cytokines and Chemokines to determine levels of cytokine/chemokines that were different in these 2 mice at this time point. We were interested in the immune responses that may change in response to tumor cells entering and establishing in the lung (as a model of metastasis) in the presence or absence of IL-15.

Project description:Previous stimulation experiments of stimulated primary murine colonic fibroblasts with IL-36R ligands for 4h in vitro showed an upregulation of pro-inflammatory cytokines e.g. IL-6, IL-1b and chemokines e.g. CXCL1, CCL2 indicating an important role for IL-36 in inflammation. We were interested in analyzing long-term stimulation (9 days) of intestinal fibroblasts to identify a putative differential expression profile. The experimental setup included 6 samples from 3 colons in a pairwise design (3 stimulated vs 3 unstimulated).

Real-time quantitative PCR analysis of lungs from Litomosoides sigmodonis-infected mice

Project description:A pool of cDNA from naive BALB/c mice lungs (n=8) was compared with one from D70 p.i. Litomosoides sigmodontis-infected mice (n=8) by profiling 84 cytokine-related genes simultaneously. Screening of inflammatory lung environment was performed with a qRT-PCR array (Mouse Cytokines & Chemokines RT2 Profiler PCR Array, Qiagen, Germany) according to manufacturer’s instructions.

Project description:We report the differences of gene expression pattern of 4 different groups of mouse lung CD45- cells (Naive mouse without IL-15/IL-15Ra complex, Naive mouse with IL-15/IL-15Ra complex, MCMV-infected mouse without IL-15/IL-15Ra complex, and MCMV-infected mouse with IL-15/IL-15Ra complex). We found that mouse with MCMV-memory CD8 T cells response to IL-15/IL-15Ra complex and the behaviors of cancer were changed compared to other groups.

Expression data of MMTV-PyMT mice mammary tumor with or without JAM-A

Project description:Junction Adhesion Molecule-A (JAM-A) is present on leukocytes and platelets where it promotes cell adhesion and motility. We are interested in an interaction between JAM-A and tumor progression/metastases. To address this point, we mated JAM-A-/- mice and mouse mammary tumor model MMTV-PyMT mice which, which express polyoma middle T antigen under the control of mouse mammary tumor virus. MMTV-PyMT mice show 100% penetration of mammary tumor and highly metastases to lung. MMTV-PyMT mice without JAM-A show less primary tumor progression, therefore JAM-A enhance primary tumor progression. Then we are addressing the molecular mechanism of this phenomenon by in vivo. Furthermore, we would like to examine JAM-A deficient MMTV tumor signature.

Project description:GM-CSF receptor-β deficient (Csf2rb–/– or KO) mice develop a lung disease identical to hereditary pulmonary alveolar proteinosis (hPAP) in humans with recessive CSF2RA or CSF2RB mutations that impair GM-CSF receptor function. We performed pulmonary macrophage transplantation (PMT) of bone marrow derived macrophages (BMDMs) without myeloablation in Csf2rb–/–mice. BMDMs were administered by endotracheal instillation into 2 month-old Csf2rb–/– mice. Results demonstrated that PMT therapeutic of hPAP in Csf2rb–/– mice was highly efficacious and durable. Alveolar macrophages were isolated by bronchoalveolar lavage one year after administration subjected to microarray analysis to determine the effects of PMT therapy on the global gene expression profile.

Real-time quantitative PCR analysis of human low-density (LDNs) and normal-density neutrophils (NDNs)

Project description:NDNs and LDNs were isolated by density gradient centrifugation of PBMCs from 8 patients affected by tubercolosis (TB). In detail, RNA was extracted from NDNs and LDNs by Maxwell® RSC miRNA Tissue kit (Promega, Madison, USA), following manufacturer’s instructions. RNA concentration and quality was measured using NanoDrop 2000 Spectrophotometer (Thermo Fisher Scientific). Total RNA (600 ng) was reverse transcribed using the RT2 First Strand Kit (SABiosciences, Qiagen, UK). 84 human cytokines and chemokines were tested using the RT2 Profiler PCR Array Human Innate & Adaptive Immune Responses array (Qiagen). A semiquantitative real-time RT-PCR was performed using a real-time PCR detection system (QuantStudio 7 Flex Real-Time PCR System, Thermo Fisher Scientific) with a two-step thermal cycling: 95 °C for 10 min, followed by 40 cycles (95 °C for 15 sec, 60 °C for 1 min). Data were analysed using the RT² Profiler PCR Array Qiagen data analysis software, and ACTB and B2M as housekeeping genes.

Project description:We are interested in having the gene microarray done on untreated and PMA treated (10 nM x 24 hr) human and mouse epithelial cells lines including CALU-3, MLE-15, primary bronchial human airway epithelial cells. We are especially interested in seeing whether sialyl or sulfo transferases, and MUC genes, are present and if they are induced by PMA. We hope to use this information to optimize future experiments designed to identify Siglec lung ligands. The lab is interested in studying untreated and PMA treated (10 nM x 24 hr) human and mouse epithelial cells lines including CALU-3, MLE-15, primary bronchial human airway epithelial cells. With special interest in understanding whether sialyl or sulfo transferases, and MUC genes, are present and if they are induced by PMA.
