Primary and adjacent non-malignant tissue samples of 100 HCC patients (mRNA)

ABSTRACT: Genome-wide mRNA expression profiles of 100 primary and matched non-malignant tissues of HCC patients from a Singapore patient cohort. HCC is a hetergenous disease and it is important to understand the underlying molecular mechanisms. Here, we studied the role of microRNAs in HCC by integrating microRNA and gene expression profiles of HCC patients.

Project description:Hepatic cirrhosis or advanced fibrosis caused by chronic hepatitis may be the main risk factor contributing to the occurrence of HCC. Growing clinical evidence show that patients with hepatic cirrhosis or advanced fibrosis may have a 7-fold higher incidence rate of HCC than those without, suggesting a crucial role of inflammation-carcinogenesis malignant transformation in HCC development and progression. Applied Biosystems Human TaqMan Low Density Array (TLDA) in HCC tumor and adjacent non-tumor tissues

Project description:MicroRNAs (miRNAs) are aberrant expressed in hepatocellular carcinoma (HCC) tissue and play a central role in diverse biological processes. We conducted a genome-wide miRNAs screening in 10 pairs of HCC tumor and adjacent non-tumor tissues to test the hypothesis that dysregulation of miRNAs in HCC tumor tissue are partially due to aberrant methylation in relevant miRNAs host genes. Taqman low density arrays were used to examine miRNA profiles in paired HCC tissues, and quantitative RT-PCR was used to validate candidate miRNAs for both discovery and validation sets. A cross-sectional study was conducted in 10 HCC tumor tissues and 10 adjacent non-tumor tissues in Columbia University Medical Center (CUMC), which is approved by the Institutional Review Board.

Project description:Current selection criteria for liver transplant in patients with HCC (the Milan criteria) do not incorporate biologic metrics. MiRNA expression profiles correlate with HCC recurrence after transplant and can add significant value to the Milan criteria. MiRNA expression profiles were studied in HCC specimens from patients undergoing liver transplant and correlated with their tumor recurrence status and Milan criteria status.
