Expression data from wild-type (WT) and lrrc19 gene knockout (KO) mice intestines

ABSTRACT: LRRC19 (Leucine repeat receptor containing 19) is a newfound Toll-like receptor without definite function reported. We have detected that the lrrc19 gene had a high expression level in both human and mouse intestines and concentrated on the effect of this immune receptor in mediating the process of enteritis and intestinal cancer. To have a systematic understanding on the influence of this gene on the intracelluar environment of intestinal cell, we have conducted a microarray experiment to compare the transcriptomes of the WT and KO mice intestines under the same experimental condition.

To have a systematic understanding on the influence of this gene on the intracelluar environment of intestinal cell, we have conducted a microarray experiment to compare the transcriptomes of the WT and KO mice intestines under the same experimental condition. Gut epithelial cells of WT and KO mice of the same physiological and maintaining condition were isolated and then total RNAs were extracted from them, which could reflect the respective transcription profile of WT and KO mice. These RNAs were detected by microarray and the influence of lrrc19 gene on intestine could be revealed to some extent.

Transcription profiling of mouse small intestines

Project description:Rb and E2F are thought to play antagonistic roles in celll proliferation. However, this model is based mostly from in vitro cell culture systems. We used small intestines to test this model in vivo. We found that deletion of E2f1-3 in the small intestine of mice suppressed the ectopic expression of E2F targets and cell proliferation caused by Rb-deficiency. Surprisingly, E2f1-3 deletion failed to arrest the proliferation of intestinal cells containing an intact Rb gene, and instead led to E2F target derepression and apoptosis. Experiment Overall Design: Total RNA of crypts and villi from wild-type, Rb-/-, E2f1-/-, E2f2-/-, E2f3-/-, E2f1-/-, E2f2-/-, and E2f3-/- small intestines. Small intestines were harvested 7 days after mice were injected intraperitoneally with beta-napthoflavone.

RNA-seq mouse intestines from PN, Germ-free, SPF and Antibiotics-treated mice

Project description:We hypothesized that there were genes involved in the maintenance of intestinal tolerance to commensal bacteria. These genes should meet the following criteria: expressed in intestines, upregulated in the presence of commensal bacteria and downregulated in absence of microbes. To find these genes, we collected small intestines from postnatal mice that neither affected by the bacteria nor by the food, adult mice bred in germ-free or specific pathogen-free conditions, and adult SPF mice treated with antibiotics. We therefore performed bulk RNA sequencing using these cells.

The ChIP-Sequence data of STAT92E in Drosophila adult intestines

Project description:JAK/STAT pathway plays important roles in controlling Drosophila intestinal homeostasis and regulating the ISC proliferation and differentiation. However,the downstream targets of its transcription factor-STAT92E remain largely unknown.To further identify the regualtory mechanisms of the JAK/STAT pathway in controlling intestinal homeostasis,we performed the ChIP-Seq assay with mouse raised STAT92E antibody using JAK/STAT signaling highly activated adult intestines.Through the ChIP assay, we have identified over 1000 significant peaks (p<0.01) around the putative targets.The well-characterized JAK/STAT downstream targets including Domeless,Socs36E,STAT92E and chinmo were identified in our ChIP assay,indicating that our experiment is workable to identify novel JAK/STAT downstream targets in adult intestines.This work will provide insights into our understanding of regulatory mechanisms of JAK/STAT signaling during Drosophila intestinal development. Culture conditions for each intestinal segment were optimized by correlating monolayer global gene expression with the corresponding tissue segment. The monolayers polarized, formed tight junctions, and contained a diversity of intestinal epithelial cell lineages.
