Interleukin-6 modifies the expression profile of microRNA in mouse 3T3-L1 adipocytes

Project description:In the present study we examined the expression profile of microRNA in mouse 3T3-L1 adipocytes treated with IL-6, a known inflammatory cytokine and an adipo-myokine. The miRNA microarray analysis revealed increased expression of 9 miRNA species in adipocytes exposed to IL-6 (1 ng/ml) for 8 days of differentiation. In order to verify the microarray data, qRT-PCR analysis was performed to quantify miRNA expression and we confirmed the increased expression of miR-146b-3p (fold change =1.49±0.17). Using Target Scan database we found potential targets for these miRNAs with expression modified by IL-6 and focused our further interest on genes/proteins involved in cytoskeleton organization and vesicle transport regulation, that is: actin, dynein, myosin light chain kinase, Rab4a, and glucose transporter Glut4.

Project description:Obesity is an energy balance disorder in which nutrient intake chronically exceeds energy expenditure, resulting in the accumulation of white adipose tissue. Increased adiposity is due to increases in the number and size of adipocytes, which leads to increased body fat and metabolic consequences. Adipocytes induce insulin resistance by promoting lipotoxicity and modulating adipokine secretion. Therefore, a thorough understanding of the mechanisms that regulate adipogenesis could have clinical relevance in preventing and treating obesity and the metabolic syndrome. In this study, we performed miRNA array to measure miRNA profiles of undifferentiated and differentiated 3T3-L1 adipocytes using Agilent(r) miRNA array. We show that miRNA profile changes during adipogenesis. Thus, this data would be useful to find the distinct role of miRNAs for the regulation of adipogenesis.

Project description:Purpose: The goals of this study are to compare NGS-derived 3T3-L1-NC transcriptome profiling (RNA-seq) to LIGHT overexpression 3T3-L1 cells and find out the DEGs Methods: mRNA profiles of 3T3-L1-NC and 3T3-L1-LIGHT before and after differentiation into beige adipocytes were generated by deep sequencing, in triplicate, using Illumina HiSeq. The sequence reads that passed quality filters were analyzed at the transcript isoform level. Results: Using an optimized data analysis workflow, we mapped about 6.53 Gb data per sample. The average genome mapping rate is 87.27% and the average gene mapping rate is 80.18%. 18,255 genes were identified in which 17,367 of them are known genes and 1,001 of them are novel genes. 13,086 novel transcipts were identified in which 9,304 of them are previously unknown splicing event for known genes, 1,001 of them are novel coding transcripts without any known features, and the remaining 2,781 are long noncoding RNA. Conclusions: Our study represents the first detailed analysis of the impact of LIGHT on gene expression in process of beige adipocytes biogenesis with biologic replicates, generated by RNA-seq technology.
