After measuring the respective relative water content, RNAs were isolated from leaves of moderately and severely droughted plants and from control plants, and Illumina RNA sequencing was performed to analyze RNA synthesis in these tissues.

Transcriptional profiling of leaves of poplar plants subjected to 8 or 13 days of water deprivation. Sequence data were generated by deep sequencing, on four replicates (2 per 2 hybrids), using Illumina HiSeq2000.

RNAseq of drought treated Populus trichocarpa

Project description:To identify genes that are drought-responsive we conducted drought (soil water depletion) experiments on 3-month-old *P*. *trichocarpa *clonal plants. The plants undergo five different stages based on the appearance of their shoots and leaves during the drought experiments. Stage I: The shoot and leaves are green, and the leaves are well-spread. Stage II: The leaves are droopy. Stage III: The shoot is droopy, and the leaves are partially dry. Stage IV: The leaves are brown and totally dry. Stage V: The shoot is brown. With fully irrigation, the soil water content is 74% and the xylem water content is 80.6%. Plants in Stage III (Day 5) are under a mild drought state. The soil and xylem water content in Stage III dropped to 33% and 75.3%, respectively. Stage IV (Day 6-10) is a severe drought state where the soil and xylem water content continued decreasing to 29% and 74.3%, respectively in Day 7. The stressed plants from Stage I-IV could all recover in 3 days after rehydration, but the plants in Stage V could not recover after rehydration.

We use the Affymetrix Poplar GeneChip to decrypt the gene functions and mechanisms in Populus simonii leaves and detail the global program of gene expression during water deficit treatments. Populus simonii leaves were taken from short-term water-deficit-treated plants and control plants for RNA extraction and hybridization on Affymetrix microarrays. D1 and D2 are from water-deficit-treated plants, CK1 and CK2 are controls.

Identification of rust-induced genes in poplar using Populus 2.0 NimbleGen oligoarray

Project description:Oligoarray expression profiling was carried out in poplar leaves upon infection with rust in order to identify genes expressed during tree defense response. For this purpose, we inoculated detached leaves of the interamerican hybrid poplar Populus trichocarpa x Populus deltoides 'Beaupré' grown in greenhouse either with spores of avirulent strain 93ID6 (incompatible interaction I48) or spores of virulent strain 98AG31 (compatible interaction C48) of the pathogenic rust fungus Melampsora larici-populina. Besides, we mock-inoculated 'Beaupré' leaves with water (control condition, T48). Detached leaves were maintained in vitro in controled conditions to allow fungal infection and colonization of plant tissue. Populus Drought Stress - Quantitative proteome profile of water deficit stress responses in eastern cottonwood (Populus deltoides) leaves

Project description:We sought to generate a comprehensive proteomic profile for a Populus plant that captures protein abundance behavioral differences in mature leaves under various water deficit conditions. The sudden infliction of severe drought is unlikely to reflect what naturally happens to soil-grown plants. Therefore, rather than exposing plants to severe dehydration via the application of concentrated osmotica (e.g., polyethylene glycol or mannitol), we aimed to monitor plants exposed to prolonged drought as well as plants frequently experiencing cyclic drought. For the progressive drought experiment, herein referred to as acute drought, water will be withheld from the plants for a certain period of time until symptoms of wilting are observed. From this drought treatment, protein abundance changes that occur because of declining water potentials that consequently lead to worsening plant health will provide new insights to biological pathways in response to severe drought conditions. On the other hand, as a consequence of simulating a more natural condition, the cyclic drought treatment will provide novel insights into drought recovery and coping mechanisms. As first noted in a meta-analysis of microarray experiments comparing different water deficit-related treatments3, one consequence of analyzing multiple treatment conditions is that we expect only a few differentially abundant proteins to be common to both treatments. Therefore, we implemented a bioinformatic framework to provide an overall integrative picture of the conserved drought markers but also meaningful divergences in functional behavioral responses between the two experiments.

Project description:Populus euphratica is a natural population grown in semirid areas. The molecular response of the poplar to drought maintain to be elucided, especially at global genome level. We used Affymetrix poplar genome genechip microarrays to analyze the full transcript expression underlying different drought intensities and identified significantly differently expressed genes during this process. Uniformly developed seedlings of P. euphratica grown in gradually long-term drought throught water-withholding treatment. Affymetrix poplar genechip was hired to investigate the full transcripts changed of the poplar response to different drought intensity levels.
