Single cell RNASeq of whole mouse kidneys from Uromodulin/Tamm horsfall protein knock-out mice

Project description:To elucidate changes in cellular populations in the ischemic acute kidney injury, we performed single cell RNAseq on mouse kidneys following ischemic reperfusion injury and a sham surgery. Mice were subjected to bilateral clamping of the renal pedicule for 30 minutes. After 6-7 hours of reperfusion the kidneys were removed and processed by a MACs cell isolation protocol and dead cell removal (PMID:33448928) , sorted on a 10X Chromium platform version 3 and libraries generated. Sequencing was performed on an Illumina NovaSeq 6000.

Gene expression from microdissected glomeruli and tubulointerstitium of NZM2328 mice

Project description:To characterize molecular profiles through the development of lupus nephritis and identify risk factors for end organ damage, we carried out gene expression analysis of microdissected kidneys from lupus-prone NZM2328 mice. We identified a continuum of inflammatory processes associated with the progression from acute inflammatory to chronic destructuve disease initiated in the glomeruli and progressing to the tubulointerstitium. We examind male mice and the congenic NZM2328.R27 strain, both of which are resistant to the development of chronic nephritis and end organ damage, as a means to define pathogenic processes.

Project description:Using whole genome bisulfite sequencing (WGBS), we investigated dynamics of DNA methylation and chromatin changes during mouse lens fiber and epithelium differentiation between embryos (E14.5) and newborns (P0.5) using microdissected lenses. Histone H3.3 variant chromatin landscapes were also generated by ChIP-seq using microdissected newborn lenses.
