Project description:Here we reveal evidence of a shared genetic toolkit across the full spectrum of social complexity found in Vespid wasps, from simple group living where castes remain plastic throughout life, to complex superorganismal societies comprised of mutually dependent insects with irreversible castes determined during development. We generated brain transcriptomic data for castes in nine representative species; using a machine learning approach we identified thousands of shared orthologs which consistently describe castes (queens and workers), from species with the simplest (Mischocyttarus paper wasps) to the most complex (e.g. Vespine wasps) levels of social organisation. The top 400 genes were enriched in synaptic transport genes, suggesting that these changes could affect brain neural function and connectivity. Fine-scale dissection of these patterns revealed that the molecular processes underpinning the simpler societies (which likely represent the origins of social living) are conserved throughout the major transition, but that additional processes may come into play in the more complex societies, especially at the point of no return where societies transition to be committed superorganisms. These analyses provide the first evidence of a conserved toolkit regulating social behaviour across the full spectrum of social complexity in any social insect. Importantly, they also provide evidence that there may be fundamental differences discriminating superorganismal societies from non-superorganismal societies. We suggest that the evolution of irreversible caste commitment (in superorganisms) is accompanied by a fundamental shift in the underlying regulatory molecular machinery; such shifts may also typify other major evolutionary transitions that are characterised by the emergence of a committed division of labour, such as the evolution of multicellularity.

Project description:Specific genes or encoded proteins are involved in regulating various learning models of different species through certain signaling pathways，but whether there are also regulatory genes during bimodal learning and memory is largely unknown. Using a multi-omics approach to examine gene expression changes in bees brain performed with three different learning assays, a general up-regulation of genes and proteins were observed in bimodal learning compared to controls. Protein-protein network predictions of differential proteins together with FISH assays suggest ALDH7A1 may be involved in regulation of bimodal learning and memory. Injecting siRNA-ALDH7A1 to the bee brain results in significant inhibition the expressions of ALDH7A1 and regucalcin, and increase β-alanine content. Interestingly, we found that loss of ALDH7A1 only affect visual-olfactory bimodal learning and memory, but not single visual or olfactory conditioned learning after ALDH7A1-RNAi in bees. Therefore, our data suggests that ALDH7A1 may affect bimodal learning and memory though controlling β-alanine related plasticity mechanisms.

Project description:Aggressive competition is an important aspect of social interactions, but conflict can be costly. Some animals are thought to minimize the costs of conflict by using conventional signals of agonistic ability (i.e. badges of status) to assess rivals. Although putative badges have been found in a range of taxa, little research has tested whether individuals use badges to assess potential rivals before they engage in aggressive contests. Here, choice trials were used to test how the variable black facial patterns in Polistes dominulus wasps are used during rival assessment. Focal wasps were given access to two patches of food, each guarded by a wasp whose facial pattern had been experimentally altered. Wasps chose food patches based on the facial pattern of the guard, preferring to challenge guards with facial patterns indicating a low level of quality, while avoiding guards with facial patterns indicating a high level of quality. Therefore, status badges play an important role during rival assessment; paper wasps use facial patterns alone to quickly assess the agonistic abilities of strangers.

Project description:Gene expression profiles were generated from 199 primary breast cancer patients. Samples 1-176 were used in another study, GEO Series GSE22820, and form the training data set in this study. Sample numbers 200-222 form a validation set. This data is used to model a machine learning classifier for Estrogen Receptor Status. RNA was isolated from 199 primary breast cancer patients. A machine learning classifier was built to predict ER status using only three gene features.

Project description:A theoretical framework for the function of the medial temporal lobe system in memory defines differential contributions of the hippocampal subregions with regard to pattern recognition retrieval processes and encoding of new information. To investigate molecular programs of relevance, we designed a spatial learning protocol to engage a pattern separation function to encode new information. After background training, two groups of animals experienced the same new training in a novel environment, however only one group was provided spatial information and demonstrated spatial memory in a retention test. Global transcriptional analysis of the microdissected subregions of the hippocampus exposed a CA3 pattern that was sufficient to clearly segregate spatial learning animals from control. Individual gene and functional group analysis anchored these results to previous work in neural plasticity. From a multitude of expression changes, increases in camk2a, rasgrp1 and nlgn1 were confirmed by in situ hybridization. Furthermore, siRNA inhibition of nlgn1 within the CA3 subregion impaired spatial memory performance, pointing to mechanisms of synaptic remodeling as a basis for rapid encoding of new information in long-term memory. Experiment Overall Design: RNA samples from animals subjected to a spatial learning paradigm were compared to controls using Affymetirx RAE230a chips. An N of 7 was used in each of the two experimental conditions.

Project description:We are searching for common gene expression alteration in the hippocampus of animal models of learning disabilities and in post-mortem human tissues derived from patients with cognitive deficits. Our study has three phases. In phase I, we want to investigate the changes in gene expression in a transgenic mouse line that shows impairments in hippocampal-dependent learning. These animals are overexpressing the RNA-binding protein HuD in the forebrain under the control of the alpha CAMKinII promoter. For phase II, we plan to characterize the hippocampus of another animal model of learning disabilities: fetal alcohol exposed (FAE) rats. Last, for phase III, we would like to examine gene expression changes in the hippocampus and other tissues from patients with schizophrenia. To test our hypothesis, in phase 2 of our project, we propose to compare the pattern of gene expression in our mouse model with that in the hippocampus of rats prenatally exposed to alcohol (FAE model). Our hypothesis is that learning disabilities have common neural substrates regardless of the origin of the disability being genetic, environmental of a combination of these two. To test this hypothesis, we plan to examine the pattern of gene expression in the hippocampus of animals and humans with cognitive deficits and use bioinformatics tools to identify common changes in gene expression in three of such conditions: a genetic model (HuD transgenic mice), an environmental model (FAE rats) and a model for combined environmental and genetic effects (patients with schizophrenia). All the animals are adult male Sprague-Dawley rats (age 90-100 days). Animals were divided in three diet groups and three experimental treatment conditions (9 conditions altogether). Three groups of pregnant rat dams were placed on different diet regimens: a liquid diet containing 5% alcohol (A) groups, a control group fed an isocaloric liquid diet without alcohol (B) and a group of rats fed lab chow ad libitum (C). Offspring from each of these groups were allowed to grow until adulthood and placed into each of three training groups: a contextual fear conditioned group (1), a non-learning stress control group (2) and a naive/unhandled group (3). Hippocampi were dissected and used to isolate total RNA from each rat diet and training condition. Initially, we will submit 18 samples as follows: 10 samples from training group 1 and 8 samples from training group 3 (training group 2 will be analyzed at a later date). The diet assignment for the 18 samples is: 8 samples from group A, 6 samples of group B and 4 samples of group C. Please note that I was able to enter only the first 8 samples, as the sample list does not hold 18 samples altogether.
