Project description:Analysis of TH17 cells redirected with chimeric antigen receptors (CAR) expressing various signaling domains (including CD28, 4-1BB and ICOS) after surrogate antigen stimulation. Our results showed that T cells redirected with an ICOS-based CAR specifically retained a genotype of TH17 cells with expression of Il17a, Il17f, Il1r1, Ccl20, Rorc, and in the absence of Foxp3 CAR-redirected TH17 cells from three different human normal donors were stimulated with immobilized recombinant mesothelin. Gene expression levels were determined prior to stimulation (day 0) and 4, 8, 24 and 96 hours upon antigen recognition.

Combinatorial treatment with PARP and MAPK inhibitors to overcome phenotype switch-driven drug resistance in advanced melanoma

Project description:Metastatic melanoma is either intrinsically resistant or rapidly acquires resistance to targeted drugs such as MAPK inhibitors (MAPKi). Here, using a drug screen targeting chromatin regulators in patient-derived 3D melanoma cell cultures, we discovered that PARP inhibitors are capable of restoring MAPKi sensitivity. This synergy was found to be independent of DNA damage repair pathways and was effective both in vitro and in vivo in patients-derived xenografts. Strikingly, through integrated transcriptomic, proteomic and epigenomic analysis, we discovered that PARPi induces lysosomal autophagy which was accompanied by enhanced mitochondrial lipid metabolism that, ultimately, increased antigen presentation and sensitivity to T-cell cytotoxicity. Moreover, we also found that PARP inhibitors regulated EMT-like phenotype switching by dampening the mesenchymal phenotype via transcriptomic and epigenetic rearrangements. This, in turn, redirected melanoma cells towards a proliferative and, thus, MAPKi-sensitive state. Our study provides a scientific rational for treating patients with PARPi in combination with MAPKi to annihilate acquired therapy resistance.

T cell dependent immune responses: B cell activation and differentiation Group1 Fo, Group2 GC1, Group3 GC2

Project description:Upon immunization with a T cell dependent antigen naive follicular B cells (Fo) are activated and a germinal center reaction is induced. Within the next 2 weeks large germinal centers develop where the process of affinity maturation takes place. To analyze the gene expression profile of resting and activated B cells, follicular B cells (Fo), B cells from early (GC1) and late germinal centers (GC2) were isolated and their gene expression profile compared. Gene expression profiles of Fo versus GC1 and GC2 B cells, respectively. Naïve Fo B cells were isolated from non-immunized BALB/c mice. Germinal center B cells sorted from spleen cell suspensions of BALB/-c mice immunized with the T cell dependent antigen 2-phenyl Oxazolone. GC1 B cells were isolated 7 days after primary immunization. GC2 cells were isolated 15 days after primary immunization. After total RNA extraction, reverse transcription, cDNA extraction, the biotinylated cRNA was transcribed, fragmented, and 15 µg cRNA hybridized in duplicates for each of the three groups to the GeneChip arrays. Group1: Fo, Group2: GC1, Group3: GC2. Lists of differentially regulated genes were created using High Performance Chip Data Analysis (HPCDA) with Bioretis database (http://www.bioretis-analysis.de). IL12 integrated into the CAR exodomain converts CD8+ T cells to poly-functional NK-like cells with superior killing of antigen-loss tumors

Project description:Chimeric antigen receptor (CAR)-redirected T cell therapy often fails to control tumors in the long-term due to selecting cancer cells that down-regulated or lost CAR targeted antigen. To reprogram the functional capacities of CAR T cells, we inserted IL12 into the extracellular moiety of a CD28- CAR; both the CAR and IL12 were functional indicated by antigen-redirected activation and STAT4 phosphorylation, respectively. CD8+ T cells engineered with a IL12-CAR gained a so far not recognized NK cell-like RNA expression signature and a CD94+CD56+CD62Lhigh phenotype closely similar, but not identical, to NK and cytokine activated killer (CIK) cells. IL12-CAR T cells with specificity for CEA additionally acquired antigen-independent, HLA-E restricted cytotoxicity eliminating both CEA-negative and CEA-positive cancer cells in an antigen-dependent and independent fashion that was in contrast to conventional CEA CAR T cells. Simultaneous signaling through CD3 of the CAR and through IL12 were required and sufficient for inducing NK-like cytotoxicity. Overall, we present a prototype of a new family of CARs that harbor a cytokine integrated into the extracellular module in order to reprogram the functional capacities of CAR T cells towards augmented tumor recognition and elimination.

Project description:Specific metabolic program is required for generation of immune cells and their proper function. Since the description of age-associated or autoimmune-associated B cells (ABCs), there has been a growing interest in the role of these cells in autoimmunity. ABCs have many distinct functional properties, including antigen presentation to T cells and autoantibody production in lupus mouse models. Metabolic properties which support these functions are unknown in ABCs. In this study, we showed that ABCs are metabolically hyperactivated than follicular (FO) B cells. There was an increased glycolysis and oxidative phosphorylation (OXPHOS) in ABCs. In line with this observation, ABCs express key costimulatory molecules for T cell activation with distinct group of cytokines. T cell proliferation and activation were induced by both FO B and ABCs in antigen-dependent manner. However, ABCs induce stronger T cell receptor signaling from naïve CD4+ T cells, leading to differentiation of follicular helper T (Tfh) cells. ABCs actively internalize OVA protein and increased autophagy formation. Treatment of ABCs with metformin suppressed antigen presentation by decreasing antigen uptake, leading to a defective generation of Tfh cells. Taken together, these findings reveal distinct metabolic activity in ABCs and define fundamental connection between metabolism and function within ABCs.
