ABSTRACT: To investigate which genes are up- or down-regulated by aptf-1 transcription factor in C. elegans, aptf-1(gk794) mutants pretzel-stage embryos were used to performe whole genome microarray expression profiling using the C. elegans (V2) Gene Expression Microarray, 4x44K from Agilent Technologies.

Project description:To investigate which genes are up- or down-regulated during L4 sleep in C. elegans mutant aptf-1(gk794) we performed whole genome microarray expression profiling using the C. elegans (V2) Gene Expression Microarray, 4x44K from Agilent Technologies. Approximately 200 sleeping L4 worms were used per sample. As a control we have used N2 C. elegans wild type strain. Mutant condition (aptf-1(gk794)) and control (N2) were done in four replicates.

Transcriptional response to ethanol exposure in Caenorhabditis elegans measured by microarray

Project description:This study investigated the transcriptional response of C. elegans after 0, 30, 60, 120, and 480 minutes of ethanol or mock exposure (n=4 per timepoint and treatment). Age synchronized Bristol N2 animals were obtained by allowing fertile adults to lay eggs for 3 hours. After which adults were removed and the population was incubated for 3 days at 20 ˚C. After ethanol exposure, RNA was isolated, labeled and hybridized on Agilent C. elegans (V2) Gene Expression Microarray 4X44K slides.

Project description:Transcriptional profiling of early C. elegans embryos comparing control (N2) embryos with mes-2 mutant embryos at different developmental stages: 2E (24-40 cells), 4E (50-90 cells) and 8E stage (100-200 cells). Goal was to determine the effects of mes-2 loss on global gene expression as embryos transit from a developmentally plastic state (2E stage) to the onset of differentiation (8E stage). Our microarray data showed that early-expressed genes remain active, differentiation genes fail to reach wild-type levels in mes-2 mutant embryos at the 8E stage.
