SynapTRAP on SNAP25 TRAP Cortex

ABSTRACT: SynapTRAP. Identification of Synaptic mRNA of neurons of the cortex. Technique combines sucrose percoll fractionation of a synaptically rich sample (SN) and TRAP tagged ribosome IP (PreIP and PostIP). This experiment uses pan neuronal SNAP25 mice and a cortical dissection.

Upregulation of SNAP25 by HDAC inhibition ameliorates NPC disease phenotypes via autophagy induction

Project description:Niemann-Pick Type C disease (NPC) is a rare fatal neurodegenerative disorder caused by mutations in NPC1 and NPC2 gene leading to abnormal accumulation of unesterified cholesterol in lysosomes. There have been no available treatments and FDA approved drugs for treating NPC. SAHA, a HDAC inhibitor (HDACi), was shown to ameliorate NPC disease phenotypes and considered as a therapeutics candidate but its mechanism of action is not fully clarified yet. Here, we investigated whether SAHA and HNHA, a new derivative of SAHA, reduced cholesterol level and then performed NGS analysis for these two compounds in NPC-iNSCs. Intracellular trafficking, secretion, and vesicular transport factors were highly enriched by two compounds treatment in NPC-iNSCs. Notably, SNAP25, a subunit of SNARE complex was significantly upregulated. Since autophagic flux is impaired in NPC cells due to SNARE complex deficiency, we hypothesized that increasing SNAP25 could contribute to form autolysosomes. When SNAP25 was overexpressed in NPC cells, it compensated deficient Vamp8-SNAP29-STX17 complexes to Vamp8-SNAP25-STX17 complexes then it restores autolysosomes leading to reduce accumulated LC3, LAMP1a, and p62. Consistently, SNAP25 was upregulated by HDAC inhibitors treatment and recovered deficient autophagic flux in vitro, and in vivo. Therefore, upregulation of SNAP25 by HDAC inhibition ameliorated NPC phenotypic and behavioral disorder in 2D culture system and NPC-/- mouse model. These results demonstrated a key role of SNAP25 to ameliorate NPC disease phenotypes and provided HDAC inhibitors as new therapeutic candidates for NPC treatment

Detection of lower levels of SNAP25 using multiple microarray systems and its functional significance in medulloblastoma

Project description:Medulloblastoma (MB) is the most common pediatric malignant brain tumor and patients with a high-risk or recurrent MB respond poorly to current therapies, with higher related mortality. Potential molecules related to MB must be identified to develop targets for therapeutic development. In the present study, we compared MB microarray data obtained using different microarray systems and significant targets were selected by gene annotation and enrichment analysis. Genes for soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) annotated with the function “vesicle” were identified and one of these proteins, synaptosomal-associated protein 25 (SNAP25), had significantly lower expression levels in MB. In addition, SNAP25 was detected in a very low number of MB cells according to western blot analysis and immunohistochemical analyses of archived and formalin-fixed/paraffin-embedded human MB specimens. We found that SNAP25 could change the morphology and the chemotherapeutic effect of arabinofuranosyl cytidine (Ara-C) on SNAP25-expressing MB cells. In conclusion, the expression of SNAP25 is crucial for the dendrite formation and correlate to the targeted chemotherapy. The detection of SNAP25 expression in MB cells may be essential for the chemotherapeutic application of Ara-C.

Trap-80-dependence of TNF-alpha-induced genes

Project description:In fibroblasts, p65-dependent genes can be sub-divided, depending on whether they are Trap-80-dependent or -independent. To examine the generality of this grouping, we performed a microarray analysis of wild-type and Trap-80 knock-down fibroblasts, before and after stimulation of NF-kappaB activity using TNF-alpha. RNA was extracted from three independent cultures of wild-type and Trap-80 knock-down fibroblasts, before and after stimulation for 1 hour with 5ng/ml TNF-alpha. The unstimulated and stimulated wild-type samples, and the stimulated Trap-80 knock-down samples, were used for microarray analysis.
