Project description:We report RNA-sequencing from zebrafish explants generated from embryos injected with mRNA encoding the Nodal ligand ndr2 (N), constitutively active Nodal receptor CA-acvr1b* (T), and uninjected (U) controls. Explants of all 3 conditions were compared at each of 7 developmental stages: sphere (SR), 30% epiboly (30), 50% epiboly (50), shield (SH), 75% epiboly (75), 90% epiboly (90), and 2-somite (2S) (corresponding to 4, 4.7, 5.3, 6, 8, 9, and 11 hours post fertilization, respectively).
Project description:Overexpression of high levels of human UHRF1 tagged with EGFP (hUHRF1-EGFP)high in zebrafish hepatocytes induces cellular senescence by 5 days post fertilization (dpf) and hepatocellular carcinoma by 15-20 dpf. The goal of this study is to compare the transcriptome profile of zebrafish livers with (hUHRF1-EGFP)high overexpression compared to WT siblings. RNAseq was performed on RNA extracted from pools of microdissected liveres by using RiboZero libary prep to capture all transcripts, including transposable elements, that are expressed in the livers.

Project description:Transcriptional profiling of Mtx2 morphants at 30% epiboly stage, shield stage and tailbud stage of embryonic development: Coordinated cell movements of gastrulation reshape the uniform blastula into a structured gastrula with three fundamental axes and an internal germ layer of mesoderm. Genetic regulation of the forces which drive gastrulation is incompletely understood. We employed a microarray screen in zebrafish for genes which participate in epibloy, and identified many with biochemical roles in endocytosis, microtubule remodeling, cell adhesion and transcription.
