Chemo-senolytic therapeutic potential against angiosarcoma

Project description:Angiosarcoma is an aggressive soft-tissue sarcoma with a poor prognosis. Chemotherapy for this cancer typically employs paclitaxel, one of the taxanes (genotoxic drugs), although it has a limited effect due to chemoresistance for prolonged treatment. Here we examine a new angiosarcoma treatment approach that combines chemotherapeutic and senolytic agents. We first find that the chemotherapeutic drugs, cisplatin and paclitaxel, efficiently induce cellular senescence of angiosarcoma cells. Subsequent treatment with a senolytic agent, ABT-263, eliminates senescent cells through the activation of the apoptotic pathway. In addition, expression analysis indicates that senescence-associated secretory phenotype (SASP) genes are activated in senescent angiosarcoma cells and that ABT-263 treatment eliminates senescent cells expressing genes in the type-I interferon (IFN-I) pathway. Moreover, we show that cisplatin treatment alone requires a high dose to remove angiosarcoma cells, whereas a lower dose of cisplatin is sufficient to induce senescence, followed by the elimination of senescent cells by senolytic treatment. This study sheds light on a potential therapeutic strategy against angiosarcoma by combining a relatively low dose of cisplatin with the ABT-263 senolytic agent, which can help ease the deleterious side effects of chemotherapy.

Human melanoma MeWo cell phosphopeptide enriched Orbitrap MS

Project description:Our newly developed high-throughput screening assay of bioactive compounds discovered a cellular phosphatase, calcineurin, was critical in the regulation of Varicella-Zoster Virus (VZV) glycoprotein complex gB/gH-gL mediated cell fusion. To better understand how calcineurin regulates fusion, the potential substrates for calcineurin phosphatase were investigated in human melanoma MeWo cells upon treatment with specific calcineurin phosphatase inhibitor, pimecrolimus. Phosphopeptide enrichment and Orbitrap mass spectrometry analysis identified seven proteins that were exclusively phosphorylated in MeWo cells treated with pimecrolimus compared to DMSO vehicle control, including two known calcineurin substrates. The other five proteins were novel targets for calcineurin dephosphorylation. These data implicated that one or more of these seven proteins in the calcineurin mediated regulation of VZV gB/gH-gL mediated cell fusion.

Gene expression analysis of eight-cell stage rat embryos sired by paternally exposed males to saline or BEP

Project description:8-cell stage rat embryos were sired by males treated with saline or a chemotherapeutic cocktail, BEP, consisting of Bleomycin, Etoposide, and Cisplatin for 9 weeks. Animals were given an additional 9 weeks of recovery, without any treatment, prior to mating. qPCR gene expression profiling for 84 stem cell transcription factors. Total RNA extracted from 25-35 embryos at the eight-cell stage sired by control or BEP-treated males after a 9-wk recovery period (2 females mated per male, n= 4 males/treatment).

Clinical impact of a novel microRNA chemo-sensitivity predictor in gastroesophageal cancer

Project description:Background: miRNAs might be potentially useful biomarkers for prediction of response to chemotherapeutic agents, radiotherapy and survival. The aim of this retrospective study was to validate miRNA response predictors in a cohort of patients with gastroesophageal cancer in order to predict overall survival (OS) and disease-specific survival (DSS). Material and methods: The study population encompassed 53 patients treated with curative intended for loco-regional gastroesophageal cancer. miRNA expression was quantified from pre-therapeutic and diagnostic, formalin-fixed, paraffin embedded tumor specimens using Affymetrix GeneChip miRNA 1.0 Array. Based on growth inhibition of the NCI60 panel in the presence of cisplatin, epirubicine and capecitabine, a miRNA based response predictor was developed. The Cox proportional hazards model was applied to assess the correlations of the response predictor with OS and DSS. Results: A univariate analysis demonstrated a statistical significant improvement of OS for patients who had undergone surgical resection with prediction scores above the median prediction score (HR: 0.41 (95% CI: 0.17-0.96). Adjusting for surgery and stage, this predictor was identified to be independently associated with both OS (HR: 0.37 (95% CI: 0.16-0.87)) and DSS (HR: 0.32 (0.12-0.87)). Conclusion: The miRNA profile predictive for sensitivity to cisplatin, epirubicine and capecitabine was shown to be independently associated with OS and DSS in patients with gastroesophageal cancer.
