Project description:Restoration of hematopoiesis upon bone marrow (BM) transplantation relies on the engraftment, expansion and function of transplanted hematopoietic stem cells (HSC). However, the number of donorâderived HSC typically remain much below that present in normal individuals and this likely limits and delays hematopoietic recovery. HSC depend on supportive stromal niches, however, whether the preâconditioning required for BM transplantation damages this stromal niche has not been evaluated extensively. Using mouse models, we now find that that BM stroma cells (BMSC) are severely and permanently damaged by preâconditioning. Transplantation of primary but not cultured BMSC quantitatively reconstitutes stroma function in vivo, which is mediated by a multipotent CD73+ CD105â Sca1+ BMSC subpopulation. BMSC coâtransplantation significantly ameliorates clinical side effects of BM transplantation and doubles expansion of functional, donorâ derived HSC, demonstrating the potential of stroma transplantation to improve HSC transplantation. Purified CD45âTer119âCD73+ CD105â and CD45âTer119âCD73+ CD105+ cells were directly sorted on lysis buffer with FACS Aria 2 in triplicate.

Project description:Engraftment of donor cells is a prerequisite to successful allogeneic hematopoietic stem cell transplantation. However, it has been poorly characterized in detail both in experimental models and in human. Bone marrow examination by mass cytometry in 26 patients with myeloid malignancies and 20 healthy donors provides evidence that the proportions of myeloid and B-cell progenitors profoundly differed as compared to healthy controls with a shift toward terminal myeloid differentiation and sharp decrease of myeloid and B-cell progenitors. Principal component analysis clearly separated in 2 recipient groups (R1 and R2) driven by previous GVHD (in R2 patients). We then used single cell CITEseq to decipher donor cell engraftment in 10/26 patients (compared with 4 healthy donors). Forty-one clusters were resolved encompassing all bone marrow cellular components. Projection of patients and R1 and R2 showed drastic segregation of the 2 patient groups which were highly significant different repartition in a compositional analysis. In R2 patients sustained engraftment relied on only 10 clusters that included only committed myeloid progenitors, one stem-cell memory T-cell cluster and activated NK-cell clusters. Gene expression hallmarks disclosed repeatedly similar pathway in transplant recipients, namely, TNFα signaling via NFĸ, interferonƔ response, and allograft rejection. Thus, the cellular composition of the engrafted bone marrow in human is strongly influenced by T-cell mediated processes and inflammation leading to an emergency hematopoiesis.

Project description:Adipose tissue engraftment has become a promising strategy for reconstruction in the field of regenerative surgery. However, there are significant challenges such as reabsorption of 50–90% of the transplanted fat or cyst formation due to fat necrosis after fat transplantation. Thus, identifying novel materials or methods to improve the engraftment efficiency is crucial. Here, we investigated the effects of nervonic acid (NA) on adipogenesis and fat transplantation and its underlying mechanisms. We found that NA treatment accelerated adipogenesis through activation of the AKT/mTOR pathway and inhibition of Wnt signaling. Additionally, NA treatment enriched the expression of AKT-mTOR signaling-related genes and increased the expression of genes involved in angiogenesis and fat differentiation. Furthermore, NA effectively improved the outcome of adipose tissue engraftment. Treatment of NA reduced the resorption of transplanted fat and increased the proportion of perilipin-1+ adipocytes with a lower portion of vacuoles. Moreover, the NA-treated group showed decreased pro-inflammatory response and had more CD31+ vessel structures, which were relatively evenly distributed among viable adipocytes, facilitating the successful engraftment. In summary, we demonstrated that NA not only stimulates adipogenesis by regulating in human MSCs but improve the outcome of fat transplantation by reducing inflammation and stimulating angiogenesis. We suggest that NA could be an adjuvant strategy to support the fat transfer in regenerative surgery.

Project description:We found that transient inhibition of JNK pathway increased short term-HSC frequency in cord blood CD34+ cells by 12.56 folds. Transcriptome analysis shows that inhibition of JNK pathway upregulated HSC-specific and anti-oxidative gene expression, prevented upregulation of cell cycle entry, oxidative phosphorylation and glycolysis related gene expression, and downregulated reactive oxygen species (ROS) active gene expression. Consistently, cell cycle and metabolic analysis show that inhibition of JNK pathway prevented HSC from cell cycle entry, glucose uptake increase and ROS accumulation. Accordingly, transient inhibition of JNK pathway also enhanced long term-HSC recovery during cord blood CD34+ cell collection. Collectively, these findings suggest that transient inhibition of JNK pathway could promote quiescent state of HSCs by preventing cell cycle entry and metabolic activation, thus improving HSC recovery and engraftment ability.

Project description:Through variation in the cancer cell and their immune infiltrates each tumor represents a unique problem, but therapeutic targets can be found in the shared features. Radiation therapy can change the interaction between the cancer cells and the stroma through release of innate adjuvants including the STING agonist cGAMP. Enpp1 is a phosphodiesterase that can be expressed by cancer cells and can degrade cGAMP. Enpp1 can therefore limit innate adjuvant availability following radiation therapy. We observed that many cancer cells lack Enpp1 expression, but that Enpp1 expression is retained in cells of the tumor stroma and this expression in the tumor stroma limits tumor control by radiation therapy. We demonstrate the efficacy of a novel Enpp1 inhibitor and show that this inhibitor improves tumor control by radiation even where the cancer cells lack Enpp1. We show that the mechanism requires STING and type I IFN receptor expression by non-cancer cells, and is dependent on CD8 T cells as a final effector mechanism of tumor control. This suggests that Enpp1 inhibition may be an effective partner for radiation therapy regardless of whether cancer cells express Enpp1, and identifies a novel therapeutic Enpp1 inhibitor suitable for combination therapies in immuno-oncology.

Project description:Adoptive cell therapies for cancer treatment often fail due to a lack of T cell survival and persistence within the patient. Redirecting glycolytic processing with dichloroacetate (DCA) during in vitro expansion improves immediate and long-term T cell engraftment by up to 8x - ultimately resulting in a drastic improvement in tumor clearance. Here we use bulk RNA sequencing to interrogate the changes in transcriptional programs resultant of DCA treatment that may confer the observed improvement in engraftment and tumor clearance of DCA-treated T cells.
