Expression profile of (inducible) homozygous Apc loss in the mouse intestinal epithelium

ABSTRACT: To asses the effect of Apc loss on the intestinal epithelium we induced homozygous VillinCreERT2 Apc flox/flox mice with tamoxifen on 3 consecutive days (day 0, 1 and 2), and harvested small intestinal epithelium on day 3.

Transcriptome analysis of Brg1 deficient small intestinal epithelium in the context of normal and aberrant Wnt signalling

Project description:Brg1 has been reported to act as a trans-activator for the Wnt pathway by interacting with beta-catenin. Given this interaction and the crucial role Wnt signalling plays in the intestinal homeostasis, we aimed to investigate the effect of Brg1 loss on gene expression in normal and Wnt activated small intestinal epithelium. We used VillinCreERT2 Cre recombinase and loxP targeted allels of Brg1 and Apc to generate 4 cohorts of conditional knock-out mice: Cre-negative controls (n=4), Brg1 deficient (n=4), Apc deficient (n=3) and double Brg1-Apc deficient (n=4). All mice were induced by 4x80mg/kg daily injections of Tamoxifen. Epithelium enriched (gut scrapes) samples of small intestine (jejunum) were collected at day 4 post induction. Loss of Brg1 expression in the small intestinal epithelium at this time point was confirmed by immunohistochemistry.

Non-tumor/tumor intestinal tissue of control or intestine-specific HAI-1 deficient Apc(Min/+) mice

Project description:To analyse roles of HAI-1/Spint1 in intestinal tumorigenesis, we examined the effect of intestine-specific deletion of Spint1 gene on Apc(Min/+) mice. The loss of Hai-1/Spint1 significantly accelerated tumor formation in ApcMin/+ mice and shortened their survival periods. Mouse small intestine tumor tissue or background mucosa lacking macroscopically visible tumors were proceeded to RNA extraction and hybridization on microarrays (Affymetrix Mouse Genome 430 2.0 Array). Non-tumor or tumor intestinal mucosa tissues of Apc (Min/+)/Spint1 (flox/flox) mice and non-tumor or tumor intestinal mucosa tissues of Apc (Min/+)/Spint1 (flox/flox)/Vil-Cre mice were analysed. The experiment was repeated respectively.

Gene expression changes in Apc-mutant mouse intestinal organoids with and without deleting the Prox1 transcription factor

Project description:We isolated and selected intestinal adenoma organoids from villin-CreER; Apcflox/flox and villin-CreER; Apcflox/flox; Prox1flox/flox mice and added tamoxifen to induce the deletion of the Apc and Prox1 genes in the intestinal epitheliul ex vivo. Microarray experiments were carried out 7 days after the addition of tamoxifen. Total RNA obtained from villin-CreER; Apcflox/flox and villin-CreER; Apcflox/flox; Prox1flox/flox organoids were compared 7 days after the addition of tamoxifen and 5 days after the selection for Apc-mutant organoids in the absence of the Wnt-agonist R-Spondin1.

Total RNA obtained from Lgr5-EGFP-IRES-CreER; Apcflox/flox organoids were compared 7 days and 20 days after the addition of tamoxifen, cultured without the Wnt-agonist R-Spondin1.

Cold atmospheric plasma affects stem cell renewal and differentiation and is associated with a regeneration process in an intestinal organoid culture model

Project description:Cold atmospheric plasma (CAP) has recently been proposed as an anti-cancer therapy, mainly driven by the concentration of reactive oxygen and nitrogen species (RONS), whereas several studies suggest its selectivity on cancer cells. However, the underlying mechanisms of this “selectivity” are largely poorly understood and rely exclusively on 2D (dimensional) unicellular monolayers, which do not recapitulate the in vivo epithelial context. In this study, we aimed to evaluate phenotypically and transcriptomically the effect of different modalities of CAP’s application (direct, indirect and plasma activated medium) on intestinal organoids derived from wild type (WT), VillinCreERT2/Apc floxflox and VillinCreERT2/Apc WT adult mice. The phenotype analysis of different modalities (direct, indirect, plasma activated medium) of CAP’s application revealed that direct and indirect CAP treatment decrease WT intestinal organoid survival associated to a predominance of spheroids organoids, this reduction in survival was not observed for organoids derived from Villin CreERT2/Apc flox flox mice. Moreover, transcriptome analyses of control’s organoids showed that indirect and plasma activated applications induced a transcriptomic reprogramming, highlighted by a decrease of intestinal stem cell pool, cell cycle arrest, cell death, altogether associated to a regeneration repair process. These results have been validated at the protein level. In conclusion, in a 3D organoid model, the selectivity of CAP on cancer cells does not seem to be confirmed.

Msi1 integrates APC loss and mTORC1 activation to promote intestinal stem cell transformation

Project description:Loss of the APC tumor suppressor in the intestinal epithelium initiates the majority of human colorectal adenocarcinomas. Constitutive Î²-catenin activation is thought to underlie tumorigenesis induced by loss of APC, however Î²-catenin activation alone does not recapitulate all APC-loss phenotypes, suggesting that additional pathways are required. We demonstrate that aberrant activation of the Msi1 RNA binding protein occurs upon APC loss and that constitutive Msi1 activation alone is sufficient to phenocopy APC loss in the intestinal epithelium. Msi1 elicits these effects through binding of mRNAs encoding pleiotropic tumor suppressors resulting in promiscuous activation of quiescent intestinal stem cells, proliferative expansion of the stem cell compartment, crypt fission, and blocked differentiation. Further, we find these phenotypes to be largely dependent on mTORC1 activity, and demonstrate that loss of Msi activity is sufficient to abrogate tumorigenesis in mouse and human systems. Our findings implicate Msi1 as a central coordinator of APC loss-induced intestinal stem cell transformation and adenocarcinoma progression. 2 wild-type, 2 transgenic samples
