RNA-seq sequencing of the transgenic plants seeds

Project description:To analyze influence of ZmDWF4 on seed development, we we chose 10DAP and 15DAP from the transgenic line L100 seed to RNA-seq sequence. Total RNA was extracted from the seeds (10 days after pollination, 10DAP, 15 days after of pollination,15DAP) of transgenic line L100 and wild type Q319. cDNA library was builded in the BIOMARKER, original reads were sequenced based on Illumina HiSeq2000, and through rRNA, low quality fragment DNA of filtering, ultimately, we obtained 99.30Gb high quality clean reads. Compared with reference gene sequence(AGPv3), according to genes function annotation, we found 463, 675 different expression genes, respectively.We classified the different expression genes according to Cellular Component, Molecular Function and Biological Process. It showed that BRs could affect many genes expression involved in different biological process in the seed development.

Transcriptional profiling during pollination in Ophrys fusca, through cDNA microarrays

Project description:Survey of post pollination events in a sexually deceptive orchid (Ophrys fusca): a transcriptional approach Pollination through deception is a widespread phenomenon in angiosperm, and is extremely common in Orchidaceae family. One of the most striking pollination mechanism in orchids is known as sexual deception, in which flowers lure pollinators by foraging chemical (sex pheromones), visual (e.g. labellum colour and/or shape) and tactile (e.g. labellum pilosity) cues of the female insect pollinator. Ophrys has been used as a model genus to study sexual deception mechanism, mainly regarding chemical analysis in plant-insect association. Study was focused on Ophrys fusca, a species widely distributed in Mediterranean Basin. The main objective rely on Ophrys fusca gene expression study after pollination, through a transcriptional approach using cDNA microarrays. In order to evaluate pollination enhanced events, two different time points were selected: 2 days and 4 days after pollination. Ophrys fusca plants were sampled from a Portuguese natural occurring population. Plants were covered with a white and inert net, built specially for preventing pollinator’s visits in both pollinated and unpollinated flowers. Cross- pollination was performed manually with a sterile plastic stick. Five biological replicates (5 plants in each replicate) from each condition (pollinated and unpollinated) were collected in each time-point Flowers that demonstrate strict pollination regulation, as orchids, provide an excellent model system to unravel pollination- elicited mechanisms (i.e. petal senescence, pigmentation changes, ovary growth). Therefore, this study aims to contribute to the overall knowledge on orchid pollination biology, which is still lacking.

Transmission of xanthomonads to Medicago truncatula seeds

Project description:Plant pathogenic bacteria disseminate and survive through transmission to and by seeds of hosts and non-hosts plants. To investigate the interaction between xanthomonads and developing seeds of Medicago truncatula, plants at the M-oM-,M-^Bower bud stage were spray inoculated until runoff with xanthomonads suspensions. Using the Medicago NimbleGen chip, a transcriptomic analysis was performed on seeds to characterize the molecular dialogue between Xanthomonas campestris pv. campestris in an incompatible situation with M. truncatula seeds and Xanthomonas alfalfae pv. alfalfae in a compatible situation at two developmental time points (16 and 32 days atfter pollination (DAP). Six-condition experiment, 16dap_Mock versus 16dap_Xaa, 16dap_Mock versus 16dap_Xcc, 32dap_Mock versus 32dap_Xaa, 32dap_Mock versus 32dap_Xcc. Biological replicates: 6 controls (16dap_Mock, 32dap_Mock), 12 treatments (16dap_Xaa, 16dap_Xcc, 32dap_Xaa, 32dap_Xcc), independently grown and harvested. One replicate per array.

Transcription profiling of Medicago truncatula radicles incubated in polyethylene glycol solution and embryos at different time points after pollination to study dessication tolerance

Project description:Time course of the transcriptome of desiccation-sensitive 2.7-2.9 mm-long radicles of Medicago truncatula seeds at different time points during incubation in a polyethylene glycol (PEG) solution at -1.7 MPa and 10°C, resulting in a gradual re-establishment of desiccation tolerance. Gene profiling was also performed on embryos before (14 days after pollination) and after acquisition of desiccation tolerance during maturation (20 days after pollination).
