Next Generation Sequencing Facilitates Quantitative Analysis of Wild Type and Stat3f/f K5cre Skin Transcriptomes

ABSTRACT: Atopic dermatitis (AD) is a common chronic skin disease, affecting >15% of children and 2-10% of adults worldwide. Understanding pathogenic mechanisms of the onset and dynamic progression of AD is required to design its effective treatments. The kinetical analysis of transcriptome in a mouse AD model revealed the pathways that led a vulnerable background and were critical to the onset of disease progress.

Project description:Atopic dermatitis (AD) is a common pruritic dermatitis with macroscopically nonlesional skin that is often abnormal. Therefore, we used high-density oligonucleotide arrays to identify cutaneous gene transcription changes associated with early AD inflammation as potential disease control targets. Skin biopsy specimens analyzed included normal skin from five healthy nonatopic adults and both minimally lesional skin and nearby or contralateral nonlesional skin from six adult AD patients. Early-onset pediatric atopic dermatitis is characterized by Th2/Th17/Th22- centered inflammation and lipid alterations

Project description:Background: While atopic dermatitis (AD) often starts in early childhood, detailed tissue profiling of early-onset AD in children is lacking, hindering therapeutic development for this patient population with a particularly high unmet need of better treatments. Objective: We sought to globally profile the skin of infants with AD compared to adults with AD and healthy controls. Methods: We performed microarray, RT-PCR and fluorescence microscopy studies in infants and young children (<5yo) with early-onset AD (<6mo) compared to age-matched controls and adults with longstanding AD. Results: Transcriptomic analyses revealed profound differences between early-onset pediatric vs. longstanding adult AD, not only in lesional but also non-lesional tissues. While both patient populations harbored Th2-centered inflammation, pediatric AD also showed significant Th17-skewing, but lacked the Th1 upregulation that characterizes adult AD. Defects in lipid barrier (e.g. ELOVL3, DGAT2) and tight junction regulation (e.g. Claudins 8 and 23) were evident in both groups but, unlike adult AD which showed the classic downregulation of epidermal differentiation and cornification products, pediatric AD exhibited relatively normal expression of these genes. Some lipid-associated mediators (such as FAR2 and FA2H) even showed preferential downregulation in pediatric AD, and lipid barrier genes (FA2H, DGAT2) showed inverse correlations with transepidermal water loss/TEWL, a functional measure of the epidermal barrier. Conclusions: Children and adult AD skin samples share lipid metabolism and tight junction alterations, but epidermal differentiation complex defects are only present in adult AD, potentially resulting from chronic immune aberration that is not yet present in early-onset disease.

Molecular Networks in Atopic Mothers Impact the Atopy Risk of Infants

Project description:The prevalence of atopic diseases has increased with atopic dermatitis (AD) as the earliest manifestation. We assessed if molecular risk factors in atopic mothers influence their offsprings’ susceptibility to an atopic disease. Pairs of pregnant women and their infants with or without parental atopy were followed over the first 2 years of life. Global DNA methylation and differentially methylated regions (DMR) were determined in atopic and non-atopic mothers. During the first 2 years of life, AD was more prevalent in children of atopic compared to non-atopic mothers with an increase in food sensitization in children with AD. 165 DMRs distinguished atopic from non-atopic mothers. Maternal atopy combined with DMRs increased the offsprings’ predicted risk to develop AD from an odds ratio of 2.56 to 4.26.

Project description:Elderly atopic dermatitis is a subtype of AD defined by age (over 60 years), which has different clinical manifestations and distinct inflammatory patterns from AD in infants, children, and adolescents/adults. However, the molecular characteristics of elderly AD remain to be clarified.We sought to characterize the molecular features of skin lesions and peripheral blood mononuclear cells (PBMCs) in patients with AD across various age groups, focusing on elderly AD.We identified the molecular phenotypes of skin lesions and PBMCs in elderly individuals with AD. Fibroblasts, innate immune cells, and SASP might play important roles in the pathogenesis of elderly AD.
