Comparative physiological and proteomic signatures reveal contrasting stay green and senescence phenotypes in drought tolerant and susceptible pearl millet and wheat genotypes

Project description:The marker- and genome-based development of drought resistant and high yield cereal crops is the most pressing activity in a constantly more stress- and harmful environment for plant productivity. Genome-based assisted breeding is only capable to cover 30-40 % of phenotypic variance according to the most recent GWAS studies. There are many processes which are not predictable by genome information, especially protein translation and activity which is crucial for phenotypic responses and survival of the plants under severe stresses. Therefore, we present comparative proteomic and physiological analysis under drought stress in two of the most important staple food crops pearl millet and wheat thereby representing C4 and C3 plants. Here, we have selected contrasting genotypes, and performed a large-scale comparative analysis from the molecular to the phenotypic level under drought stress. We were able to establish molecular-physiological phenotypes for: 1. Stay green protein signature in contrasting pearl millet genotypes which is highly correlated to the physiological data, in the submitted manuscript. 2. No clear indication of stay green proteome signatures in contrasting wheat genotypes but instead differential senescence proteome signatures not capable to cope with similar drought stress. These mechanisms are decisive for drought resistance and yield/grain filling under stress conditions and for the first time these physiological phenotypes (seed yield, root growth, and photosynthesis) are directly linked to the molecular proteomic phenotype. We think these results are of broad importance for the scientific community. This study demonstrates the enormous molecular and phenotypic plasticity because the selected genotypes represent the extreme points of stress adaptation and yield protection.

Unravelling the molecular mechanism underlying drought stress response in chickpea via integrated multi-omics analysis

Project description:This study was aimed to deal with a comparative proteome analysis of the two chickpea genotypes with contrasting response to drought stress, ICC 4958 (drought-tolerant, DT) and ICC 1882 (drought-sensitive, DS). Proteins were extracted from the root tissues collected from the control and drought stressed plants of both the genotypes. NanoLC-MS/MS analysis of the protein sample was performed using EASY-nLC 1000 system for the separation and identification of peptides/proteins. This study provided a mechanistic insight of drought stress tolerance in chickpea.

Project description:Background Maintaining global food security in the context of climate changes is an important challenge in the next century. Improving abiotic stress tolerance of major crops, like wheat, can contribute to this goal. Therefore, new genes improving tolerance to abiotic stresses, like drought, are needed to support breeding programs aimed at producing more adapted cultivars. Recently, we screened five wheat Zinc Finger Proteins (TaZFPs) and identified TaZFP13D as a new gene improving water-stress tolerance. However, a more detailed characterization of this gene is required to better evaluate its potential in drought tolerance and to decipher the underlying molecular basis. Results We used the Barley Stripe Mosaic Virus to up- or down-regulate TaZFP13D expression in wheat. Overexpression of TaZFP13D under well-watered conditions enhances growth as indicated by improved biomass. Exposing plants to a severe drought stress revealed that TaZFP13D strongly increases survival rate and stress recovery. In addition, TaZFP13D reduces drought-induced oxidative damages, at least in part by improving key antioxidant enzymes activity. Conversely, down-regulation of TaZFP13D decreases drought tolerance and protection against drought-induced oxidative damages. RNA-seq-based transcriptome analysis showed that many genes regulated by TaZFP13D were previously shown to improve drought tolerance, while many others are related to the photosynthetic electron transfer chain and are proposed to improve photosynthesis efficiency and chloroplast protection against ROS damages. Conclusion This study highlights the important role of TaZFP13D in wheat drought tolerance, contributes to unravel the complex regulation governed by TaZFPs, and provides a useful marker to select more drought tolerant wheat cultivars.

Project description:The aim of the study was to identify candidate genes responsible for drought tolerance trait between a pair of wheat varieties ( WL711 and C306) and correspondng progeny bulks (10 drought susceptible RILs and 10 drought tolerant RILs) by combining QTLs analysis with expression analysis. Microarray analysis of RNA extracted from the flag leaves showed large number of differentially expressed genes. The number of differentially expressed genes was reduced to 37 on the basis of their occurance in a major QTL region (responcible for drought tolerance) detected in RIL population derived from WL711 and C306. Transcript profiling of drought responsiveness in two contrasting rice genotypes during vegetative stage stress

Project description:Rice (Oryza sativa), the major staple food crop is being cultivated under varying ecosystems ranging from irrigated lowland to rainfed upland environments. Improvement in the rice production under drought prone unfavourable environment depends on the development of drought tolerant genotypes which needs thorough understanding of physiological and molecular events behind the tolerance mechanism. There is considerable genetic variation for drought tolerance mechanism within the cultivated gene pool. To understand the diversity of drought response, two indica rice genotypes namely, i) Apo, an up-land drought tolerant indica veriety from Philippines and ii) IR64, a popular high yielding drought susceptible genotype were selected for this study. We used the 22K rice Oligoarray from Agilent technologies to study the transcript profile in the leaves of the two contrasting rice genotypes under control and drought stressed conditions during vegetative phase. Keywords: Drought response We used Agilent rice gene chips (G4138A) to investigate the transcript level changes in rice leaf tissues during drought stress. We used two contrasting rice genotypes (IR64 drought susceptible and Apo drought tolerant) differing in their degree of drought tolerance. Plants were grown under green house conditions and drought stress was imposed on 33rd DAS. Leaf sampling was done in both control and drought stressed plants after 6 days of drought stress. Three replications of microarray experiments were carried out by hybridizing the control samples against the drought stressed samples.
