MiroRNA expression from pig endometrium exposed to developing pre-implantation embryo versus unfertilized oocytes in vivo

ABSTRACT: In vivo evidence suggests a dynamic change of microRNA expression with the phase of the endometrial cycle to achieve endometrial receptivity. In vivo animal studies outlined post – implantation changes in microRNA profiles. Little information is available on the role of microRNAs in the early stages of the embryonic – endometrial dialogue prior to established implantation .Our aim is to investigate the possible role of microRNAs the early endometrial response to the developing pre- attachment blastocyst prior to established implantation

HB-EGF-loaded nanovesicles enhance trophectodermal spheroid attachment and invasion

Project description:The ability of trophectodermal cells (outer layer of the embryo) to attach to the endometrial cells and subsequently invade the underlying matrix are critical stages of embryo implantation during successful pregnancy establishment. Extracellular vesicles (EVs) have been implicated in embryo-maternal crosstalk, that reprogram endometrial cells towards a pro-implantation signature and phenotype. However, challenges associated with EV yield and direct loading of biomolecules limits their therapeutic potential. We have previously established generation of cell-derived nanovesicles (NVs) from human trophectodermal cells (hTSCs) and their capacity to reprogram endometrial cells to enhance adhesion and blastocyst outgrowth. Here, we developed a rapid NV loading strategy to encapsulate potent implantation molecules such as HB-EGF (NVHBEGF). We show these loaded NVs upon uptake to low-receptive endometrial HEC1A cells elicit EGFR-downstream pathways and enhance target cell attachment and invasion in an EGF-signalling dependent manner. This phosphoproteomics and proteomics approach reveals that NVHBEGF regulate short-term signalling (e.g., ERBB/EGFR signalling) and long-term reprogramming capabilities on endometrial cells, and demonstrate their functional regulation on trophectodermal-endometrial interactions and trophectodermal invasion. This proof-of-concept study demonstrate feasibility in enhancing the potency of NVs in the context of embryo attachment and establishment.

Endometrial luminal epithelium transcriptome during early diestrus defines pregnancy outcome in cattle

Project description:Prior to implantation, embryonic development occurs in the confinement of the reproductive tract lumen. The endometrial epithelia determines the uterine luminal composition and, consequently, the molecular milieu available to the pre-implantation embryo. This cow RNA-seq study was especially designed to (i) evaluate the impact that progesterone concentration has on the transcriptome of luminal epithelial cells, and (ii) evaluate the impact that endometrial gene expression has on pregnancy outcome. Luminal epithelial cells were collected using a cytology brush three days prior to embryo transfer, so that the molecular evaluation and the pregnancy outcome, measured 30 days after estrus, pertained to the same reproductive cycle.

Proteomics of U1810 cells upon treatment with microRNAs with an AAGUGC seed motif.

Project description:microRNA dysregulation is a common feature of cancer cells, but the complex roles of microRNAs in cancer are not fully elucidated. Here we used functional genomics to identify oncogenic microRNAs in non-small cell lung cancer and to evaluate their impact on response to EGFR targeting therapy. Our data demonstrate that microRNAs with an AAGUGC-motif in their seed-sequence increase both cancer cell proliferation and sensitivity to EGFR inhibitors. Global transcriptomics, proteomics and target prediction resulted in the identification of several tumor suppressors involved in the G1/S transition as targets of AAGUGC-microRNAs. The clinical implications of our findings were evaluated by analysis of public domain data supporting the link between this microRNA seed-family, their tumor suppressor targets and cancer cell proliferation. In conclusion we propose that AAGUGC-microRNAs are an integral part of an oncogenic signaling network, and that these findings have potential therapeutic implications, especially in selecting patients for EGFR-targeting therapy.

Human trophectoderm extracellular vesicle proteome

Project description:Embryo implantation into maternal endometrium is critical for initiation and establishment of pregnancy, requiring developmental synchrony between endometrium and blastocyst. However, factors regulating human endometrial-embryo cross talk and facilitate implantation remain largely unknown. Extracellular vesicles (EVs) are emerging as important mediators of this process. Here, human trophectomderm stem cell-derived EVs were shown to transfer to and regulate human endometrial cells towards processes associated with implantation. Importantly, transfer of trophectoderm EV cargo proteins to endometrial cells to mediate changes in polarity is demonstrated.

microRNAs in early pregnant porcine endometrium revealed by Illumina sequencing

Project description:Preimplantational estrogen exposure to pregnant gilts has been associated with implantation failure, embryonic losses and changes in endometrial mRNA expression. Small non-coding RNAs (ncRNA), especially miRNAs, play a key role in regulation of gene expression. Effects of estrogens on endometrial microRNAs (miRNA), however, have not been investigated in this context so far. Thus, we studied the influence of estradiol-17β (E2) on the endometrial expression profile of miRNAs in the pig at gestational day 10. E2 administered early during pregnancy resulted in significantly higher endometrial estrogen concentrations even at a low dose, but did not disrupt the expression profile of endometrial miRNAs.

Endometrial extracellular vesicle decidualization

Project description:Proper decidualization is vital in preparation for a potential embryo receptivity, placentation, menstrual health and subsequent endometrial regeneration. Given the importance of extracellular vesicles (EVs) in intercellular communication, and recently in embryo implantation and indicators of menstrual cycle and fertility, we investigated their role during decidualization. Overall, this study provides an insight into distinct variation in sEV composition depending upon the level of decidualization of endometrial stromal cells, with the signaling potential to coordinate endometrial health ranging from embryo implantation, facilitating placentation and subsequent endometrial regeneration.

Enhancing Uterine Receptivity for Embryo Implantation through Controlled Collagenase Intervention

Project description:Ineffective endometrial matrix remodeling, a key factor in infertility, impedes embryo implantation in the uterine wall. Our study reveals the cellular and molecular impact of human collagenase-1 administration in mouse uteri, demonstrating enhanced embryo implantation rates. Collagenase-1 promotes remodeling of the endometrial extracellular matrix (ECM), degrading collagen fibers and proteoglycans. This process releases matrix-bound bioactive factors, (e.g. VEGF, decorin), facilitating vascular permeability and angiogenesis. Collagenase-1 elevates embryo implantation regulators, including NK cell infiltration and the key cytokine LIF. Remarkably, uterine tissue maintains structural integrity despite reduced endometrial collagen fiber tension. In-utero collagenase-1 application rescues implantation in the heat stress and embryo transfer models, known for low implantation rates. Importantly, ex-vivo exposure of human uterine tissue to collagenase-1 induces collagen de-tensioning and VEGF release, mirroring remodeling observed in mice. Our research highlights collagenase potential to induce and orchestrate cellular and molecular processes enhancing uterine receptivity for effective embryo implantation. Here in this study extracellular vesicles from trophoblast cells were shown to modulate endometrial epithelial cell secretome in favour of embryo implantation and embryo development.

The analysis of the α2,6-sialylated proteins in endometrium from porcine gestational day 15 by LC-MS/MS

Project description:Implantation is the attachment of embryo in the endometrium. Failure in implantation is a major cause of early pregnancy loss. During implantation, the temporal uterine lumen closure can help embryo attach to the uterus. In pigs, extending of endometrial folds to form interlocking finger-like projections is a main cause leads to uterine lumen closure during attachment time, but the underlying mechanisms are largely unknown. Our data reveal that pig uterine luminal epithelium (LE) migrate in coordinated groups during extending of endometrial folds. Moreover, the MALDI-TOF MS based N-glycomic characterization of porcine endometrium revealed α2,6-linked sialic acid are highly expressed in pig uterine LE during extending of endometrial folds. To investigated the mechanisms by which α2,6-sialylated proteins in formation of the endometrial folding during implantation in pigs, the α2,6-sialylated proteins in pig uterine LE were characterized by proteomic analysis and those proteins that are involved in cell adhesion, such as E-cadherin, were detected. Finally, our in vivo and in vitro data show that α2,6-sialylation of E-cadherin occurs in accompany with collective epithelial migration. The results provide new insight into the mechanism of pig implantation by identifying that α2,6-sialylation of cell adhesion molecules may participate in formation of extending of endometrial folds through promoting of collective migration of uterine LE.
