Project description:Glyphosate-based herbicides are the major pesticides used worldwide. There is an intense debate on the estrogenic effects of their ingredients. We have compared the estrogenic effects of glyphosate (the active principle), polyethoxylated tallowamine (a co-formulant), and a commercial formulations containing different co-formulants to those of estradiol and bisphenol A in the MCF-7 human breast cancer cell line. The gene expression profiles were determined using the Affymetrix Human Transcriptome 2.0 Array.

Project description:Exposures to environmental endocrine disruptors is growing and human contact in industrialized countries has become signficant and constant. We studied the effect of chronic exposure to two endocrine disrupting compound, bisphenol A and genistein, in an in vitro cell culture system. MCF7 cells were cultured for greater than 70 passages under normal (MCF7-F) conditions or with the addition of 50 nM BPA (MCF7-B) or GEN (MCF7-G). We performed transcriptome analysis of the all three cell lines in the absence of any estrogenic compounds and in the presence of 10 nM estradiol for 3 hours. The MCF7-F, B, G cell lines were starved of all estrogenic compounds by culturing the cells for 72 hours in phenol red free DMEM containing 5% charcoal/dextran treated FBS. Cells were then treated with either ethanol vehicle or 10 nM 17b-estradiol for 3 hours. Total RNA was harvested and utilized for whole genome analysis on the Affymetrix Human Trascriptome Array 2.0. The experiment was performed in duplicate

Project description:Plasticizers with estrogenic activity, such as bisphenol A (BPA), have been reported to have potential adverse health effects in humans, especially in fetal and infant stages. Due to mounting evidence and public pressure BPA is being phased out by the plastics manufacturing industry and is being replaced by other bisphenol variants in “BPA-free” products. We have compared estrogenic activity of 7 bisphenol analogues (BPA; bisphenol S, BPS; bisphenol F, BPF; bisphenol AP, BPAP; bisphenol AF, BPAF; bisphenol Z, BPZ; bisphenol B, BPB) in human breast cancer cell lines. Identification of ERα AF-1 or AF-2 mediated genes in mouse uterus

Project description:Estrogen receptor alpha (ERα) is a ligand dependent transcription regulator, which contains two transactivation functional domains, AF-1 and AF-2. These activities are regulated differently by the ligands. Specifically, the selective estrogen receptor modulators (SERMs) regulate AF-1 rather than AF-2. It is important to know whether AF-1/AF-2 predominantly regulated genes exist in the tissues for a better understanding of the SERMs functionality. We sought out AF-1 dependent estrogenic genes by using the AF-2 mutated knock-in (KI) mouse model, AF2ERKI. AF2ER is an estrogen-insensitive AF-2 disrupted ERα mutant mouse but unique to this model, AF-1 can be activated by the estrogen-antagonist, fulvestrant (ICI), in vitro and in vivo. The information of ICI-mediated gene expression profile of AF2ERKI could define whether AF-1/AF-2 predominantly regulated genes exist in the tissues.
