Project description:The response of tumors to PDT treatment is expected to provide information on effects of oxygen depletion, induced apoptosis, induction of an inflammatory response and induction of an ani-tumor immune response. Experiment Overall Design: NCI-H69 human SCLC xenografts were induced in SCID mice. Three pairs of xenograft tumors, PDT treated and untreated controls, were harvested four hours after PDT treatment. RNA was isolated and prepared for hybridization to human Affymetrix GeneChip arrays.

Transcriptional analysis of time course in response to HY-PDT on human nasopharyngeal carcinoma cell line CNE-2

Project description:Analysis of photodynamic therapy with hypericin of nasopharyngeal carcinoma cells CNE-2 in different time-points at gene expression level. The hypothesis tested in the present study was that HY-PDT could induce apoptosis on CNE-2 cells via intrinsic pathways. Results provide important information of the response of apoptosis, such as specific oxidative stress genes, mitochondrial related genes, cell cycle related genes, etc. Total RNAs were isolated from the cultured cells treatment with 20 μg/ml HY and irradiation then cultured in dark for 0, 2, 6, 12, 20 h respectively compared to cells treatment with 20 μg/ml HY only then cultured in dark for 20 h.

Targeted PDT agent eradicates TrkC expressing tumors via photodynamic therapy (PDT).

Project description:This contribution features a small molecule that binds TrkC (tropomyosin receptor kinase C) receptor that tends to be overexpressed in metastatic breast cancer cells but not in other breast cancer cells. A sensitizer for (1)O2 production conjugated to this structure gives 1-PDT for photodynamic therapy. Isomeric 2-PDT does not bind TrkC and was used as a control throughout; similarly, TrkC- cancer cells were used to calibrate enhanced killing of TrkC+ cells. Ex vivo, 1- and 2-PDT where only cytotoxic when illuminated, and 1-PDT, gave higher cell death for TrkC+ breast cancer cells. A 1 h administration-to-illumination delay gave optimal TrkC+/TrkC--photocytotoxicity, and distribution studies showed the same delay was appropriate in vivo. In Balb/c mice, a maximum tolerated dose of 20 mg/kg was determined for 1-PDT. 1- and 2-PDT (single, 2 or 10 mg/kg doses and one illumination, throughout) had similar effects on implanted TrkC- tumors, and like those of 2-PDT on TrkC+ tumors. In contrast, 1-PDT caused dramatic TrkC+ tumor volume reduction (96% from initial) relative to the TrkC- tumors or 2-PDT in TrkC+ models. Moreover, 71% of the mice treated with 10 mg/kg 1-PDT (n = 7) showed full tumor remission and survived until 90 days with no metastasis to key organs.

Low Dose PDT - Murine Tumors

Project description:Tumors were induced in C57BL/6 mice by subcutaneous injection of murine prostate carcinoma cell line TRAMP-C2. Tumor-bearing mice were narcotized, injected with photosensitizer 5-aminolaevulinic acid (5-ALA), shaven at their back, and 3-4 h later the whole tumor area was irradiated perpendicular (under anesthesia) through the intact skin using a 635 nm laser (75 or 100 J/cmM-2 at 200 mW/cmM-2). After treatment the mice were kept in the dark, then the tumor was excised and total RNA was extracted for expression profiling.
