Project description:Human tumour cell lines PC3, DU145, U87 and U373 (prostate carcinoma and glioblastoma) were treated with photosensitizers 5-aminolaevulinic acid (5-ALA) or photofrin. Then, the cells were irradiated sublethally with 635 nm laser light.<br>After photodynamic therapy, the cells were grown at 37°C for 4 or 24 hours in the dark until extraction of total RNA and expression profiling.

Low dose PDT - Murine Cells

Project description:Murine prostate carcinoma cell lines TRAMP-C1 and TRAMP-C2 were grown for 16 h at 37M-0C with photosensitizer 5-aminolaevulinic acid (5-ALA; 50 M-5g/ml). Then, the cells were sublethally irradiated with laser light (635 nm) at room temperature (using RPMI1640 without phenol red as medium). After photodynamic therapy, the cells were cultivated at 37M-0C for 4 h and 24 h, respectively. After that time total RNA was extracted for expression profiling.

Low Dose PDT - Murine Tumors

Project description:Tumors were induced in C57BL/6 mice by subcutaneous injection of murine prostate carcinoma cell line TRAMP-C2. Tumor-bearing mice were narcotized, injected with photosensitizer 5-aminolaevulinic acid (5-ALA), shaven at their back, and 3-4 h later the whole tumor area was irradiated perpendicular (under anesthesia) through the intact skin using a 635 nm laser (75 or 100 J/cmM-2 at 200 mW/cmM-2). After treatment the mice were kept in the dark, then the tumor was excised and total RNA was extracted for expression profiling.

Transcription profiling of human H69 xenograft tumors in mice following PDT treatment.

Project description:The response of tumors to PDT treatment is expected to provide information on effects of oxygen depletion, induced apoptosis, induction of an inflammatory response and induction of an ani-tumor immune response. Experiment Overall Design: NCI-H69 human SCLC xenografts were induced in SCID mice. Three pairs of xenograft tumors, PDT treated and untreated controls, were harvested four hours after PDT treatment. RNA was isolated and prepared for hybridization to human Affymetrix GeneChip arrays.
