Growth is required for perception of water availability to pattern plant root branches

ABSTRACT: Lateral root branching in higher plants is promoted in regions locally contacting a source of water, and suppressed in regions exposed to low water availability. We found that developmental competence to respond to this environmental signal is limited to growing tissues. We profiled gene expression in regions of the maize primary root exposed to low and high water availability both within and outside of the zone of competence.

Project description:Water availability is a potent regulator of plant development and induces root branching through a process termed hydropatterning. Hydropatterning enables roots to position lateral branches toward regions of high water availability, such as wet soil or agar media, while preventing their emergence where water is less available, such as in air. The mechanism by which roots perceive the spatial distribution of water during hydropatterning is unknown. Using primary roots of Zea mays (maize) we reveal that developmental competence for hydropatterning is limited to the growth zone of the root tip. Past work has shown that growth generates gradients in water potential across an organ when asymmetries exist in the distribution of available water. Using mathematical modeling, we predict that substantial growth-sustained water potential gradients are also generated in the hydropatterning competent zone and that such biophysical cues inform the patterning of lateral roots. Using diverse chemical and environmental treatments we experimentally demonstrate that growth is necessary for normal hydropatterning of lateral roots. Transcriptomic characterization of the local response of tissues to a moist surface or air revealed extensive regulation of signaling and physiological pathways, some of which we show are growth-dependent. Our work supports a "sense-by-growth" mechanism governing hydropatterning, by which water availability cues are rendered interpretable through growth-sustained water movement.

Magnesium Availability Regulates Development of Root Hairs in Arabidopsis thaliana

Project description:Root hairs are frequently reported to be plastic in response to nutrient supply, but relatively little is known about their development in response to magnesium (Mg) availability, and evidence is scarce about the signals involved in this process. Here, we showed that both density and length of root hairs of Arabidopsis decreased logarithmically with increasing Mg supply in the media , which correlated with the initiation of new trichoblast files and likelihood of trichoblasts to form hairs. Low Mg resulted in greater concentrations of reactive oxygen species (ROS) and Ca2+ in the roots and displayed a stronger tip-focused gradient of ROS and cytosolic Ca2+ concentration ([Ca2+]c) during initiation and elongation of root hairs. This gradient could be eliminated by DPI or BAPTA. Application of either DPI or BAPTA to low Mg treatment blocked the enhanced development of root hairs. The opposite was true when the plants under high Mg were supplied with Ca2+ or PMS. Whole-genome transcriptome data revealed that the maximum differential expressed genes involved in ‘stress’, ‘oxidation reduction’, ‘ion transport and homeostasis’ and ‘cell wall organization’. A greater fraction of morphogenetic H-genes and root hair -specific genes as well as genes involved in ‘cell wall structure’ were up-regulated by 7-d treatment of 0.5 μM Mg but down-regulated by 7-d treatment of 10,000 μM Mg. It is concluded that a distinct and previously poorly characterized response of root hair development to Mg availability is presented in Arabidopsis where ROS and Ca2+ are the signaling molecules that control this response.

Distinct early transcriptional regulations by turgor and osmotic potential changes in the root of Arabidopsis

Project description:In a context of climate change, deciphering signaling pathways leading to plant adaptation to drought, water availability, and salt tolerance is a crucial question. A common crossing point of these plant stresses is their impact on plant water potential, a composite physico-chemical variable reflecting the availability of water for the plant biological and biochemical processes such as growth or stomatal aperture. The water potential of plant cells is mainly driven by their turgor pressure and osmotic potential. Here we investigated the effect of a variety of osmotic treatments in the root of Arabidopsis plants grown in hydroponics. Measurement of the cortical cells turgor pressure with a cell pressure probe allowed to control the intensity of the treatments and, in particular, preserve the cortex from plasmolysis. Transcriptome analyses at early time point (15min) showed specific and quantitative transcriptomic responses for both osmotic and turgor pressures in the root. Our results highlight how water-related biophysical parameters can shape the transcriptome and provide putative candidates to explore further the early perception of water stress in plants.

Project description:Nitrogen availability in the soil is a major determinant of crop yield. While the application of fertilizer can substantially increase the yield on poor soils, it also causes nitrate pollution of water resources and high costs for farmers. Increasing the nitrogen use efficiency in crop plants is a necessary step to implement low input agricultural systems. We exploited the genetic diversity present in the world-wide Arabidopsis thaliana population to study adaptive growth patterns and changes in gene expression associated with chronic low nitrate stress, with the aim to identify biomarkers associated with good plant performance under low nitrate availability. Transcription and epigenetic factors were identified as important players in the adaptatiion to limited nitrogen in a global gene expression analysis using the Affymetrix ATH1 chip.
