Project description:Transcriptome profiles of control Lactobacillus plantarum WCFS1 cells were compared with 8% ethanol adapted cells and with 10 min or 30 min 8% ethanol shocked cells. A continuous culture was performed in duplicate on medium containing ehtanol (tE). A shock experiment was performed in duplicate in which ethanol was added to a batch culture immediately after t0 (=control). The culture was sampled at time=0 (t0), time =10 min (t1) and time=30 min (t2).

Project description:Lactobacillus acetotolerans is a major microbe contributing to the Chinese liquor fermentation with unknown function. It can be grown well in a high concentration of ethanol. RNA sequencing (RNA-seq) was performed on L. acetotolerans F28 growing in 12% ethanol to determine important genetic mechanisms for both a short and long term adaption to this environment. A genome-wide transcriptional analysis revealed that the most important genetic elements for L. acetotolerans F28 grown in ethanol are related to high levels of stress response and fatty acid biosynthesis, and a reduction of amino acid transport and metabolism after both a short and long time stress. The fatty acid methyl ester analyses showed that most fatty acids were increased in L. acetotolerans F28 after exposure to ethanol while the unsaturated fatty acid octadecenoic acid (C18:1) was significantly increased. The increasing unsaturated fatty acid biosynthesis in L. acetotolerans F28 might enhance cell membrane fluidity and protect the cells against high concentration of ethanol. Overall, the transcriptome and functional analysis indicated that the elevated stress response and fatty acid biosynthesis, and the decrease of amino acid transport and metabolism might play important roles for L. acetotolerans F28 to adapt to environmental ethanol.

Project description:This paper describes the molecular responses of Lactobacillus plantarum WCFS1 toward ethanol exposure. Global transcriptome profiling using DNA microarrays demonstrated adaptation of the microorganism to the presence of 8% ethanol over short (10-min and 30-min) and long (24-h) time intervals. A total of 57 genes were differentially expressed at all time points. Expression levels of an additional 859 and 873 genes were modulated after 30 min and 24 h of exposure to the solvent, respectively. Ethanol exposure led to induced expression of genes involved in citrate metabolism and cell envelope architecture, as well as canonical stress response pathways controlled by the central stress regulators HrcA and CtsR. Correspondingly, cells grown for 24 h in medium containing 8% ethanol exhibited higher levels of citrate consumption and modified cell membrane fatty acid composition and showed invaginating septa compared with cells grown in liquid medium without ethanol. In addition, these physiological changes resulted in cross-protection against high temperatures but not against several other stresses tested. To evaluate the role of HrcA and CtsR in ethanol tolerance, ctsR and hrcA gene deletion mutants were constructed. The growth rate of the L. plantarum ?ctsR::cat strain was impaired in de Man-Rogosa-Sharpe (MRS) medium containing 8% ethanol, whereas growth of the L. plantarum ?hrcA::cat and ?ctsR ?hrcA::cat mutants was indistinguishable from that of wild-type cells. Overall, these results suggest that the induction of CtsR class III stress responses provides cross-protection against heat stress.

Project description:Bacterial membrane vesicles have been implicated in a broad range of functions in microbial communities from pathogenesis to gene transfer. Though first thought to be a phenomenon associated with Gram-negative bacteria, vesicle production in Staphylococcus aureus, Lactobacillus plantarum, and other Gram-positives has recently been described. Here we characterize MVs from three different Lactobacillus species (L. acidophilus, L. casei, and L. reuteri), determining that the size and protein composition of Lactobacillus-derived MVs have both similarities and differences with those produced by Gram-negative bacteria. Using proteomics, we identified more than 80 protein components from Lactobacillus-derived MVs, including some that were enriched in the vesicles themselves. For each species, vesicular proteins were categorized based on biological pathway and examined for subcellular localization signals in an effort to identify possible sorting mechanisms for MV proteins. Additionally, differences between MVs of other Lactobacillus species and Gram positive bacteria were highlighted. Information in this study will assist in elucidation of the formation and functions of MVs, as well as the development of therapeutic tools for vaccines, diagnosis, and therapeutic delivery.

Project description:Breast cancer is the second leading cause of cancer-related deaths in women (*). Many genetic and behavioral risk factors can contribute to the initiation and progression of breast cancer, one being alcohol consumption. Numerous epidemiological studies have established a positive correlation between alcohol consumption and breast cancer; however, the molecular basis for this link remains ill defined. Elucidating ethanol-induced changes to global transcriptional programming in breast cells is important to ultimately understand how alcohol and breast cancer are connected mechanistically. We investigated early induced transcriptional changes in response to cellular exposure to moderate levels of alcohol. We treated the non-tumorigenic breast cell line MCF10A, and the tumorigenic breast cell lines MDA-MB-231 and MCF7, with ethanol for 6 h, then captured the changes to ongoing transcription using 4-thiouridine metabolic labeling followed by deep sequencing. Only the MCF10A cell line exhibited statistically significant changes to ongoing transcription in response to ethanol treatment. Further experiments revealed some ethanol-upregulated genes are sensitive to the dose of alcohol treatment, while others are not. Gene ontology and biochemical pathway analyses revealed that ethanol-upregulated genes in MCF10A cells are enriched in biological functions that could contribute to cancer development.

Project description:Grapevine is a plant of agronomic interest, which is able to cope with several stresses: heat, drought, and hypoxia, among others. A recent study showed very low oxygen levels inside grape berries, linked to ethanol content. Other studies have established the link between ethanol and tolerance to various stresses: heat stress, drought, and high salinity. The causes of such tolerance induced by ethanol are not well understood. In our study, three-week-old Gamay calli were characterised for their endogenous O2 levels (78 ± 43 µM at 2.1 mm inside the callus) and endogenous ethanol concentration (406 ± 2 µM). Subsequently, a transcriptomic study of these cells was conducted, at 6 and 24 hours post-treatment with 1 mM ethanol. After 6 hours post-treatment, ethanol addition led to 386 Differentially Expressed Genes (DEGs) compared to the controls. Analyses of the gene classes modulated by ethanol showed that many genes involved in heat response were up-regulated, notably the small Heat Shock Proteins (sHSP). Next, we showed that grape cells primed with ethanol experienced less pigment leakage due to heat stress compared to the controls. This result was further supported by a similar observation in Arabidopsis seedlings, for which heat stress-induced electrolyte leakage was reduced by ethanol priming. his study confirms previous observations regarding the beneficial role of ethanol priming in protecting plants against heat stress and provides a comprehensive set of RNA-seq data to further decipher the mechanisms involved in such adaptation. The small heat shock proteins seem involved in this chemical priming response.
