Project description:Purpose: To identify the cellular processes upregulated in fibroblasts after treatment with selected protein(s) or E16.5 skin extract Methods: To explore the mechanism of extract-induced HF neogenesis, adult dermal fibroblasts were cultured in the presence of E16.5 extract or selected protein(s) for 3 days before being tested. Results: we determined how the exposure of adult fibroblasts to E16.5 extract, the selected protein mix or each of the selected proteins and compared the resultant transcriptomes with those of adult DP and embryonic dermal fibroblasts. Conclusions: Gene expression of adult fibroblasts exposed to E16.5 extract and selected proteins mix, the key DP signature genes became upregulated.

Project description:Quiescent (Q) and stress induced premature senescent (SIPS) fibroblasts were treated for the duration of 4 days with growth medium supplemented with a plant extract (1201) from Solidago vigaurea subspecies alpestris. Rna was isolated with Trizol and sent to GATC for next generation sequencing with Ilumina technology. The plant extract proofed to block the negative effects of senescence in human skin fibroblasts in various experiments by delaying the acquisition of a senescent phenotype/favouring a papillary-like phenotype and attenuating the senescence associated secretory phenotype. The RNAseq was performed to understand the underlying molecular mechanism of the observed effects. SIPS was induced by chronic oxidative stress treatment (9 days with 1 h 100 µM H2O2 per day).

Gene expression study in skin fibroblasts

Project description:To investigate the in vivo developmental potential, we generated dedifferentiated-skin fibroblast from adult skin fibroblasts with a FVB background, a different genetic strain from the protein-donor embryonic stem (ES) cell. Primary cardiac fibroblast (cFB) was cultered from an adult C57/BL mouse to compare with ES cells(E14-mES, C57-mES). We also introduced C57 background ES cell-derived extract proteins into cFB by reversible permeabilisation. The ES-like cells (colonies) were expanded with subculture every 3 to 5 days using the standard ES cell culture protocol.

Single-cell RNA-seq of the developing C57/BL6 mouse back skin at E13.5, E16.5 and P0

Project description:We performed single-cell RNA seq on C57/BL6 mouse back skin at E13.5, E16.5, and P0 to study embryonic hair follicle development. We analyzed 15,086 single cell transcriptome profiles from E13.5, E16.5 and newborn mice (postnatal day 0, P0) dorsal skin cells across hair follicle induction, organogenesis, cytodifferentiation stage. Based on t-distributed Stochastic Neighbor Embedding (tSNE) clustering, we identified 14 cell clusters from skin cells and delineated their cell identity gene expression profile. By using Monocle pseudotime ordering analysis, we constructed epithelium/dermal cell lineage differentiation trajectory and revealed sequential activation of key regulons involved during embryonic hair follicle morphogenesis. Our findings here provide molecular landscape during hair follicle epithelium/dermal cell lineage fate decisions.

Project description:We used whole transcriptome sequencing (RNA-Seq) to assess differential gene expression of normal human melanocytes with respect to keratinocytes, fibroblasts and whole skin. We have identified a number of genes and pathways, which are characteristic or unique for MC compared to KC and FB. We also demonstrated the extent of the difference between genes detectable in MC culture and the whole skin. The present results reveal melanocytes as highly proliferative cells with stem cell-like properties.

Project description:Pomegranate skin extract could prevent fatty liver due to high fat diet in adult male Sprague Dawley rat. There are 3 groups of rats, feeding chow diet, high fat diet and high fat diet combined with pomegrante skin extract. After 8 weeks feeding, high fat diet group developped fatty liver but the other two groups still have healthy liver. We used microarrays to detail the global programme of gene expression underlying the fatty liver development and preventive effect of pomegranate skin extract on fatty liver.
