Project description:The aim of this data set is to perform a differential expression analysis between wild type eye-antennal imaginal disc and discs that are homozygous glass mutant gl[60j]. This data set is used to validate Glass target gene predictions identified by i-cisTarget on a set of conserved eye-specific genes. RNA-seq was performed in eye-antennal imaginal discs of two D.melanogaster wild-type strains (Canton S and strain RAL-208 (Jordan et al. 2007, Ayroles et al. 2009)), representing two biological replicates; and in glass mutant (gl[60j]) discs for two technical replicates.

Project description:Transcriptional profiling of mammary tissue irradiated at 10 weeks of age with either 100 cGy sparsely ionizing gamma-rays, or 10 cGy or 30 cGy densely ionizing radiation (350 MeV/amu Si). Mammary tissue was collected 1 weeks, 4 weeks, and 12 weeks post-irradiation. Four radiation treatment groups: sham, 100 cGy sparsely ionizing gamma-rays, 10 cGy or 30 cGy densely ionizing radiation (350 MeV/amu Si). Three time points post-irradiation (1, 4, and 12 weeks). Three or four replicates per time point.

Project description:The edible cyanobacterium Arthrospira is highly resistant to ionising radiation. How theses cells can escape, protect or repair the radiation damage is not known, and requires additional molecular investigation. Therefore, in this study, Arthrospira cells were shortly exposed to different doses of 60Co gamma rays and the dynamic response of Arthrospira cells to irradiation was investigated by monitoring its gene expression and cell physiology, after irradiation. The dynamic transcriptome and associated physiological traits showed clearly two events: (1) an intense global emergency response with general reprogramming of carbon and energy metabolism and (2) a recovery period. Genes expressed during early response indicated a reduction of photosynthesis and reduced tricarboxilic acid (TCA) and Calvin Benson Cycles, while a likely activation of pentose phosphate pathway. Activation of biosynthesis of additional carbon storage molecules, compatible solutes, vitamins and sugars transport occurred after irradiation. In addition the cells enhance the restoration of the redox homeostasis. The repair mechanism of Arthrospira cells involve mainly proteases for cellular cleaning and removal of damaged proteins, single strand DNA repair and restriction modification system. During recovery, Arthrospira relies on the powerful antioxidant Glutathione molecule, for ROS detoxification. The exposed cells expressed during recovery period also again highly the arh genes, coding for a group of novel proteins which were detected in our previous irradiation studies, which confirms our hypothesis that they are a key element in the radiation resistance and merit further detailed investigation. This study provides new insights into phasic response and the cellular pathways involved the radiation resistance of microbial cells, in particularly for photosynthetic organisms as the cyanobacterium Arthrospira.

Project description:Curcumin, a yellow pigment extracted from the rhizome of the plant Curcuma longa (turmeric) has been widely used as a spice and herbal medicine in Asia. It has been suggested to have many biological activities such as anti-oxidative, anti-inflammatory, anti-cancer, chemopreventive, and anti-neurodegenerative properties. We evaluated the impact of curcumin on lifespan, fecundity, feeding rate, oxidative stress, locomotion and gene expression in two different wild type Drosophila melanogaster strains, Canton-S and Ives, under two different experimental conditions. We report that curcumin extended the lifespan of two different strains of Drosophila and was accompanied by protection against oxidative stress, improvement in locomotion and chemopreventive effects. Curcumin also modulated the expression of several aging related genes (genes with age-dependent changes in gene expression) such as mth, thor, InR, and JNK. In order to evaluate the impact of curcumin and aging on gene expression, we first determined which genes were affected by aging alone in Canton S flies. Age-related changes in gene expression were defined as changes in expression levels that occurred between 3 and 40 days of age (median lifespan). Among the 18,880 probe sets in the Affymetrix GeneChip® Drosophila Genome 2.0 Array, 1,383 genes (Data on file, 7.3%, P < 0.05) had statistically significant changes in expression levels during this time frame. We next determined the effect of curcumin on gene expression levels in young and aged Canton S flies. Gene expression were defined as changes in expression levels that occurred between 3 and 40 days of aged flies with or without curcumin-feeding.
