Effect of glucocorticoid steroid regimens on murine dystrophic muscle

ABSTRACT: The goal of this study is to compare the effect of weekly and daily steroid regimens (prednisone) on transcriptional regulation of candidate genes for muscle repair and atrophic response in mdx mice (n=5 mice/group).

Project description:Duchenne muscular dystrophy is caused by genetic defects in the gene encoding dystrophin and leads to progressive muscle degeneration. Glucocorticoid steroids are current mainstay pharmacological regimen to decrease muscle inflammation and prolong the ambulatory period in these patients, but daily intake of glucocorticoids like prednisone and deflazacort causes adverse side effects like osteoporosis, adrenal suppression, insulin resistance and obesity. Intermittent steroid dosing has been proposed as alternative to maintain benefits and limit side effects, but a detailed understanding of the mechanisms underpinning the regimen-specific effects in muscle is still missing. Here we explore how once-daily versus once-weekly prednisone (4 week-long treatment) affect the epigenomic landscape in mdx mouse muscle (genetic model of Duchenne muscular dystrophy; DBA/2J background) through H3K27 acetylation profiles.

Project description:Exogenous glucocorticoids regulate the muscl-fat axis communication, but little attention is paid to their frequency of intake. Here we investigated the transcriptional effects of prednisone in muscle and adipose tissue when dosed as intermittent once-weekly 1mg/kg i.p. versus once-daily 1mg/kg in WT obese mice. Prior to treatment, mice were fed a high-fat diet for 12 weeks. After 12 weeks of treatment, we performed RNA-seq analysis in muscle (quadriceps) and white adipose tissue (ventral fat pad) to quantitate gene expression at isoform level.

Project description:CD4+Foxp3+ regulatory T cells (Tregs) accumulate in skeletal muscle from dystrophin-deficient mdx mice. Analysis of global gene expression in muscles from mdx mice treated with anti-CD25 compared with muscles from mdx mice treated with control antibody revealed that Tregs partially protect mdx mice from muscle pathology and promote muscle repair/regeneration. Mdx mice received 2 ip injections of 100 mg of anti-CD25 mAb (clone PC61) or rat IgG (Jackson Immunoresearch) at days 17 and 20 of age. Muscles were dissected 7 days after the last injection.

Project description:Glucocorticoid steroids are commonly prescribed for inflammatory conditions such as rheumatoid arthritis and acute respiratory distress, but chronic daily use produces an array of adverse effects including muscle wasting and weakness. In contrast, shorter and lower dose glucocorticoid pulses may improve athletic performance, although the mechanisms by which this occurs remain unclear. Muscle is sexually dimorphic and comparatively little is known about how male and female muscle respond to glucocorticoid steroids. In this work, we investigated the impact of a once-weekly glucocorticoid exposure on body mass and skeletal muscle performance, comparing male and female mice. We performed RNA sequencing of isolated myofibers from quadriceps of C57BL/6 male and female mice given vehicle, weekly, or daily prednisone for one month.

Project description:MicroRNA-expression profile of dystrophic single fibers compared to wild type single fibers isolated from different muscles of mdx and C57BL mice. Myofibers were isolated from different muscle types (tibialis, diaphragm and quadriceps) of gender- (male) and age- (3 month old and half) matched wt and dystrophic mice. 9 total samples per animal model (C57BL, mdx), 3 replicates per muscle type.
