The effect of SPARCL1 overexpression on osteosarcoma MNNG-HOS cell line

ABSTRACT: To investigate the altered gene expression levels in SPARCL1 overexpressed cells and vector cells. Each group had a biological repeat (n=3). Results provide insight into the role of SPARCL1 in the progression of osteosarcoma.

Project description:Investigation of whole genome gene expression level changes in human osteosarcoma cell line MNNG/HOS treated by TGF-beta1 for three days (mesophase) and five days (sarcospheres iOSCs), compared to non-treatment cells (residual adherent cells). A three chip study using total RNA cover from three cultures of non-treatment human osteosarcoma cell line MNNG/HOS (residual adherent cells), TGF-beta1 treated three days osteosarcoma cell line MNNG/HOS (mesophase) and TGF-beta1 treated five days osteosarcoma cell line MNNG/HOS ( only collected the suspending sarcospheres iOSCs). Effect of SET knock down on human osteosarcoma cell line HOS gene expression

Project description:SE translocation (SET) functions as an inhibitory protein of protein phosphatase 2A (PP2A), an important tumor suppressor, and contributes to the maintenance of stemness in various cancers. In the present study, we used the Tet on system to suppress SET expression in the osteosarcoma cell line HOS in a doxycycline-dependent manner and analyzed its effect on gene expression. The results revealed that SET is involved in mammalian target of rapamycin complex 1 (mTORC1) and B-lymphoma Moloney murine leukemia virus insertion region-1 (Bmi-1).

Syndecan-2 and apoptosis in osteosarcoma

Project description:Syndecan-2 was found to act as a tumor suppressor in osteosarcoma and to mediate the apoptotic response to cytotoxic agents. To determine new mechanisms that control cell survival and apoptosis in osteosarcoma cells, we aim to show target genes that are involved downstream of this proteoglycan during apoptosis induction. We aim to compare the modifications induced by syndecan-2 overexpression in two different human osteosarcoma cell lines using a lentiviral vector coding the human syndecan-2. Syndecan-2 lentiviral vector (SY) and control empty lentiviral vector (VV) were transducted in two different human osteosarcoma cell lines in a dye-swap experiment.
