Project description:Understanding how trees respond to global change drivers is central to predict changes in forest structure and functions. Although there is evidence on the mode of nitrogen (N) and drought (D) effects on tree growth, our understanding of the interplay of these factors is still limited. Simultaneously, as mixtures are expected to be less sensitive to global change as compared to monocultures, we aimed to investigate the combined effects of N addition and D on the productivity of three tree species (Fagus sylvatica, Quercus petraea, Pseudotsuga menziesii) in relation to functional diverse species mixtures using data from a 4-year field experiment in Northwest Germany. Here we show that species mixing can mitigate the negative effects of combined N fertilization and D events, but the community response is mainly driven by the combination of certain traits rather than the tree species richness of a community. For beech, we found that negative effects of D on growth rates were amplified by N fertilization (i.e., combined treatment effects were non-additive), while for oak and fir, the simultaneous effects of N and D were additive. Beech and oak were identified as most sensitive to combined N+D effects with a strong size-dependency observed for beech, suggesting that the negative impact of N+D becomes stronger with time as beech grows larger. As a consequence, the net biodiversity effect declined at the community level, which can be mainly assigned to a distinct loss of complementarity in beech-oak mixtures. This pattern, however, was not evident in the other species-mixtures, indicating that neighborhood composition (i.e., trait combination), but not tree species richness mediated the relationship between tree diversity and treatment effects on tree growth. Our findings point to the importance of the qualitative role ('trait portfolio') that biodiversity play in determining resistance of diverse tree communities to environmental changes. As such, they provide further understanding for adaptive management strategies in the context of global change.

Project description:The growing-season length of temperate and boreal trees has a strong effect on the global carbon cycle. Yet, a poor understanding of the drivers of phenological processes, such as autumn leaf senescence in deciduous trees, limits our capacity to estimate growing-season lengths under climate change. While temperature has been shown to be an important driver of autumn leaf senescence, carbon source–sink dynamics have been proposed as a mechanism that could help explain variation of this important process. According to the carbon sink limitation hypothesis, senescence is regulated by the interplay between plant carbon source and sink dynamics, so that senescence occurs later upon low carbon inputs (source) and earlier upon low carbon demand (sink). Here, we manipulated carbon source–sink dynamics in birch saplings (Betula pendula) to test the relevance of carbon sink limitation for autumn leaf senescence and photosynthetic decline in a widespread deciduous tree. Specifically, we conducted a gradient of leaf and bud removal treatments and monitored the effects on autumnal declines in net photosynthesis and the timing of leaf senescence. In line with the carbon sink limitation hypothesis, we observed that leaf removal tended to increase total leaf-level autumn photosynthesis and delayed the timing of senescence. Conversely, we did not observe an effect of bud removal on either photosynthesis or senescence, which was likely caused by the fact that our bud removal treatment did not considerably affect the plant carbon sink. While we cannot fully rule out that the observed effect of leaf removal was influenced by possible treatment-level differences in leaf age or soil resource availability, our results provide support for the hypothesis of carbon sink limitation as a driver of growing-season length and move the scientific field closer to narrowing the uncertainty in climate change predictions.

Project description:Hormones effect various plant developmental processes by altering gene expression. The expression of several genes is regulated by plant hormones and many of these genes are regulated commonly and specifically by various hormones. We used microarrays to study the global effect of plant hormones on rice gene expression and identify the genes involved in operlapping and specific transcriptional responses. Rice seedlings of IR64 variety were grown hydroponically for 7-days in a culture room with a daily photoperiodic cycle of 14h light and 10h dark. Seedlings were incubated in water (control) or 50 µM solution of indole-3-acetic acid (auxin, IAA) and benzyl aminopurine (cytokinin, BAP) and 100 µM solution of abscisic acid (ABA), 1-aminocyclopropane-1-carboxylic acid (ethylene derivative, ACC), salicylic acid (SA) and jasmonic acid (JA) for 3h. The 5 micrograms of total RNA sample isolated from each tissue sample was processed for microarray analysis according to Affymetrix protocol. Two biological replicates for each sample (two controls, IAA, BAP, ABA, ACC, SA and JA) were used for microarray analysis.

Project description:Technology advances can revolutionize Precision Forestry by providing accurate and fine forest information at tree level. This paper addresses the question of how and particularly when Unmanned Aerial System (UAS) should be used in order to efficiently discriminate deciduous tree species. The goal of this research is to determine when is the best time window to achieve an optimal species discrimination. A time series of high resolution UAS imagery was collected to cover the growing season from leaf flush to leaf fall. Full benefit was taken of the temporal resolution of UAS acquisition, one of the most promising features of small drones. The disparity in forest tree phenology is at the maximum during early spring and late autumn. But the phenology state that optimized the classification result is the one that minimizes the spectral variation within tree species groups and, at the same time, maximizes the phenologic differences between species. Sunlit tree crowns (5 deciduous species groups) were classified using a Random Forest approach for monotemporal, two-date and three-date combinations. The end of leaf flushing was the most efficient single-date time window. Multitemporal datasets definitely improve the overall classification accuracy. But single-date high resolution orthophotomosaics, acquired on optimal time-windows, result in a very good classification accuracy (overall out of bag error of 16%).

Project description:Amplicon-based analysis of 16S rRNA genes and transcripts was used to assess the effect of tree species composition on soil bacterial community structure and function in a temperate deciduous forest. Samples were collected from mono and mixed stands of Fagus sylvatica (beech), Carpinus betulus (hornbeam), Tilia sp. (lime), and Quercus sp. (oak) in spring, summer, and autumn. Soil bacterial community exhibited similar taxonomic composition at total (DNA-based) and potentially active community (RNA-based) level, with fewer taxa present at active community level. Members of Rhizobiales dominated at both total and active bacterial community level, followed by members of Acidobacteriales, Solibacterales, Rhodospirillales, and Xanthomonadales. Bacterial communities at total and active community level showed a significant positive correlation with tree species identity (mono stands) and to a lesser extent with tree species richness (mixed stands). Approximately 58 and 64% of indicator operational taxonomic units (OTUs) showed significant association with only one mono stand at total and active community level, respectively, indicating a strong impact of tree species on soil bacterial community composition. Soil C/N ratio, pH, and P content similarly exhibited a significant positive correlation with soil bacterial communities, which was attributed to direct and indirect effects of forest stands. Seasonality was the strongest driver of predicted metabolic functions related to C fixation and degradation, and N metabolism. Carbon and nitrogen metabolic processes were significantly abundant in spring, while C degradation gene abundances increased from summer to autumn, corresponding to increased litterfall and decomposition. The results revealed that in a spatially homogenous forest soil, tree species diversity and richness are dominant drivers of structure and composition in soil bacterial communities.

Project description:Two fiber tissues harvested 10 days post anthesis from upland cotton trees grown under the same green house conditions except for different seasons of the year were used for RNA extraction. Small RNA molecules under 30 bases were amplified and isolated from an agarose gel. The purified DNA was used directly for cluster generation and sequencing analysis using the Illumina Genome Analyzer according to the manufacturer's instructions. The 35nt sequence tags from sequencing went through data cleaning first, which included getting rid of the low-quality tags and several kinds of contaminants from the 35nt tags. All clean tag sequences with copy numbers were then summarized into a fasta format file.

Project description:Lateral organ development is important for cucumber yield, while the molecular mechanism controlling leaf and floral organ development in cucumber remain elusive. In this report, a novel EMS-mutaginized mutant, round leaf (rl) was distinguished with remarkable round leaf shape, abnormal floral organ and inhibited tendril outgrowth in early development phase. Moreover, the ovule organogenesis disrupted completely in parthenocarpy fruit of rl. MutMap+ analysis revealed that RL encodes a protein kinase PINOID (CsPID, Csa1G537400). A non-synoymous SNP in the second exon of CsPID resulted in an amino-acid substitution from Arg in the wild type to Lys in the rl mutant. CsPID was down-regulated in rl mutant and preferentially expressed in young leaf, and flower buds. IAA quantification showed that rl plants exhibited a lower IAA content than wild type in ovary and blade edge. IAA immunolocalization results confirmed the IAA content alteration in rl plants. Transcriptome profile analysis further suggested IAA biosynthesis, polar transport and signal transduction genes participated in the leaf and floral development process by CsPID. Biochemical analyses showed that CsPID may regulate leaf shape by interacting with CsREV. In conclusion, this study revealed that the extensive genetic architecture of lateral organ organogenesis and development via CsPID regulating auxin polar transport action in cucumber.
