Comparison of microarray expression data from 23 day mouse germ cells from control and Mgat1 conditional knockout mice

ABSTRACT: Loss of Mgat1 in spermatogonia was investigated in germ cells from 23 day males. Gene expression changes induced by deletion of Mgat1 were determined using the Affymetrix GeneChip Mouse Gene 2.0 ST Array. We include the expression data obtained from three replicate aliquots of pooled testis total RNA obtained from six control or six conditional knockout Mgat1 testis germ cells from 23 dpp males, Three replicates of the respective control and Mgat1 cKO pools were used for microarray analysis using Affymetrix GeneChip Whole Transcript Plus Reagent Kit. We identified differential expressed genes with a significance level of ANOVA and FDR p < 0.05 and an absolute fold-change (linear) less than -2, and greater than 2.0.

Translational activation of developmental mRNAs during neonatal mouse testis development

Project description:The sequence of gene regulatory events that drive neonatal germ cell development in the mammalian testis is not yet clear. We assessed changes in mRNA utilization in the neonatal testis at 1 and 4 dpp, times when the testis contains quiescent gonocytes (1 dpp) and proliferating spermatogonia (4 dpp). There are not thought to be major changes in the nature or number of somatic cells over that interval. We used microarrays to detail the global expression levels of mRNA distribution between non-translating mRNAs and efficiently translating mRNAs during testis development at 1 dpp and 4 dpp Extracts of mouse testes from 1 dpp and 4 dpp CD-1 mice were fractionated over a sucrose gradient in triplicate. Gradients were fractionated using an ISCO gradient fractionation system equipped with a UA-6 detector. The position of the 80S peak defined the boundary between translating and non-translating mRNAs. Pooled non-polysomal (non-translating; fractions 1-3) and heavy polysomal (efficiently translating, fractions 7-13) RNAs were isolated from sucrose gradient fractions for hybridization on Affymetrix 420 2.0 microarrays.

Transcription profiling of rat testis aafter acute antibody mediated FSH suppression

Project description:To unravel the genes that underpin the effects of FSH on spermatogenesis, testicular gene expression was compared in juvenile rats after FSH suppression to littermate controls. Based on existing data, it is apparent that the dominant function of FSH shifts between 9 and 18 day postpartum (dpp) during the first wave of spermatogenesis from driving Sertoli cell proliferation to support of germ cells. To enable comprehensive analysis of the impact of acute in vivo FSH suppression on Sertoli and germ cell development and to determine the genes that underpin these affects, FSH was selectively suppressed in Sprague Dawley rats by passive immunisation for 4 days prior to testis collection at 18 days postpartum. FSH suppression did not affect Sertoli cell proliferation at 18 dpp, but germ cell numbers were decreased at 18 dpp following FSH suppression, with a corresponding increase in germ cell apoptosis measured. Sixty transcripts were measured as changed at 18 dpp in response to 4 days of FSH suppression, as assessed using Affymetrixâ¢ microarrays. Some of these are known as Sertoli cell-derived FSH-responsive genes (e.g. StAR, cathepsin L, insulin-like growth factor binding protein-3), while others encode proteins involved in cell cycle and survival regulation (e.g. cyclin D1, scavenger receptor class B 1). These data demonstrate that FSH affects germ cells in vivo, by supporting germ cell viability at day 18. This model enabled identification of candidate genes that contribute to the FSH-mediated pathway by which Sertoli cells support germ cells. Experiment Overall Design: Sprague Dawley male rats at day 14 postpartum received either a rat FSH antibody raised in sheep or a control sheep immunoglobulin for 4 days. RNA was extracted from the testis and hybridisation on Affymetrix microarrays was conducted.

Expression data from gonads of Meioc+/+ and Meioc-/-mice

Project description:Meiosis is a specialized division that is specifically engaged in germ cells. This program is finely and dichotomically regulated: germ cells enter meiosis at fetal life in ovaries (13.5 dpc) and only at post-natal life in testes (8 dpp). We used microarrays to determine the gene expression modifications in Meioc mutant gonads displaying early meiotic defects. 14.5 dpc ovaries and 8 dpp testes from Meioc+/+ and Meioc-/- mice (NMRI) were dissected and processed for RNA extraction and hybridization on Affymetrix microarrays. Two pools were analysed for each genotype and each sex. We sought to constitutes pairs (A and B) of wild type and knock-out pools from embryos and pups that were homogenous for developmental stages and from matched littermates. 10 ovaries from 5 embryos composed each pool for females and 4 testes from 4 pups composed each pool for males.

Ubb Knockout Mouse Testis

Targeted disruption of the polyubiquitin gene Ubb results in male and female infertility in mice with germ cells arrested at meiotic prophase I. Whole testes from wild-type (WT) and Ubb−/− (KO) mice were harvested at 7, 14, 21, and 28 days postpartum. Total RNA was extracted and hybridized to Affymetrix Mouse Genome 430 2.0 arrays.

Deletion of Mgat2 in Spermatogonia Blocks Spermatogenesis

Project description:Identifying factors required for spermatogenesis is important for understanding mechanisms of male fertility. Inactivation of the Mgat1 or Man2a2 gene leads to a block in spermatogenesis causing infertility in male mice. In both cases multi-nucleated germ cells (MNC) are formed and there are no sperm in the epididymis. MGAT1 GlcNAc-transferase initiates complex N-glycan synthesis and MAN2A2 mannosidase generates the substrate for MGAT2 GlcNAc-transferase which transfers GlcNAc to form a biantennary complex N-glycan. Deletion of Mgat2 can leave its substrate, a hybrid N-glycan terminating in a single GlcNAc, to accumulate or the single GlcNAc may be extended to with Gal and GlcNAc to form polylactosamine. In this paper we show that conditional deletion of Mgat2 in spermatogonia via Stra8-iCre caused a block in spermatogenesis prior to the formation of round spermatids. This phenotype differs from deletion of Mgat1 by Stra8-iCre which generates MNC of round and elongated spermatids. In addition, RNA-seq analysis of germ cells prepared at 15 days after birth revealed a unique transcriptomic landscape in Mgat2[-/-] germ cells. Bioinformatic analyses predicted potential roles for ERK and AKT activities which were validated by western analyses. By contrast, Mgat1[-/-] germ cells have reduced ERK activity and unchanged AKT activity. Therefore, the defective spermatogenesis observed following removal of MGAT1 or MGAT2 must arise from the different immature N-glycans that accumulate rather than from the loss of complex N-glycans which is common to both.
