Project description:MS/MS spectra were collected from face of 6 individuals before and during 2 weeks of application of a Colgate facial cleanser. During weeks 1-4 of washout period, volunteers did not use any personal beauty product and they washed their face only with water. During the following 2 weeks, all participants applied the Colgate facial cleanser daily on their face.
Samples were collected at the end of the washout period on week 4. During facial cleanser application (weeks 5 and 6), samples were collected once a week (beginning of week 5 and end of week 6) right after the first usage then 4, 8 and 24 hours after the first usage. For each timepoint, 9 samples were collected from different locations on the face.

Project description:In the present study, a defect was created in the patellar enthesis of a rat model, and treatment was provided using two versions of a biphasic silk fibroin scaffold showing two different transition morphology between the phases. The study included two scaffold-treated groups, two control groups, and one native/healthy group. For the scaffold-treated groups, the defect was treated by implanting one of the two versions of the biphasic scaffolds. One scaffold showed a mixed/interconnected transition zone between the tendon and bony part of the construct (S-MT) while the transition in the other scaffold was abrupt (S-AT). Two control groups were included. In the first group, a defect was created in the patellar enthesis and the tendon was sutured back over the defect and to the bone. In the second group, a transversal cut was performed at the tendon part of the enthesis and sutured back. Samples were collected (n=8) per group at two time points (4 weeks and 12 weeks) after the surgery. Healthy (contralateral) samples were used as reference. The Qiagen Osteogenesis Pathway-focused PCR array was utilized to profile the expression of relevant osteogenic, chondrogenic, and tenogenic genes in samples from healthy, injured and treated patellar entheses samples.

Project description:Epigallocatechin-3-gallate (EGCG), a polyphenol, influences cutaneous wound healing due to its anti-angiogenic, anti-inflammatory and anti-oxidant properties. We previously demonstrated the role of EGCG in scarring in ex vivo human scar models. A double-blind randomised-controlled trial. Here, we evaluated application of topical EGCG compared with placebo in 62 healthy human volunteers, over 8 weeks, in scars created in their upper inner arms using skin punch biopsies. RNA sequencing was used here based on 45 samples: day 0 (n=3), week 1 (n=3 placebo, n=3 EGCG), week 2 (n=3 placebo, n=3 EGCG), week 3 (n=3 placebo, n=3 EGCG), week 4 (n=3 placebo, n=3 EGCG), week 5 (n=3 placebo, n=3 EGCG), week 6 (n=3 placebo, n=3 EGCG), week 8 (n=3 placebo, n=3 EGCG). The common differentially expressed genes identified between treated (EGCG) and placebo samples were grouped according to time point (Zonal priming: weeks 1 and 2; Direct application post scar formation (2 weeks post-biopsies): weeks 1, 2, 3, 4, 6). Further exploration of these genes using Gene Ontology and Ingenuity Pathway Analysis revealed gene signatures of relevant biological processes and pathways. Here, RNA sequencing revealed a number of angiogenic, inflammatory and antioxidant genes that were significantly downregulated with EGCG particularly at week 1 including; TPSAB1, VEGFA, EDF, IL17F, IL1A, IL1RL1, IL36A, IL36G, SOD3.

Project description:Interventions: This study is a cross-over design and will be done in 4 weeks and include 2 periods. The volunteers will be randomized in two groups: one group will start with the intervention period and the other one will start with the control period. After, these 2 weeks of intervention or control periods, the volunteers will start the control period and intervention period respectively for another 2 weeks. The control period, is meant to reduce to a minimum the ingestion of E171 in order to have all the volunteers to start the same level of E171. During this period the volunteers will receive a list of products to avoid like cookies, chewing gum, certain toothpaste that are known to contain a significant amount of E171.The intervention period, is meant to control the amount of E171 that the volunteers will ingest. A concentration, 0.82 mg/kg bw/day, corresponding to a normal daily consumption will be given to the volunteers. The quantity will be adjusted to the body weight and dispersed in yoghurt that will be eaten at breakfast, lunch and dinner.
Primary outcome(s): The main outcome parameter in this study is the difference in the gene expression profiles due to exposure to the food additive E171.
In order to evaluate the impact of the food additive E171 on gene expression changes and to modify molecular processes involved in human cancer development colon biopsies will be analysed for transcriptomic responses to the exposure to E171.

Project description:Previous work has demonstrated that elevated maternal lipid intake (particularly from dairy products) is associated with increased lipids and altered fatty acid profile in milk produced by healthy lactating women. We investigate our primary hypothesis that a maternal diet rich in full-fat dairy products would simultaneously increase milk lipid percent and expression of genes related to the uptake and/or de novo biosynthesis of milk lipids. Within a a randomized, crossover study we performed a microarray analysis comparing 6 lactating women with diets rich in full-fat or non-fat dairy products. Each woman consumed one of the two diets during timepoint one, and after a two week washout period, switched over to the other diet. The timepoint value indicates whether the particular dietary treatment was consumed first or second.

Project description:Dietary supplementation of vitamin D is commonly recommended to patients with multiple sclerosis (MS). We investigated the effect of 1,25-dihydroxyvitamin-D3 (1,25-(OH)2D3) on the brain proteome in the cuprizone model during remyelination. Mice were demyelinated with dietary cuprizone for 7 weeks. The mice received intra-peritoneal injections of 1,25-(OH)2D3 or placebo twice a week, from week 6 and throughout week 10. Brain samples were taken after 7 weeks (demyelinated), after 8 weeks (1 week remyelination) and after 10 weeks (3 weeks of remyelination). The six experimental groups were labeled with TMT, mixed mode HPLC fractionation, and applied to LC-MS which enabled quantification of 5062 proteins with high confidence.

Project description:Interventions: This study is a cross-over design, including only healthy volunteers. A cross-over design was chosen because it is the most appropriate design for this study: it will reduce the individual differences by allowing every subject to be his own control.
This study will be done in 4 weeks and include 2 periods. The time of exposure is 2 weeks. Indeed, it takes 5 to 7 days for the turnover of the epithelial cells in the gut. This will ensure the exposure of all the cells of the luminal surface of the colon.

In order to verify the compliance of the subjects, once in every period the subjects will prepare a package that include exactly what they ate and drank all day. This food package will be mixed and tested for TiO2 content.
Primary outcome(s): The primary outcome parameters are differences in transcriptomic markers after consumption of food additive E171. These outcomes in humans will shed light on the relevance of markers identified in preclinical studies, and how the animal data on the risk of facilitation of tumour growth can be extrapolated to humans

Project description:Scottish trained individuals (SCO; n = 18) living and training at or near sea-level and Kenyan runners (KEN; n = 20) living and training at ~2,150 m received rHuEPO injections of 50 IUâ kg-1 body mass every two days for four weeks. Blood was collected at baseline, during rHuEPO administration and for four weeks after administration. Eight time points per subject were selected for gene expression analysis; two baseline samples, then one sample at two days, two weeks and four weeks after the first rHuEPO injection and one week, two weeks and four weeks after ceasing rHuEPO administration. RNA was extracted from blood stabilized in Tempus Blood RNA tubes (Life Technologies), amplified, labelled and hybridized to Illumina HumanHT-12v4 Expression BeadChips (Illumina). Expression data was analysed using Rank Products with a Benjamini-Hochberg 5% false discovery rate and an additional 1.5 fold-change threshold. The expression profiles of a subset of target genes was further confirmed using QuantiGene Plex assay (Affymetrix) and Limma (linear models for microarray).

Project description:We performed survival analysis of control and MCD groups, and explored underlying tumor suppression mechanisms after dietary intervention, focused on alterations in the energy-dependent signaling pathways, histone modifications, and global gene expression differences on cDNA microarray study. Illumina arrays: Five- week- old male C57BL6 mice were randomly divided into two groups and fed control diet (control group, LabDiet, Brentwood, MO, USA) or moderate restriced carbohydrate diet formula (MCD, Treat group) in a specific pathogen free zone. All procedures were approved by the institutional animal use and care committee. Following a preliminary feeding of each diet formula for one week, 1 × 106 B16F10 cells (suspended with 100 μl of PBS) were subcutaneously injected into the back of the mice. After 2 weeks of diet supplementation, all mice were sacrificed. Tumor tissue was excised for cDNA microarray.
