GNPS - Colgate facial cleanser longitudinal study_Skin samples

ABSTRACT: MS/MS spectra were collected from face of 6 individuals before and during 2 weeks of application of a Colgate facial cleanser. During weeks 1-4 of washout period, volunteers did not use any personal beauty product and they washed their face only with water. During the following 2 weeks, all participants applied the Colgate facial cleanser daily on their face. Samples were collected at the end of the washout period on week 4. During facial cleanser application (weeks 5 and 6), samples were collected once a week (beginning of week 5 and end of week 6) right after the first usage then 4, 8 and 24 hours after the first usage. For each timepoint, 9 samples were collected from different locations on the face.

Volunteers were asked to follow instructions required over our 9 weeks study: - We provided the same head to toe shampoo to all volunteers, that was used during shower for the first 6 weeks. - Week 1 to week 3: no other beauty products was used. - Week 4 to week 6: we provided 5 selected commercial beauty products that all volunteers applied daily on specific body part (deodorant for armpits, soothing foot powder for feet, sunscreen for face and moisturizer for arm back). - Week 7 to week 9: back to normal routine. - Samples were collected every week, before during and after application of beauty products.

Human plasma LC-MSMS for predicting drug-response progonosis in depressed patients at 0, 1, 4 and 10 visiting weeks

Project description:Patients with major depressive disorder (MDD) have a high relapse rate and first prescribed selective serotonin reuptake inhibitors (SSRIs). The selection process of an antidepressant agent is primarily guided by trial and error. If patients do not benefit from the initial course of antidepressant medication for at least 4-6 weeks, additional therapeutic strategies are required to gain remission, including switching within and between classes of antidepressants. In order to discovery SSRI-related plasma protein markers, we designed a retrospective study. In this study, four longitudinal plasma samples were collected from 10 patients during the 10-week drug reaction period after being diagnosed with depression, and more than a thousand plasma proteins were identified by highly sensitive nanoflow liquid chromatography-tandem mass spectrometry (LC−MS/MS).

Project description:Analysis of changes of gene expression profiles in the left ventricle (LV) during the progression of heart failure (HF) in the canine tachypacing induced HF model. Gene expression profiling was performed on samples collected at different time points representing various stages of LV dysfunction, i.e. tachypaced for 3 days (Day-3), 1 week (Week-1), 2 weeks (Week-2), 3-4 weeks (End stage), and unpaced controls (Day-0). Results provide insights into molecular mechanisms of heart failure progression. Keywords: time course This work aims to study changes of gene expression profiles in the left ventricle (LV) during the progression of heart failure (HF) in the canine tachypacing induced HF model. LV transmural tissue sections were collected at different time points after initiating rapid pacing representing various stages of LV dysfunction, i.e. tachypaced for 3 days (Day-3), 1 week (Week-1), 2 weeks (Week-2), 3-4 weeks (End stage), and unpaced controls (Day-0). As biological replicates, tissues were collected separately from 3 individual dogs at each of the time points. In total we collected 15 tissue samples for all 5 time points. Three RNA samples were independently isolated from each of the 15 tissue samples as technical replicates. The resulting 45 RNA samples were used for microarray experiment. Affymetrix gene expression profiles of the 45 RNA samples were used for all data analysis in this study.

Project description:Afferent Lymph was collected from up to 5 pen raised, nematode naïve, outbreed Romney sheep (Tag No’s 1001, 1002, 1003, 1004, 1006) over a 12 week period. During this time a Pre-Infection sample was collected, the sheep were immunised by 3 truncated (2 week) T. colubiformis infections (40,000 Tc) followed by a Challenge Infection with 40,000 T. colubiformis L3 (Challenge Infection weeks 1-3). The Challenge Infection will enable us to determine changes in gene expression when the sheep gut rejects the parasitic nematodes.

Exposure of Apis mellifera to anti-Varroa destructor formic acid treatment induces significant proteomic alterations.

Project description:The parasitic mite Varroa destructor is a major threat to the health and productivity of Apis mellifera hives and induces its effect by feeding on the fat body of larvae/pupae and transmitting viruses. Control of Varroa populations can be attempted using a variety of treatments and the effect of one of these (the formic acid containing - Mite Away Quick (MAQ) strips) on the proteome of A. mellifera was assessed in this work. Samples of A. mellifera were isolated from hives one week prior to MAQ treatment, during the week of treatment and for two weeks after end of treatment, proteins were extracted and analysed by label free quantitative proteomics. The results indicated that samples isolated during the week of treatment showed increased abundance of a range of cuticular proteins (+ 2.65 fold to + 6.64 fold) and decreased abundance of proteins that deal with xenobiotics (Cytochrome P450 subunits -11.38 fold to -2.16 fold) Interestingly some proteins associated with the oxidative phosphorylation pathway were increased in abundance (Cox5a and Cox5b) but others such as (Coxfa4) were decreased. The results presented here reveal that application of MAQ strips caused a dramatic disruption to the proteome of A. mellifera but the effect is transient and that by two weeks after the end of treatment the proteome has returned to resemble that of the untreated control. While MAQ strips are effective in reducing Varroa populations, the results presented here indicate they can adversely affect the proteome of A. mellifera and may contribute to the elevated stress in hives previously affected by Varroa parasitisation.
