ABSTRACT: Formulations with herbal medicines (HM) may emerge as alternatives for topical treatment of vaginal candidiasis when the resistance to antifungal drugs has increased in Candida species. We developed a vaginal formulation with HM (S. adstringens shell extract characterized by molecular networking, and M. alternifolia Chell essential oil, 46% of terpinen-4-ol).

RNAseq analysis of antibacterial mechanism of Cinnamomum camphora essential oil against Escherichia coli

Project description:Transcriptome analysis plays a central role in elucidating the complexity of gene expression regulation in Escherichia coli. By analyzing the transcriptomics of E. coli treated with water, acetone, and Cinnamomum camphora essential oil, the inhibitory mechanism of the essential oil on the human intestinal microbe was studied. The results showed that the inhibitory effect of the essential oil on E. coli increased with an increase in concentration; 1/4 minimum inhibitory concentration was the reaction equilibrium point. RNAseq transcriptomic comparison indicated that the essential oil inhibited the growth of E. coli by inhibiting the metabolism, chemotaxis, and some resistance reactions of them, while E. coli maintained its life activities by enhancing its resistance reactions. These results are of great importance to the study of the medical use of C. camphor essential oil and gene regulation in E. coli under stress conditions.

Genome-wide Translational Response of Candida albicans to Fluconazole Treatment

Project description:Azoles are commonly used for the treatment of fungal infections and the ability of human fungal pathogens to rapidly respond to azole treatment is critical for the development of antifungal resistance. While the role of genetic mutations, chromosomal rearrangements and transcriptional mechanisms in azole resistance has been well-characterized, very little is known about post-transcriptional and translation mechanisms that drive this process. In addition, most previous genome-wide studies have focused on transcriptional responses to azole treatment, and likely serve as an inaccurate proxies due to extensive post-transcriptional and translational regulation. In this study we use ribosome profiling to provide the first picture of the global translational response of a major human fungal pathogen, Candida albicans, to treatment with fluconazole, one of the most widely used azole drugs. We identify sets of genes showing significantly altered translational efficiency (TE), including genes associated with a variety of biological processes such as the cell cycle, DNA repair, cell wall/cell membrane biosynthesis, transport, signaling, DNA- and RNA-binding activities and protein synthesis. Importantly, while there are similarities and differences among gene categories that are regulated by fluconazole at the translational vs. transcriptional levels, we observe very little overlap among individual genes controlled by these mechanisms. Our findings suggest that C. albicans possesses distinct translational mechanisms that are important for the response to antifungal treatment, which could eventually be targeted by novel antifungal therapies.

Interplay between acetylation and ubiquitination of Isw1 confers multidrug resistance in Cryptococcus neoformans

Project description:Lysine acetylation and ubiquitination are one of many protein modifications and play a crucial role in the biological regulation of many organisms, but little is known about the relationship between acetylation and ubiquitination few. Here, the Isw1 protein is an important member of the chromatin remodeling complex, and we performed single-protein modification mass spectrometry detection of the C. neoformans Isw1 protein and site mutations for both detected modifications. The data showed that the two modifications of Cryptococcus neoformans Isw1 protein have a balance of each other. Acetylation can maintain protein stability and maintain protein function, while ubiquitination can reduce protein level and maintain Isw1 protein expression. The expression level of Isw1 protein leads to resistance to antifungal drugs. These results reveal the resistance mechanism of Isw1 protein of Cryptococcus neoformans to antifungal drugs.

Lansoprazole interferes with fungal respiration and acts synergistically with amphotericin B against multidrug-resistant Candida auris

Project description:Candida auris has emerged as a problematic fungal pathogen associated with high morbidity and mortality. Amphotericin B (AmB) is the most effective antifungal used to treat invasive fungal candidiasis, with resistance rarely observed among clinical isolates. However, C. auris possesses extraordinary resistant profiles against all available antifungal drugs, including AmB. In our pursuit of potential solutions, we conducted a screening of a panel of 727 FDA-approved drugs and identified the proton pump inhibitor lansoprazole (LNP) as a potent enhancer of AmB’s activity against C. auris. LNP also potentiates the antifungal activity of AmB against other medically important species of Candida and Cryptococcus. Our investigations into the mechanism of action unveiled that LNP metabolite(s) interact with a crucial target in the mitochondrial respiratory chain (complex III, known as cytochrome bc1). This interaction increases oxidative stress within fungal cells. Our results demonstrated the critical role of an active respiratory function in the antifungal activity of LNP. Most importantly, LNP restored the efficacy of AmB in an immunocompromised mouse model, resulting in a 1.7-log (~98%) CFU reduction in the burden of C. auris in the kidneys. Our findings strongly advocate for a thorough and comprehensive evaluation of LNP as a cytochrome bc1 inhibitor for combating drug-resistant C. auris infections.

Identification and characterization of potent, broad-spectrum antifungal compounds using a yeast reporter bioassay.

Project description:New antifungal drugs are urgently needed due to the currently limited selection, the emergence of drug resistance, and the toxicity of several commonly used drugs. To identify drug leads, we screened small molecules using a Saccharomyces reporter bioassay in which the yeast heterologously expresses Hik1, a group III hybrid histidine kinase (HHK) from Magnaporthe grisea. Group III HHKs are integral in fungal cell physiology, and highly conserved throughout this kingdom; they are absent in mammals, making them an attractive drug target. Our screen identified compounds 13 and 33, which showed robust activity against numerous fungal genera including Candida, Cryptococcus and molds such as Aspergillus and Rhizopus. Drug-resistant Candida from patients were also highly susceptible to compounds 13 and 33. While the compounds do not act directly on HHKs, microarray analysis showed that compound 13 induced transcripts associated with oxidative stress, and compound 33, transcripts linked with heavy metal stress. Both compounds were highly active against Candida biofilm, in vitro and in vivo, and exerted synergy with fluconazole, which was inactive alone. Thus, we identified potent, broad-spectrum antifungal drug leads from a small molecule screen using a high-throughput, yeast reporter bioassay. Coordination of fungal biofilm development by extracellular vesicle cargo

Project description:The fungal pathogen Candida albicans can form biofilms that protect it from drugs and the immune system. The biofilm cells release extracellular vesicles (EVs) that promote extracellular matrix formation and resistance to antifungal drugs. Here, we define functions for numerous EV cargo proteins in biofilm matrix assembly and drug resistance, as well as in fungal cell adhesion and dissemination. We use a machine-learning analysis of cargo proteomic data from mutants with EV production defects to identify 63 candidate gene products for which we construct mutant and complemented strains for study. Among these, 17 mutants display reduced biofilm matrix accumulation and antifungal drug resistance. An additional subset of 8 cargo mutants exhibit defects in adhesion and/or dispersion. Representative cargo proteins are shown to function as EV cargo through the ability of exogenous wild-type EVs to complement mutant phenotypic defects. Most functionally assigned cargo proteins have roles in two or more of the biofilm phases. Our results support that EVs provide community coordination throughout biofilm development in C. albicans.
