Project description:Polycyclic aromatic hydrocarbons (PAHs) are a class of hundreds of structurally similar chemicals ubiquitously present in our environment. They are created during the incomplete combustion of organic materials, such as oil, wood, tobacco, and charbroiled meat. As such, human exposure to mixtures of PAHs can occur through consumption of PAH-containing foods and water, inhalation of polluted air, or dermal contact. Several PAHs have been classified as carcinogenic to humans or probably carcinogenic to humans by the International Agency for Research on Cancer. The mice in this study were exposed to a mixture of 4 PAHs, benzo(a)pyrene, benz(a)anthracene, benzo(b)fluoranthene, and chrysene. In the present study, we sought to determine the dose-dependent changes in gene expression upon oral exposure to this PAH mixture in the lung tissue. Adult male MutaTMMouse were exposed to three doses of the mixture of 4 PAHs or vehicle control (olive oil) for 28 days and sacrificed three days after the final exposure.

Project description:Perfluoroalkyl substances (PFAS) are a group of synthetic chemicals that are resistant to biodegradation and are environmentally persistent. PFAS are found in many consumer products including non-stick cookware, food packaging materials, upholstery, and personal care products. Accordingly, PFAS are a major source of water and soil contamination. Although use of many PFAS have been phased out, they continue to be detected in human and animal fluids, including human follicular fluid. This study investigated the effects of an environmentally relevant PFAS mixture [perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), perfluorohexanesulfonic acid (PFHxS)] on the transcriptome and function of human granulosa cells (hGCs). PFOA, PFOS, and PFHxS were detected in 100% of follicle fluid samples. Increased cell proliferation was observed in hGCs treated with the PFAS mixture with no impacts on cellular apoptosis. The PFAS mixture also altered steroid hormone synthesis, increasing both FSH-stimulated and basal progesterone secretion and concomitant upregulation of STAR protein. RNA sequencing revealed inherent differences in transcriptomic profiles in hGCs after PFAS exposure. This study demonstrates functional and transcriptomic changes in hGCs after exposure to a PFAS mixture, improving our knowledge about the impacts of PFAS exposures and female reproductive health. These findings suggest that PFAS compounds have the potential to disrupt normal granulosa cell function with possible long-term consequences on overall reproductive health.

Project description:Transcriptional profiling of mussel (Mytilus galloprovincialis) digestive gland tissue comparing control tissue with tissue obtained from animals exposed for four days to sublethal amounts of Nickel, Chlorpyrifos, or their mixture. Background: Mixtures of chemicals present in aquatic environments may elicit toxicity due to additive or synergistic effects among the constituents or ‘vice versa’ the adverse outcome may be reduced by antagonistic interactions. Deviations from additivity should be explained either by the perturbations of toxicokinetic parameters and/or chemical toxicodynamics. We addressed this important question in marine mussels exposed subchronically to a binary mixture made of two wide-spread pollutants: the heavy metal Nickel and the organic phosphorus pesticide Chlorpyrifos. To this aim, we proposed the use of a systems approach based on the evaluation and integration of different disciplines, i.e. high throughput gene expression profiling, functional genomics, stress biomakers and toxicokinetics. Results: Stress biomarkers showed statistically significant antagonistic deviations from the reference model systems to predict mixture toxicity which are based either on simple additivity or non-interaction. While toxicokinetic modeling did not explain mixture interactions, gene expression profiling and further functional genomics analysis provided clues that the decrement of toxicity may arise from the development of specific toxicodynamics. Multivariate statistics of microarray and quantitative PCR data showed two separate patterns for the single chemicals and a composite complex profile for the mixture suggesting the occurrence of interactive molecular responses. The latter signature accounted for differentially expressed genes whose relative expression values were either in trend or in contrast with those found due to single substance treatments and a relevant set of sequences which were exclusive of the mixture gene list. Conclusion: The functional genomics assessment fits with biological data to indicate the occurrence of different toxicodynamic events and ‘in general’ a decrease of toxicity, driven by the mitigation or even abolishment of lysosomal responses such as the lipid metabolic disorder observed exclusively in single chemical-exposed samples. Furthermore, our results emphasized the importance of the application of mechanistic approaches and the power of systems assessment to study toxicological responses in ecological relevant organisms.

Project description:One important obstacle in using softwood in biorefineries is its recalcitrance to enzymatic saccharification. In the present work, Thermothielavioides terrestris LPH172 was cultivated on three different steam-pretreated spruce materials and on non-treated spruce, harboring different hemicellulose content and structural characteristics. The aim of the work was to map the substrate-induced changes in the secretome of T. terrestris when grown on differently treated spruce materials and evaluate the hydrolytic efficiency of the secretome as supplement of a commercial enzyme mixture. The cultivation of T. terrestris was monitored by enzyme activity measurements of endo-cellulase, endo-xylanase, endo-mannanase, laccase and peroxidase. Proteomic analysis was performed on the secretomes induced by these materials to map the differences in enzyme expression.

Project description:Human bocavirus (HBoV) is a newly discovered parvovirus identified from pooled nasopharyngeal aspirate specimens. Human bocavirus 1 (HBoV1) is a respiratory virus observed in respiratory samples from small children presenting bronchiolitis, wheezing, cough, fever, and rhinorrhea. It is the fourth most common virus detected in respiratory infections. DNA of HBoV1 was detected in up to 18% of nasal or nasopharyngeal samples and another study has been shown that over than 85% of children in the United States have antibodies to this virus. HBoV1 is a small DNA virus with a nonenveloped icosahedral capsid. This virus previously has been associated with wheezing, acute otitis media, severe pneumonia and respiratory failure. HBoV 1 has been also detected in the blood of acute respiratory patients and the selected group of immunocompromised children and also determined in healthy blood donors. The role of HBoV1 in the inflammatory process is poorly known. The aim of this project is to clarify the role of HBOV1 in the immunoregulatory mechanisms.
