ABSTRACT: We performed quantitative metabolomics and 13C-glucose isotopic profiling experiments on AML MOLM14 cell line following 24h of treatment with venetoclax (50 nM) and aracytine (25 uM) or combination compared to untreated samples. In addition, we also performed quantitative metabolomic experiments on MOLM14 silenced for BCL2 (shRNA).

Project description:We used quantitative mass spectrometry-based proteomics to unravel global changes in the phosphoproteome upon treatment of ASP14 cells (a Ewing Sarcoma cell line) according to the setup below. The combination of drugs were experimentally shown to infer synergy in terms of cell killing. Project description:The quinoline-3-carboxamide ABR-215050 (Tasquinimod (INN)), exhibits anti-tumor activity via inhibition of tumor angiogenesis in human and rodent tumors. To further explore the mode of action of ABR-215050, in vivo experiments with gene microarray analysis were performed using samples of LNCaP prostate tumors excised from nude mice exposed to ABR-215050 for 24h at 10 mg/kg(ad.lib.). The array data were validated by real-time PCR (sqRT-PCR), as well as by protein expression experiments. Among several significant differentially expressed gene after exposure to ABR-215050, were cytokine receptor CXCR4, cytochrome P450 1A1 (CYP1A1), thrombospondin-1 (THBS1) and Lysyloxidase preprotein (LOX). To further explore the mode of action of ABR-215050, in vitro experiments with gene microarray analysis were performed using LNCaP prostate tumor cells. The array data were validated by real-time PCR (sqRT-PCR), as well as by protein expression experiments. One of the most significant differentially expressed genes after exposure to ABR-215050, was thrombospondin-1 (THBS1).

Project description:In this dataset, we include the expression data obtained from untreated and blast pathogen treated rice seedlings using a variety of blast resistant rice line H4, as well as the susceptible rice line Zhonger-Ruanzhan. These data are used to obtain 4087 genes that are differentially expressed in response to blast pathogen in both of rice lines,as well as 717 genes that are differentially expressed between different lines both in the moch-treated and the blast treated. We used microarrays to detail the global gene expression in leaf from blast resistant rice line and susceptible rice line Four total samples were analyzed, The untreated rice line Zhonger-Ruanzhan: Zhonger-0;the untreated rice line H4:H4-0;Zhonger-Ruanzhan inoclutated with blast pathogen after 24h:Zhonger-24;H4 inoculated with blast pathogen after 24h: H4-24. FGFR inhibition blocks NF-kappaB-dependent glucose metabolism and confers metabolic vulnerabilities in cholangiocarcinoma

Project description:This study explored the response to FGFR inhibition in FGFR2-fusion+ intrahepatic cholangiocarcinoma. We conducted phosphoproteomics experiments in a patient-derived FGFR2-driven ICC cell line model upon FGFR inhibition. Samples were treated with the FGFR inhibitor, Futibatinib/TAS120 (75 nM) for 4 or 24 hours or with DMSO vehicle. The experiments were done using an Orbitrap Fusion Lumos mass spectrometer. Multiplexing was achieved using either TMT10/11 reagents and the SPS-MS3 method. Basic pH reversed-phase chromatography (bRPLC) was used for off-line pre-fractionation; 12 fractions were analyzed for proteome mappings (PMID: 26700037) and 24 fractions plus a phosphotyrosine-enriched sample for phosphoproteome mappings (PMID: 31606085). Phosphoproteome mappings were done using both HCD fragmentation with Orbitrap fragment ion detection and CID fragmentation with ion trap fragment ion detection (PMID: 29487189, PMID: 31606085).
