GNPS Staphylococcus aureus lipopeptides

ABSTRACT: Septic arthritis, most often caused by Staphylococcus aureus (S. aureus), is a rapidly progressive and destructive joint disease with substantial mortality and morbidity. S. aureus lipoproteins (Lpps) are known to induce arthritis and bone destruction. Here, we aimed to investigate the bone resorptive effect of S. aureus Lpps in a murine arthritis model by intra-articular injection of purified S. aureus Lpps, synthetic lipopeptides, and live S. aureus strains. Analyses of the bone mineral density of the distal femur bone were performed. Intra-articular injection of both live S. aureus and purified S. aureus Lpps were shown to significantly decrease total- and trabecular the bone mineral density. HPLC-high resolution MS/MS analyses revealed that the Lpps expressed by S. aureus SA113 strain contain both diacyl and triacyl lipid moieties. Therefore, tryptic digestion of Lpps was carried out, and the resulting lipopeptides were analyzed by HPLC-MS/MS, in which peptide fragments and neutral losses of fatty acids were used to identify the lipopeptide structures. Expression data from monoiodoacetate-induced arthritis in a rat model

Project description:The progression of arthritis involves sequential regulation of catabolic and anabolic genes that degenerate cartilages. We used microarrays to determine temporal regulation of genes during the progression of monoiodoacetate-induced arthritis in the knees of rats. Experimental arthritis was induced by intra-articular injection of monoiodoacetate (2 mg/ 50 ul) in the knees of rats. Cartilages from distal end of femur were collected on day 5, 9 and 21 post-injection. Saline injection was used as a sham control.

Gene expression profiling of mammary epithelial cells and their role in the predisposition to Staphylococcus aureus infections

Project description:Background: Staphylococcus aureus is a major pathogen of humans and animals. Host genetics influence the susceptibility to S. aureus infections, and genes determining infection outcome remain to be identified. Here, we used outbred animals from a divergent selection on susceptibility towards Staphylococcus infection to explore host immunogenetics. Methodology/Principal Findings: We investigated in vitro how mammary epithelial cells (MEC) respond to live S. aureus or S. aureus supernatant and whether MEC from animals with different degree of susceptibility to intra-mammary infections have distinct gene expression profiles. We measured the expression of 15K probes in MEC after each kind of stimulation (living bacteria or culture supernatant) at three different time points (a reference without bacteria, 1 and 5 hours) with ovine Agilent microarrays. Furthermore, a selected number of genes were confirmed by RT-qPCR. Gene signatures of stimulated MEC were obtained and genes involved in the cell cycle and metabolic processes were down-regulated during the kinetics and the apoptosis pathways were highly modified after both live bacteria and supernatant stimulations. Genes involved in immune response were up-regulated over-all after supernatant exposure. Furthermore 23 genes were differentially expressed between the resistant and susceptible animals, two of them were involed in oxidative processes, but the differences between the animals were very few. Conclusion/Significance: we successfully obtained Staphylococcus aureus associated gene expression of ovine MEC in a 5 hour kinetics experiment. The in vitro MEC model does not provide much information on the differences between Staphylococcus resistant and susceptible animals. Gene expression profiling of cancer cells purified from prostate cancer bone metastasis mouse model

Project description:The study aimed to determine mechanisms of macrophage promotion of anti-androgen resistance of prostate cancer bone disease. For this purpose, we developed a novel prostate cancer bone metastasis mouse model, MycCaP-Bo, via multiple rounds of in vivo selection of bone homing cells following intra-cardiac inoculation of a murine prostate cancer cell line, MycCaP cells. Cancer cells were purified from bone metastasis using fluoresent accelerated-cell sorting (FACS) based on their iRFP expression. Cancer cells were purified from treatment- naïve tumours, resistant tumours and resistant tumours with short-term depletion of macrophages. RNA was extracted from purified cancer cells and profiled using next-generation sequencing.
