Effect of Streptomyces roseolus cell free supernatant on fungal development, transcriptome and aflatoxin B1 production of Aspergillus flavus

Project description:Frequently observed in tropical and sub-tropical regions, crops contamination by aflatoxin B1 (AFB1) produced by Aspergillus flavus, is emerging in Europe, due to climate change. Many alternative methods are currently developed to reduce the use of chemical inputs to prevent mycotoxin contamination, such as biocontrol agents (BCAs). Actinobacteria are known to produce many bioactive compounds and some of them are able to reduce in vitro AFB1 concentration. In this context, the present study aims to analyze the effect of a cell free supernatant (CFS) from Streptomyces roseolus liquid culture on A. flavus development, as well as on its transcriptome profile using microarray assay and its impact on AFB1 concentration. To study the impact of Streptomyces roseolus cell free supernatant on global transcriptome of Aspergillus flavus we have employed whole genome microarray expression profiling.

Transcriptome response of Aspergillus fumigatus cultures under different temperatures

Project description:Microarray analysis was used to compare gene expression of Aspergillus fumigatus under two different sporulation temperatures, 17oC and 32oC, with an emphasis on genes encoding known or putative allergens of the fungus. The objective of the study was to investigate whether allergenic potencies of A. fumigatus spores produced under different sporulation temperatures would be influenced by temperature-dependent transcriptional regulation of allergenicity genes. Non-sporulating liquid culture of Aspergillus fumigatus was harvested and divided equally onto two sets of potato dextrose agar plates, one set for incubation at 17oC, the other for incubation at 32oC. After 48 hours of incubation, RNA was harvested from both sets of sporulating cultures, reverse-transcribed into dye-coupled cDNA and hybridized onto microarrays for analysis of gene expression. For each experiment, extracted RNA from the two cultures were hybridized onto two dye-swap technical replicate arrays. A total of three separate experiments were conducted for biological triplicates, for a total of six hybridizations.

Project description:Aspergillus nidulans competes in its natural environment with other microorganisms. It has been known that the bacterium Streptomyces rapamycinicus induces the production of Orsellinic acid in A. nidulans and we found that the major trigger of this induction is Polaramycin B produced by S. rapamycinicus. Here we show the transcriptome of A. nidulans following treatment with 0.5µg/ml Polaramycin B after 30 minutes and 3 hours.

Expression analysis of Aspergillus oryzae grown on different carbon sources

Project description:In this study, we focused on chemically defined inducers or substrates to drive expression of cellulases, hemicellulases and accessory enzymes in the model filamentous fungus Aspergillus oryzae. Cellohexaose (O-CHE), mannohexaose (O-MHE), xylopentaose (O-XPE), arabinoheptaose (O-AHP), 1,3:1,4-M-NM-2-glucohexaose (O-BGHEXA), 63-M-NM-1-D-glucosyl-maltotriosyl-maltotriose (O-GMH), 61-M-NM-1-D-galactosyl-mannotriose (O-GM3), xyloglucan (X3Glc4-borohydride reduced; O-X3G4R), turanose (TYR) and sophorose (SOP) were used to induce the plant polysaccharide degradation machinery of A. oryzae. The strain used in this study was the A. oryzae sequenced strain RIB40, obtained from IBT culture collection at Technical University of Denmark. To obtain a global view of the A. oryzae transcriptome activated for plant biomass conversion, mRNA from growth after 2 h on 10 different carbohydrate active enzyme inducers (di- and M-bM-^@M-^Soligo saccharides) was subjected to custom-designed Agilent microarray analysis.
