Identification of proteins from vacuoles of red algae

Project description:Vacuoles and lysosomes are single-membrane-bound organelles involved in diverse functions such as intracellular digestion and storage or secretion of metabolites. To understand their origin, evolution and fundamental features, the identification of proteins comprising these compartments in missing links would be invaluable. So, we isolated the vacuoles from Cyanidioschyzon merolae, which is considered to be one of the most primitive photosynthetic eukaryotes, and identified 46 proteins by matrix-assisted laser desorption/ionization time of flight-mass spectrometry. Identification of human FGF9 IRES interacting proteins

Project description:We have previously shown that FGF9 is overexpressed in hypoxia through the IRES located in the 5’UTR. To identify the protein that binds to FGF9 IRES and controls FGF9 protein synthesis in hypoxia, FGF9 IRES RNA was in vitro synthesized and used to pulled-down interacting proteins. The RNA-protein complexes were first visualized by sliver staining, followed by cutting specific bands for protein identification using matrix-assisted laser desorption/ionization time-of-flight mass spectrometer (MALDI-TOF MS).

Project description:The age of mosquitoes is a crucial determinant of their susceptibility to infection, probability of survival to transmit pathogens and tolerance to insecticides. We investigated changes to the abundance of proteins found in heads and thoraces of Anopheles gambiae and Anopheles stephensi as they aged. Protein expression changes were assessed using two-dimensional difference gel electrophoresis and the identity of differentially expressed proteins was determined by using either matrix-assisted laser desorption ionization tandem time-of-flight mass spectrometry or capillary high-pressure liquid chromatography coupled with a linear ion-trap (LTQ)-Orbitrap XL hybrid mass spectrometer. Protein biomarkers were validated by quantitative Western blot analysis.

Identification of type strain-specific diagnostic peptides of the Bacillus cereus group

Project description:The Bacillus cereus group consists of eight very closely related species and comprises both harmless and human pathogenic species. Yet, methods to rapidly and accurately distinguish these species are currently lacking as we demonstrate that classical matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) biotyping did not achieve reliable identification of each type strain. We assigned type strain-specific diagnostic peptides to the B. cereus group based on comparisons of their proteome profiles. The number of diagnostic peptides varies remarkably in type strain-dependent manner. The state of the art of the reference database is crucial in the process of validating candidate diagnostic peptides and may lead to a noteworthy reduction of verified diagnostic peptides as putative diagnostic peptides might be found in other species as well.
