Proteomic analysis of Cowpea aphid Aphis craccivora saliva

ABSTRACT: The cowpea aphid, Aphis craccivora (Hemiptera: Aphididae), is one of the most destructive insect pests of cowpea, peanut, and other legumes in tropical and subtropical regions. However, there are little-known molecular mechanisms underlying the host plant specification and their interaction. Aphids secretes numerous proteins while feeding on plants to counteract plant defenses which became indispensable for its survival and growth. In this study we identified salivary proteins of cowpea aphid using LC-MS/MS.

The salivary proteins acts as interface between aphid and their host plant.However, the cotton aphid salivary proteome was not studied yet. Identifying the salivary proteins helps in better understanding of aphid adaptation to their host plant which aids us to search for novel plant genetic source.

LC-MS/MS of Pseudoregma bambucicola salivary gland and saliva

Project description:In this study, liquid chromatography-tandem spectrometry (LC–MS/MS) analysis was conducted to characterize the overall salivary protein composition from dissected salivary glands and secreted saliva in P. bambucicola, and to identify putative effector proteins important for aphid-plant interactions. This study promotes our understanding for the role of salivary glands in host specialization of P. bambucicola, and provide insight into its adaptation to unique feeding habit.

Arabidopsis thaliana gene expression changes upon treatment with green peach aphid saliva

Project description:Myzus persicae (green peach aphid) feeding on Arabidopsis thaliana induces a defense response, quantified as reduced aphid progeny production, in infested leaves but not in other parts of the plant. Similarly, infiltration of aphid saliva into Arabidopsis leaves causes only a local increase in aphid resistance. Further characterization of the defense-eliciting salivary components indicates that Arabidopsis recognizes a proteinaceous elicitor with a size between 3 to 10 kD. Genetic analysis using well-characterized Arabidopsis mutant shows that saliva-induced resistance against M. persicae is independent of the known defense signaling pathways involving salicylic acid, jasmonate, and ethylene. Among 78 Arabidopsis genes that were induced by aphid saliva infiltration, 52 had been identified previously as aphid-induced, but few are responsive to the well-known plant defense signaling molecules salicylic acid and jasmonate. Quantitative PCR analysis confirms expression of saliva-induced genes. In particular, expression of a set of O-methyltransferases, which may be involved in the synthesis of aphid-repellent glucosinolates, was significantly up-regulated by both M. persicae feeding and treatment with aphid saliva. However, this did not correlate with increased production of 4-methoxyindol-3-ylmethylglucosinolate, suggesting that aphid salivary components trigger an Arabidopsis defense response that is independent of this aphid-deterrent glucosinolate. The salivary gland proteome of root-galling grape phylloxera (Daktulosphaira vitifoliae Fitch) feeding on Vitis spp.

Project description:Herbivory plant-parasite interactions depend on the delivery of effector molecules by the invading insect species. Sedentary gall forming insects, such as grape phylloxera (Daktulosphaira vitifoliae FITCH, Phylloxeridae) secrete multiple effectors into host plant tissues that alter host cellular functions to the benefit of the insect. Analyses revealed 420 putative ‘DvEffectors’ were detected in salivary glands, dissected from root-feeding vs. starving D. vitifoliae larvae reared on Teleki 5C (V. berlandieri x V. riparia) under controlled growth conditions (25±3°C, 60% rH) by proteomic mass spectrometry and in-silico secretory prediction. Slug feeding triggers dynamicmetabolomic and transcriptomic responses leading to induced resistance in Solanum dulcamara

Project description:Induced plant responses to insect herbivores are well studied, but we know very little about responses to gastropod feeding. We aim to identify the temporal dynamics of signalling- and defence-related plant responses after slug feeding in relation to induced resistance. We exposed Solanum dulcamara plants to feeding by the grey field slug (GFS; Deroceras reticulatum) for different periods and tested discs of local and systemic leaves in preference assays. Induced responses were analysed using metabolomics and transcriptomics. GFS feeding induced local and systemic responses. Slug feeding for 72h more strongly affected the plant metabolome than 24h feeding. It increased the levels of a glycoalkaloid (solasonine), phenolamides, anthocyanins, and trypsin protease inhibitors as well as polyphenol oxidase activity. Phytohormone and transcriptome analyses revealed that jasmonic acid, abscisic acid and salicylic acid signalling were activated. GFS feeding upregulated more genes than that it downregulated. The response directly after feeding was more than five times higher than after an additional 24h without feeding. Our research showed that GFS, like most chewing insects, triggers anti-herbivore defences by activating defence signalling pathways, resulting in increased resistance to further slug feeding. Slug herbivory may therefore impact other herbivores in the community.

Australian sugarcane soldier fly’s salivary gland transcriptome in response to starvation and feeding on sugarcane crops

Project description:The soldier fly is an endemic pest of sugarcane in Australia. Small numbers of larvae can cause significant damage to roots and reduce the crop yields. Little is known about the composition and function of the soldier fly salivary gland, its secretions, and their roles in insect-plant interactions. In this study, we performed transcriptome analysis of the salivary glands of starved and sugarcane root-fed soldier fly larvae. A total of 31,119 highly expressed assembled contigs were identified in the salivary glands and almost 50% of them showed high levels of similarity to known proteins in Nr databases. Of all the obtained contigs, only 9,727 sequences contain an open reading frame of over 100 amino acids. Around 31% of contigs were predicted to encode secretory proteins, including some digestive and detoxifying enzymes and potential effectors. Some known salivary secreted peptides such as serine protease, cysteine proteinase inhibitors, antimicrobial peptides and venom proteins were among the top 100 highly expressed genes. Differential gene expression analysis revealed significant modulation of 850 transcripts in salivary glands upon exposure to plant roots or starvation stress. Here, we identified some venom proteins which were significantly upregulated in the salivary glands of soldier fly larvae exposed to sugarcane roots. In other insects and nematodes some of these proteins have been used to manipulate host plant defense systems and facilitate the invasion of the host plant. These findings provide a further insight into the identification of potential effector proteins involved in soldier fly- sugarcane interactions.

Hormetic response and transcriptome profiling reveal the mechanism underlying metabolic and cuticular resistance to host-specific terpenoid defenses in an emerging insect pest, Pagiophloeus tsushimanus (coleoptera: curculionidae)

Project description:The resistance mechanisms evolved by insects to overcome host-plant allelochemicals is a key consideration in pest management. Camphor oil (EO) and its main component (i.e D-camphor) form a specific terpenoid-defensive system in camphor trees, Cinnamomum camphora. However, an emerging insect pest, Pagiophloeus tsushimanus has recently caused serious damage to this intractable plant species, which is largely elusive. Here, we used feeding bioassays and RNA-seq to investigate the mechanism underlying the resistance of the beetle to host-specific terpenoid defenses. Firstly, a hormetic response in both larval weight and developmental time was observed in terpenoid-feeding individuals, which is a highly generalized dose-response phenomenon in toxicology whereas occurs infrequently in the context of insect-plant interactions. Then, comparative transcriptome analysis between terpenoid-feeding and control groups indicated that both CYP450s-mediated metabolic resistance and CPs-mediated cuticular resistance were jointly employed to cope with terpenoid-induced stress. In addition, a small portion of genes involved in glucose transport pathway were up-regulated at the low D-camphor dose, suggesting an extra intake of glucose used for larval growth may contribute to a hormetic response. These findings probably implied that the dual terpenoid-resistance mechanisms existing in this specialist is an essential precondition for hormetic response in larval growth, ultimately contributing to successfully colonize host camphor trees with impunity. Overall, our study will open new avenues for understanding the insect-plant coevolutionary adaptation and developing durable pest control strategies.
