Project description:The cowpea aphid, Aphis craccivora (Hemiptera: Aphididae), is one of the most destructive insect pests of cowpea, peanut, and other legumes in tropical and subtropical regions. However, there are little-known molecular mechanisms underlying the host plant specification and their interaction. Aphids secretes numerous proteins while feeding on plants to counteract plant defenses which became indispensable for its survival and growth. In this study we identified salivary proteins of cowpea aphid using LC-MS/MS.

Project description:In this study, liquid chromatography-tandem spectrometry (LC–MS/MS) analysis was conducted to characterize the overall salivary protein composition from dissected salivary glands and secreted saliva in P. bambucicola, and to identify putative effector proteins important for aphid-plant interactions. This study promotes our understanding for the role of salivary glands in host specialization of P. bambucicola, and provide insight into its adaptation to unique feeding habit.

Project description:Herbivory plant-parasite interactions depend on the delivery of effector molecules by the invading insect species. Sedentary gall forming insects, such as grape phylloxera (Daktulosphaira vitifoliae FITCH, Phylloxeridae) secrete multiple effectors into host plant tissues that alter host cellular functions to the benefit of the insect. Analyses revealed 420 putative ‘DvEffectors’ were detected in salivary glands, dissected from root-feeding vs. starving D. vitifoliae larvae reared on Teleki 5C (V. berlandieri x V. riparia) under controlled growth conditions (25±3°C, 60% rH) by proteomic mass spectrometry and in-silico secretory prediction. Project description:The resistance mechanisms evolved by insects to overcome host-plant allelochemicals is a key consideration in pest management. Camphor oil (EO) and its main component (i.e D-camphor) form a specific terpenoid-defensive system in camphor trees, Cinnamomum camphora. However, an emerging insect pest, Pagiophloeus tsushimanus has recently caused serious damage to this intractable plant species, which is largely elusive. Here, we used feeding bioassays and RNA-seq to investigate the mechanism underlying the resistance of the beetle to host-specific terpenoid defenses. Firstly, a hormetic response in both larval weight and developmental time was observed in terpenoid-feeding individuals, which is a highly generalized dose-response phenomenon in toxicology whereas occurs infrequently in the context of insect-plant interactions. Then, comparative transcriptome analysis between terpenoid-feeding and control groups indicated that both CYP450s-mediated metabolic resistance and CPs-mediated cuticular resistance were jointly employed to cope with terpenoid-induced stress. In addition, a small portion of genes involved in glucose transport pathway were up-regulated at the low D-camphor dose, suggesting an extra intake of glucose used for larval growth may contribute to a hormetic response. These findings probably implied that the dual terpenoid-resistance mechanisms existing in this specialist is an essential precondition for hormetic response in larval growth, ultimately contributing to successfully colonize host camphor trees with impunity. Overall, our study will open new avenues for understanding the insect-plant coevolutionary adaptation and developing durable pest control strategies.

Project description:The reduviid bug Triatoma dimidiata is an important Chagas disease vector widely distributed along Central America. This insect has great capability for domestic adaptation as the majority of specimens caught inside human dwellings or in peridomestic areas in Central America fed human blood. Within this context, exploring the salivary compounds that overcome host haemostatic and immune responses to assist blood-feeding is of great scientific interest. Our aim is to provide a deeper insight into T. dimidiata salivary gland molecules that may contribute to its haematophagic habit. We realized T. dimidiata salivary transcriptomic analysis and complemented it through proteomics, disclosing the set complexity of 119 secreted proteins. The large-scale approach used enriches the pharmacologically active molecules database and improves our knowledge about the complexity of salivary compounds from haematophagous vectors and their biological interactions.

Project description:During blood feeding haematophagous arthropods inject into their hosts a cocktail of salivary proteins whose main role is to counteract host haemostasis, inflammation and immunity. However, animal body fluids are known to also carry miRNAs. To get insights into saliva and salivary gland miRNA repertoires of the African malaria vector Anopheles coluzzii we used small RNA-Seq and identified 214 miRNAs, including tissue-enriched, sex-biased and putative novel anopheline miRNAs. Noteworthy, miRNAs were asymmetrically distributed between saliva and salivary glands, suggesting that selected miRNAs may be preferentially directed toward mosquito saliva. The evolutionary conservation of a subset of saliva miRNAs in Anopheles and Aedes mosquitoes, and in the tick Ixodes ricinus, supports the idea of a non-random occurrence pointing to their possible physiological role in blood feeding by arthropods. Strikingly, eleven of the most abundant An. coluzzi saliva miRNAs mimicked human miRNAs. Prediction analysis and search for experimentally validated targets indicated that miRNAs from An. coluzzii saliva may act on host mRNAs involved in immune and inflammatory responses. Overall, this study raises the intriguing hypothesis that miRNAs injected into vertebrates with vector saliva may contribute to host manipulation with possible implication for vector-host interaction and pathogen transmission.

Project description:Myzus persicae (green peach aphid) feeding on Arabidopsis thaliana induces a defense response, quantified as reduced aphid progeny production, in infested leaves but not in other parts of the plant. Similarly, infiltration of aphid saliva into Arabidopsis leaves causes only a local increase in aphid resistance. Further characterization of the defense-eliciting salivary components indicates that Arabidopsis recognizes a proteinaceous elicitor with a size between 3 to 10 kD. Genetic analysis using well-characterized Arabidopsis mutant shows that saliva-induced resistance against M. persicae is independent of the known defense signaling pathways involving salicylic acid, jasmonate, and ethylene. Among 78 Arabidopsis genes that were induced by aphid saliva infiltration, 52 had been identified previously as aphid-induced, but few are responsive to the well-known plant defense signaling molecules salicylic acid and jasmonate. Quantitative PCR analysis confirms expression of saliva-induced genes. In particular, expression of a set of O-methyltransferases, which may be involved in the synthesis of aphid-repellent glucosinolates, was significantly up-regulated by both M. persicae feeding and treatment with aphid saliva. However, this did not correlate with increased production of 4-methoxyindol-3-ylmethylglucosinolate, suggesting that aphid salivary components trigger an Arabidopsis defense response that is independent of this aphid-deterrent glucosinolate. Project description:We developed a new simple method to assess the composition of proteins in the Ornithodoros moubata saliva. To collect naturally expectorated saliva from the ticks, we employed an artificial membrane feeding technique using a simple, chemically defined diet and submitted the collected native saliva samples for liquid chromatography-mass spectrometry (LC-MS) analysis. These experiments were conducted with groups of uninfected ticks as well as with B. duttonii infected ticks. The ticks exhibited a fair feeding response to the tested diet, engorgement rates reaching between 60-100% of ticks per feeding chamber. LC-MS analysis identified a total of 17 and 15 proteins in saliva samples from the uninfected and infected ticks respectively. Importantly, the analysis was sensitive enough to detect up to 9 different proteins in the saliva containing diet upon which as few as 6 nymphal ticks fed. Some of the proteins identified are well known for their immunomodulatory activity in a vertebrate host, whereas others were structural or “housekeeping” proteins and their finding in the naturally expectorated saliva confirms that they can be secreted and having some roles at the tick-host interface. Most notably, some of the proteins that have long been suspected as important for vector-pathogen interactions of Borrelia spirochetes were detected only in the samples from infected ticks, suggesting that their expression was altered by the persistent colonization of the tick’s salivary glands by spirochetes. The simple method described herein is an important addition to the toolbox available to study the vector-host-pathogen interactions in the rapidly feeding soft ticks.

Project description:Over the last 20 years, the advances in sequencing technologies highlighted the unique composition of the salivary glands of blood-feeding arthropods. Further biochemical and structural data demonstrated that salivary proteins can disrupt host hemostasis, inflammation and immune response in addition to favor pathogen transmission. Previously, a Sanger-based sialome of the adult Ochlerotatus. triseriatus female salivary glands was published based on 731 ESTs. Here we revisited O. triseriatus salivary glands contents using an Illumina-sequencing approach of both male and female tissues. In the current data set we report 10,317 coding DNA sequences classified into several functional classes. The translated transcripts also served as reference database for the proteomic analysis of O. triseriatus female saliva, in which unique peptides of 101 proteins were found. Finally, comparison of male and female libraries allowed the identification of female-enriched transcripts that are potentially related to blood acquisition and virus transmission.
