LC-MS characterization of anti-PD-L1 and CXCR4 bispecific nanobodies

Project description:Antibodies and derivative drugs targeting immune checkpoints have been approved for the treatment of several malignancies, but there are fewer responses in patients with pancreatic cancer. Here, we designed a nanobody molecule with bi-targeting on PD-L1 and CXCR4, as both targets are overexpressed in many cancer cells and play important roles in tumorigenesis. The nanobody sequences targeting PD-L1 and CXCR4 were linked by the (G4S)3 flexible peptide to construct the anti-PD-L1/CXCR4 bispecific nanobody. The bispecific nanobody was expressed in E. coli cells and purified by affinity chromatography. The purified nanobody was biochemically characterized by mass spectrometry, Western blotting and flow cytometry to confirm the molecule and its association with both PD-L1 and CXCR4. The biological function of the nanobody and its anti-tumour effects were examined.

A molecular signature for MET-targeted therapy

Project description:Constitutive MET signaling promotes invasiveness in primary and recurrent GBM; however, current MET-targeting strategies lack of effective biomarkers for selecting suitable patients for treatment. Here, we identified a predictive signature potentially valuable for indicating vulnerability to MET-targeted therapy in GBM. The use of both human and mouse gene expression microarrays showed that MET inhibitors regulate tumor (human) and host (mouse) cells within the tumor via distinct molecular processes, but overall they impede tumor growth by inhibiting cell cycle progression. Notably, GBM tumors with EGFRamp that showed resistance to erlotinib treatment also showed activation of the MET pathway, suggesting that a combination of EGFR and MET inhibitors may overcome or prevent such resistance in patients with EGFRamp GBM. GBM cell lines (DBM2, U251M2, U87M2, U118, and SF295) were treated for 7 days with either vehicle (n=14) or V-4084 (n=14) (i.e. SF295 + vehicle (n=2), SF295 + V-4084 (n=2), U118 + vehicle (n=3), U118 + V-4084 (n=3), U87M2 + vehicle (n=3), U87M2 + V-4084 (n=3), DBM2 + vehicle (n=3), DBM2 + V-4084 (n=3), U251M2 + vehicle (n=3), U251M2 + V-4084 (n=3)). GBM cell lines (DBM2, U251M2, U87M2, U118, and SF295) were subcutaneously inoculated into the flank region of nude mice to initiate tumor growth. miRNA expression data from MCF-7 cells, overexpressing ANXA1

Project description:Annexin 1 (ANXA1), an endogenous anti-inflammatory protein which modulates cellular processes such as proliferation, differentiation and apoptosis has been implicated in cancer initiation and progression. ANXA1 was previously shown to be regulated by hsa-miR-196a and promoted cell proliferation and anchorarge-dependent growth and suppressed apoptosis. However, whether ANXA1 itself regulates miRNA expression is unknown. Therefore, in this study, we investigated the regulation of miRNA by ANXA1 in breast cancer cells. Using microarray technology, 12 miRNAs were found to be significantly and consistently downregulated in MCF-7 cells (MCF-V5) overexpressing ANXA1 overexpressing MCF-7 cells (MCF-V5). Hsa-miR-26b* and hsa-miR-562 were chosen for further investigation.The data suggest that miR-26b* and miR-562 may play a role in ANXA1-induced migration and possibly angiogenesis by targeting NFKB and point towards a potential therapeutic target for breast cancer. Breast cancer MCF-7 cells (MCF-V5) overexpressing ANXA1 were cultured for RNA extraction and hybridization on Affymetrix miRNA microarrays. Gene expression of canine mammary cancer stem-like cells co-cultured with tumor associated macrophages

Project description:The aim of our study was to evaluate gene expression of canine mammary cancer stem-like cells co-cultured with tumor associated macrophages. Two canine mammary cancer cell lines (CMT-U27 and P114) were stained using anti-Sca1 (stem cell antigen 1), anti-EpCAM (Epithelial cell adhesion molecule) and anti-CD44 antibody. The FACS analysis showed 0,02-0,05% of Sca1+/EpCAM+/CD44+ in each of the cell line. Cancer stem-like cells were collected using FACS Aria II then co-cultured with tumor associated macrophages and used for further analysis of gene expression ( using Agilent Gene Expression Hybridization Kit ). Canine mammary cancer cell lines were stained using anti-Sca1 (stem cell antigen 1), anti-EpCAM (Epithelial cell adhesion molecule) and anti-CD44 antibodies. Next using FACS Aria II and Sca1+/EpCAM+/CD44+ cells were collected and co-cultured with tumor associated macrophages. Then, total RNA was isolated and hybridized at Gene Expression microarray.

Proteome Integral Solubility Alteration assay in zebrafish embryo for exploring exposure to bioactive compounds including pollutants and chemicals mixtures

Project description:The One Health initiative connects human and animal health with their shared environment. The development of novel methodology to unbiased identification of mechanisms of action of chemicals and chemicals mixtures would address one of today´s challenges of environmental and human toxicology. Zebrafish embryos offer a unique opportunity to explore the proteome integral solubility alteration (PISA) assay to study the proteins that increase or decrease their solubility as protein targets of the bioactive compounds in an organism at an developmental stage. We presented here 4 different applications of the PISA method in zebrafish embryo with different types of compounds of environmental and health concern: i) endocrine disrupting chemical, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) to outline the pathways compromised under exposure; ii) complex chemical mixtures composed by TCDD + alpha-endosulfan + bisphenol A (BPA) previously studied in vitro cells lines to evaluate single chemical versus mixture; iii) novel compounds from blue-green circular economy to map the organismal response, and a iv) novel drug directed to anti-cancer activity in lymphoma models to discharge side-effects at non-target organs. Our results demonstrated the unique opportunities of PISA assay in zebrafish for toxicology. We remark some differences to its application for drug target deconvolution. First, we evaluate both targets arising from an increase in solubility and in those decreasing, as both would be involved in modulation the cellular functions. Second, the method could be applicable to bioactive compound before chemical characterization or a chemical mixture of unknown stoichiometry. Third, the results from zebrafish embryo could provide a first glimpse of unspecific interactions with proteins in non-target tissues.The introduction of PISA assay in toxi- and ecotoxicoproteomics offers high-resolution and throughput in support with the 3R, and 3M principles.
