HLA-DO Modulates the Diversity of the MHC-II Self-peptidome


ABSTRACT: HLA-DR1 bound peptides of five biological replicates of LG2-WT(012916_WT1,022216_WT1, 02080316MHCIIWT1, 080316MHCIIWT2, 080316MHCIIWT3) and DO-KO Clone 1 LG-2 cells (012916_KO1, 022216_DO1, 081216MHCIIKO1, 080416MHCIIKO2, 080416MHCIIKO3) were processed in parallel. Each biological replicate has 3 technical replicates. In manuscript, these biological replicates are labeled as WTa, WTb, WTc,WTd, WTe,DOa, DOb,DOc, DOd, DOe respectively. HLA-DR1 bound peptides from three biological replicates of LG2-WT and DO-KO Clone 2 were processed in parallel. Each biological replicate had 3 technical replicates. H2-IAb bound peptides of mice splenic B cells from three biological replicates of WT and H2-O KO mice were processed in parallel. Each biological replicate had 3 technical replicates.

Presentation of antigenic peptides on MHC-II molecules is essential for tolerance to self and for initiation of immune responses against foreign antigens. DO (HLA-DO in humans, H2-O in mice) is a nonclassical MHC-II protein that has been implicated in control of autoimmunity and regulation of neutralizing antibody responses to viruses.
